Investigation of Physicochemical Properties, Antioxidant Activity and In vitro Bioaccessibility of Cornelian Cherry (Cornus mas. L.) Nectar Enriched Полный текст

Cornelian cherry (Cornus mas L.) fruit is a member of the cranberry family, which contains many bioactive components. However, because of its bitter taste, the number of products it can be processed is limited. In this study, beverages were prepared by adding five different herbal teas (linden, sage, chamomile, green tea, artichoke peel) to cranberry nectar sweetened with apple juice concentrate. Brix, pH, antioxidant capacity, (DPPH, FRAP, CUPRAC), total phenolic content, total monomeric anthocyanin, color (L*, a*, b*, C*, h°) and in vitro bioaccessibility analyzes were performed in the beverages. Results of antioxidant capacity analysis were determined between 129.00±11.85 µmol TE/g DM- 365.42±2.62 µmol TE/g DM; 56.61±16.42 µmol TE/g DM – 103.39±2.36 µmol TE/g DM; 157.97±25.35 µmol TE/g DM – 276.82±22.81 µmol TE/g DM with DPPH, CUPRAC and FRAP methods, respectively. The total phenolic content was found between 655.35±24.17 mg GAE/100 g DM and 1165.09±30.69 mg GAE/100 g DM. The beverages containing green tea infusion (4.13±1.69 mg/kg) had the lowest total monomeric anthocyanin content, while beverages containing artichoke peel infusion (6.33±0.67 mg/kg) had the highest. In the samples analyzed after in vitro gastric digestion, an increase was observed in the total monomeric anthocyanin amount compared to the pre-digested samples, while a decrease was found in the antioxidant capacity and total phenolic content, in general. By adding herbal teas to cornelian cherry nectar, not only improved sensorial and nutritional properties of the fruit nectar, but also a new functional beverage was developed.

Vitamin A is an essential micronutrient and has important functions such as vision, growth, reproduction embryogenesis, cellular differentiation, and proliferation, immune function and epithelial protector in the organism. Biotechnological production of vitamins is increasing due to their advantages and significant advances. The vitreoscilla hemoglobin (VHb) gene is extremely effective in binding oxygen and conducting it under hypoxic conditions. In this study, the production of vitamin A in E. herbicola (wild type) and its recombinant strains was investigated in LB medium and M9 medium (containing high concentrations (1%) of different carbon sources). The maximum production of vitamin A of the recombinant strain with the hemoglobin gene (vgb+) was observed in including glucose and sucrose M9 medium and their total product levels in vgb+ recombinant strain were 0.14 µg/ml and 0.1 µg/ml, respectively. The vitamin A production in the M9 medium with glucose and sucrose were 2-fold and 1.4- fold higher than that of the wild strain, respectively. The extracellular product level (0.07 µg/ml) in LB was 7-fold higher than wild strain at 48 h. These results reveal that the expression of VHb in E. herbicola in the both LB and M9 medium (containing 1% glucose and 1% sucrose, specially) increase the vitamin A production.

Roasting is the most important coffee processing stage that affects coffee flavour. Coffee brew is prepared by various methods and Turkish coffee brewing technique is one of the oldest methods among other coffee preparation techniques. In this study, volatile compounds of light, medium, dark roasted ground coffee and Turkish coffee brews prepared from light, medium, dark roasted coffees were evaluated and the effect of roasting process was determined. The volatile compounds from acid (2), alcohol (1), aldehyde (2), furan (8), furanone (3), ketone (1), lactone (1), phenol (3), pyrazine (19), pyridine (1), pyrrole (6), and thiophene (1) chemical group was detected in all roasted ground coffee samples. However, volatiles compounds from acid, alcohol and furanone group were not detected in any Turkish coffee brew. Pyrazines were main volatile compounds of light roasted ground coffee and Turkish coffee brew of light roasted coffee. On the other hand, furans were main volatile compounds of medium, dark roasted ground coffee and Turkish coffee brew prepared from medium, dark roasted coffees followed by pyrazines.

The purpose of this study was to estimate and identify factors affecting the willingness of households to pay for conservation of Washera sheep in West Gojam, Ethiopia. This study used both primary and secondary data. A three-stage sampling method was used to collect data from 240 respondents. Data were analyzed using descriptive statistics and econometric models. The two-dimensional test model used a method to determine the choice of the imputation method using double limited dichotomies. The study showed that education level, livestock size, extension contacts and credit source positively affected the maximum household’s willingness to pay for conservation of Washera sheep. On the other hand, age, sex, land size, and the distance of the home from the household’s market have negatively affected the maximum household’s willingness to pay for conservation of Washera sheep. Meanwhile, the total number of valid answers corresponds to 122,168 households. Therefore, the mean willingness to pay for Washera sheep conservation was US$ 3.75 per year and the total economic benefit gain from Washera sheep conservation were equivalent to US$1,069,647.23. The study recommends that households be willing to contribute to the conservation of Washera sheep breed by providing the cash. This ensures community participation in all decision making and formulation of plans and policies related to the conservation of Washera sheep breeds.

Yürütülen çalışmada sürdürülebilir kaynaklardan çevre dostu biyopolimerlerin elde edilmesi ve biyomalzeme olarak kullanılabilirliği araştırılmıştır. Bu amaçla, sığır aşil tendonundan kollajen, ipek böceği kozasından fibroin, kahverengi deniz alglerinden sodyum alginat ve aloe vera jelinden biyoaktif bileşenler izole edilerek saflaştırılmıştır. Ürün verimi kollajen, fibroin, sodium alginat ve aloe vera için sırasıyla %79.8 (w/w), %69.49 (w/w kozadan), %35.1 (w/w) ve %1 (w/v jeldeki kuru miktar) oranında hesaplanmıştır. Bu biyomoleküllerden dondurarak kurutma yöntemi ile doku iskeleleri hazırlanmıştır. Ancak, aloe vera jeli katı formada yapı bütünlüğünü koruyamayarak 3-boyutlu iskele yapı oluşturamamıştır. Fibroin, kollajen ve sodyum alginat iskelelerin FTIR analizleri ürünlerin saf olarak elde edildiğini, liyoflizasyon sırasında kimyasal yapının korunduğunu göstermiştir. SEM ile yüzey analizleri ise iskelelerin doku mühendisliği uygulamaları için uygun olduğunu desteklemiştir. Sonuç olarak, petrol kaynaklı polimer yerine sürdürülebilir kaynaklardan, genel olarak oda şartlarında bioaktif polimerler yüksek verimle elde edilmiş ve biyomalzeme olarak kullanılabilecekleri belirlenmiştir. Biyomoleküllerin bu şekilde sürdürülebilir kaynaklardan elde edilmesi hem hammadde sorununun hem de polimer kaynaklı çevresel kirliliğin çözümünde önemli potansiyele sahiptir.

Tıbbi bitki Raphanus sativus L. yumru kökünün su, etanol ve metanol ekstraktlarının Vicia faba L. (bakla) yapraklarında Botrytis cinerea fungusunun oluşturacağı kurşuni küf hastalığına karşı toplam protein içeriği ve peroksidaz (POX) aktivitesi bakımından bitkide meydana getirebileceği antioksidan savunma sistemi spektrofotometrik olarak analiz edilmiştir. R. sativus yumru kökünün su, etanol ve metanol ekstraktlarının stok solüsyonları dimetil sülfoksit (DMSO) ile hazırlanmıştır. 105 spor/mL B. cinerea spor süspansiyonu hazırlanmıştır. V. faba yapraklarına sadece distile su (negatif kontrol), sadece DMSO, sadece B. cinerea (pozitif kontrol), sadece ekstrakt uygulamaları (distile su, etanol ve metanol ekstraktı) ve ekstrakt uygulamaları sonrasında B. cinerea uygulaması gerçekleştirilmiştir. Yapraklar, analizi yapılacak olan yöntemlere uygun olarak ekstrakte edilmiştir. Ekstraktlardan elde edilen süpernatantlardan analizler spektrofotometrik olarak gerçekleştirilmiştir. Araştırma sonucunda, toplam protein içeriğinin distile su uygulanan (negatif kontrol) gruba kıyasla ekstrakt:fungus uygulama gruplarında artış gösterdiği saptanmıştır. Fakat sadece fungus uygulanan (pozitif kontrol) gruba kıyasla genel olarak azaldığı belirlenmiştir. POX aktivitesinin ise, negatif kontrol grubuna kıyasla artış gösterdiği saptanmıştır. Toplam protein içeriği ve POX aktivitesi, sadece ekstrakt uygulanan gruplarda negatif kontrol grubuna göre artış göstermiştir. En yüksek POX aktivitesi, 10 mg/mL distile su:fungus uygulamasından elde edilmiştir. Bu açıdan, ekstrakt:fungus uygulanan gruplar arasında en iyi antioksidan aktivitenin 10 mg/mL distile su:fungus uygulaması olduğu belirtilebilmektedir. Bu açıdan bakıldığında, R. sativus saf su, etanol ve metanol ekstraktlarının V. faba yapraklarında antioksidan savunma yanıtı verdiği belirtilebilmektedir.

Yield performances differ according to the environment in which the plants are grown. In order to determine the chickpea cultivars suitable for Kahramanmaraş province, 17 different chickpea cultivars were tried during the growing seasons of 2018-2019 and 2019-2020. Among the characteristics examined in the study, flowering times, pod bind times, first branch height to years, varieties and years variety interactions were significant as statistical. There were statistically significant differences in terms of plant height, years and year × variety interaction. The number of pods, pod length, pod width created statistically significant differences in terms of years and cultivars. The differences among the cultivars in terms of grain yield were found as statistically significant. It was determined that the effects of years, cultivars and year-variety interactions on the first pod height, number of branches, and pod diameter did not have a statistically significant. At the end of the two-year study, it was concluded that Seckin, Arda, Hasanbey, Çagatay, Azkan, Ubet, Damla 89 and Aslanbey chickpea cultivars can be recommended due to the over grain yield of 300 (345-301) kg from per decare in Kahramanmaras conditions.

In social & economic science disciplines, the lack of strong theories is often reflected in the lack of well-accepted common metrics defined by a UNIT. PAHU Method vs. PC is developed to investigate the feasibility of developing well-grounded common metrics/unit to advance behavioural, economic-social, food security & science research, both in terms of advancing the development of theory and increasing the utility of research for policy & practice. In addition aiming, to consider whether a set of criteria can be developed for understanding when the measurement of a particular construct is ready to be standardized & to explore how the research community can foster a move toward standardization when it appears warranted. In this globalised society, even imperial measures are defined with reference to the metric/unit standards. A radical evaluation method change in global food systems is needed to meet the challenges. State of the art of PAHU Method /Age and Gender Corrected Per Capita (PCagc) is to evaluate demographic structure, consumer & past and future food consumption potential of developed & developing countries, target populations, their food sufficiency & also food security evaluations of family and households. It involves systematic attempts to create awareness of 19.4 percentage UNIT error inherent to PC & pave the way to food - other goods consumption evaluations plus global impact of hunger & environmental issues until 2020-2050-era. PAHUM was applied & evaluated EU28 demographic structure & food consumption issues for 1999/2010/2020. Now it focuses on research with systems approach, contributing to the development of tomorrow’s food systems for family/household evaluations including CO2 emissions-biodiversity relations. A radical evaluation method change in global/EU food systems is needed to meet the global challenges, including family/household on UNIT basis. In this context, it is necessary to select and develop salt-tolerant genotypes that can grow in salty soils and have high yields, and formulate strategies which may enhance the plant survival under salinity stress. Melatonin (N-acetyl-5-methoxytryptamine) is an important biological hormone that provides resistance to abiotic stress conditions and can be secreted by plants. Melatonin concentration in plants varies depending on genotype, temperature and growth period. Increase in melatonin concentration is associated with increased SNAT and HIOMAT/ASMT enzyme activity. It plays an important role in gibberellic acid and abscisic acid biosynthesis during the germination and provides plant growth and development. Exogenous application of melatonin significantly alleviates chlorophyll degradation and stomatal closure caused by salt stress, improves photosynthesis and enhances plants' salt tolerance. Besides it significantly reduces the harmful effects of salinity by regulating plant physiology, improving plant morphology, photosynthesis and activities of antioxidant enzymes. The present review discusses the recent studies on the effect of melatonin on plant growth and physiology against salt stress that have important impacts on plant growth and development have been given according to the findings of various researches. It also highlights the mechanim/s of melatonin-induced salinity stress tolerance in plants.

A total of 46 chub mackerel and 23 Atlantic mackerel samples were taken from the commercial fishermen engaged in purse seine fishing from the Izmir Bay during the fishing period of 2020-2021. The samples were dissected in the laboratory, 15 ovaries from mature females were taken and fecundity was determined. The minimum, maximum and mean values of total length of Scomber scombrus respectively; It is 26.9-31 cm (mean: 28.89±1.44), 150 420-454 260 (mean: 259 300±97 369). A linear relationship was determined between total length fecundity in mackerel and this value was TL=25.396+0.000013×F (R2=0.83). Oocyte diameter; It is 1.1-.2 (mean: 1.16±0.10) mm. The minimum, maximum and mean values of total length of Scomber colias respectively; it is 25.1-25.9 cm (mean: 25.59±0.26), fecundity 134 853-417 500 (mean: 322 381±84 172). A linear relationship was found between total length fecundity in lichen and this value was TL=24.682+0.000003×F (R2=0.80). Oocyte diameter; It is 0.99-1 (mean:1.02±0.06) mm.
