The effects of storage temperature and storage time on the somatic cell count of Anatolian buffaloes

This research examined the effects of storage temperature and storage time on the somatic cell count (SCC) of milk from Anatolian buffaloes, which was measured with the DeLaval cell counter (DCC). Storage temperature and time are among the different factors that potentially affect the SCC of Anatolian buffalo milk. In this context, 20 milk samples were collected from Anatolian Buffaloes and analyzed. The milk samples were divided into two groups according to their measured level of SCC. These two groups were the low score (LT= 3.16 cell/ml) group and the high score (GT 3.16 cell/ml) group. The mean logSCC values of the low score and the high score groups were determined as 2.27+-0.045 and 4.06+-0.019 cells/ml, respectively. In this research, the effects of storage temperature (4 deg C, 21 deg C) and storage time (fresh milk, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12 and 24 h) on logSCC were determined to be statistically significant (P LT 0.01). Thus, increases in storage temperature and storage time were associated with an increase in the logSCC of the milk samples. In this study, the effects of storage temperature (4 deg C, 21 deg C) and storage time (fresh milk, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12 and 24 hours) on logSCC were determined to be statistically significant (P LT 0.01). Thus, increases in storage temperature and storage time were associated with an increase in the logSCC of the milk samples.

Buffalo milk is the most preferred commodity in Pakistan due to high fat contents. Hydrolysis of circulating triglycerides and uptake of fatty acids in the mammary gland is regulated by lipoprotein lipase (LPL). This study is designed to identify single nucleotide polymorphisms (SNPs) in LPL gene among high and low milk producing buffalo breeds of Pakistan. We selected samples (n=50) of each Nili-Ravi a high milk producing and Azakheli a low milk producing buffalo breeds. Blood samples were collected for DNA extraction. LPL region of exon 2 region along with exon/intron boundaries were sequenced and data was analyzed for variation detection. Allele frequency was calculated using Hardy-Weinberg equation and in-silico analysis was performed for functional prediction and genetic diversity assessment. We found one single nucleotide polymorphism c.258+43C GT T in the intronic region of LPL gene. This polymorphism followed the Hardy-Weinberg equilibrium in both Nili-Ravi (P-value = 0.538) and Azakheli (P-value = 0.077). Association of T-allele analysis predicted it significantly higher (P-value = 0.009) in Nili-Ravi. We found one novel SNP (c.258+43C GT T) in the LPL gene in both high and low milk producing buffalo breeds of Pakistan but high in Nili-Ravi. Lower splice site effect suggests its less strength of association with milk producing trait.

The study reports the physicochemical content of raw and pasteurization milk from Bhadawari, Mehsana and Nagpuri Indian buffalo breeds. The study demonstrated the protein, fat, lactose, total solid content, and percentage of casein of the raw milk for Nagpuri, Mehsana and Bhadawari as 4.5+-0.51, 5.4+-0.62, 4.8+-0.41, 7.2+-0.32, 6.1+-0.66, 7.0+-0.84, 5.4+-0.65, 6.2+-0.65, 5.1+-0.54, 17.2+-1.21, 18.8+-1.54, 18.1+-1.22, 10.8+-1.20, 11.4+-1.02 and 11.5+-0.98 respectively. The study further extended to determine the growth profile of Lactobacillus plantarum (LAB) isolated from sheep milk, in the milk of different buffalo Indian breeds. The isolate was identified as a LAB by 16S rRNA sequencing technique. These LAB thrived in the different milk were characterized by colony forming unit (CFU). The study finally revealed that CFU count was found significantly higher (P LT 0.05) in Mehsana milk as comparative to other milk treated samples of Nagpuri and Bhadawari buffalos.

The present study was conducted to investigate the normal somatic cell count (SCC), associated factors in hygienic management and to define subclinical mastitis in buffaloes in Puducherry. Samples were collected from 50 clinically normal buffaloes stationed at different farms of Puducherry region. Somatic cell count was measured for all the milk samples using Newman-Lampert staining technique. The upper limit of SCC was determined as GT= 2, 00,000/ml of milk based on the mean +- 2SD of a total SCC. California mastitis test (CMT) was conducted for all the animals. In the present study subclinical mastitis was diagnosed on the basis of samples with SCCs GT= 2, 00,000/ml. In the present study 96 percent of the milk samples were negative for California mastitis test (CMT). The SCC of CMT positive quarter was significantly higher (P LT 0.01) than CMT negative quarters. Subclinical mastitis was found only in three buffaloes in the present study. Somatic cell counts were significantly higher in the buffaloes with subclinical mastitis. Regarding the hygiene, all the farms had poor management and improper housing. Proper education on hygienic management is need of the hour for buffalo owners of Puducherry region.

Milk fatty acid composition of Murrah buffaloes was determined in present study. Samples were collected from 10 lactating buffaloes and were analysed for fatty acid profile using AOCS official method. Murrah milk fat was having 71.6 percent saturated fatty acids (SFA), 27.97 percent unsaturated fatty acids. C16:0, C18:1c, C18:0, C14:0 and C12:0 were the five most abundant fatty acid (82.5 percent of total fatty acids) in the Murrah milk. Palmitic acid, myristic acid (14:0) and stearic acid (18:0) together constituted approximately 85.8 percent of saturated fatty acids by weight. Short chain fatty acids (C4:0, C6:0), medium chain fatty acids (C8:0, C10:0, C12:0), and long chain fatty acids (C16:0, C18:0, C16:1, C18:2) were 1.82, 4.56 and 49.96 g/100 g respectively. Mono-unsaturated fatty acid were 26.79 percent of the fatty acids in milk, mostly oleic acid (18:1). Poly-unsaturated fatty acids constitute about 1.18 percent by weight of the total fatty acids. Linoleic acid (18:2) and alpha-linolenic acid (18:3) accounted for 0.88 and 0.30 percent by weight of the total fatty acids.

Data on 4971 test-day milk yield traits [milk (TDMY), fat (TDFY) and protein (TDPY)] for 691 Egyptian buffalo cows, daughters of 120 sires and 532 dams were used for fitting the lactation curve parameters of milk, fat and protein yields using multiple-trait animal model. The random effects included in the model were direct additive genetic, permanent environment and error, while the fixed effects were herd test-day, year and season of calving and parity as well as days in milk as a covariable. The means for TDMY, TDFY, TDPY and lactation curve parameters [initial (a), ascending slope (b), descending slope (c), persistency (P), and maximum milk production during lactation (Ymax) and the peak test-day (PY)] were estimated. Heritabilities of TDMY, a, b, c, P, PY and Ymax were 0.22, 0.37, 0.38, 0.39, 0.37, 0.37 and 0.38, respectively. The corresponding heritabilities for TDFY and fat curve parameters were 0.21, 0.41, 0.40, 0.39, 0.38, 0.36 and 0.42, while the estimates for TDPY and protein curve parameters were 0.22, 0.38, 0.40, 0.40, 0.38, 0.40 and 0.43, respectively. Genetic correlations among TDMY and curve parameters of a, b, c, P, PY and Ymax were 0.31, -0.23, -0.34, 0.52, 0.48 and 0.87, respectively. Genetic and phenotypic correlations between milk yield traits were high (mostly of 0.83 and 0.99), like the correlations between curve parameter in milk, fat and protein (from 0.71 to 0.96). In practice, genetic selection for lactation curve parameters (a, P and Ymax) in the Egyptian buffalo would improve total milk yield traits.

An attempt was made to know the management system of intensive buffalo farming and buffalo derived -problems under semi-arid area of Bangladesh. Direct interview with farmers, farm observation, community discussion and key informant interview (KII) had been followed over six months in Dinajpur district. It was observed that buffaloes were reared under two types of management system namely household yard intensive (HYI) and completely intensive (CI). In HYI system, farmers purchased heifer or pregnant buffalo from local market and sold mother with calf after one year. Buffaloes were kept shed at night and maintained feeding, showering and other activities in the household yard at day. Those were showered by once-two times/week in cool day, but most of them were showered in a day. On the other hand, in CI system, buffaloes were reared inside the shed all time and maintain feeding, showering and other activities in the shed. Buffaloes were showered twice in a day in winter period while three times in summer period. Average milk production for each buffalo was 5 to 8 liter/day. Major productive and reproductive traits e.g. lactation Length (day), average milk yield (L/day), dry period (day), age at first pregnancy (month) and age at first calving (month) were found better in CI system than that of HYI system and those were found highly significant (P LT 0.001). Significant difference were found on total solids, fat and protein content but no significance difference were observed on SNF, lactose and ash of buffalo milk between two farms. Insignificant difference (P GT 0.05) was found for the value of electric conductivity (ECms/cm) that showed both farms produced healthy milk (3.25+-0.49 vs 3.09+-0.89). Both systems showed as a profitable venture (BCR 1.33 vs 1.25). Problem matrix revealed that the top ranked order of problem were "inadequate knowledge on buffalo rearing", "high price of feed" and "low milk price" mentioned as 1, 2 and 3. Shortage of feeds and fodder, high temperature and lack of cow boy were ranked as 4, 5 and 6. Limited breeder bull, lake of AI workers and inadequate quality semen and credit facilities mentioned as 7 ranked problem. It was concluded that ensuring training and marketing could help to improve intensive system of dairy buffalo that meet up the growing demand of milk in urban people. Simultaneously AI facilities with quality semen, HYV fodder cultivation and credit facilities should be promoted for sustainable dairy buffalo farming.

This study aimed to determine the lactoferrin concentration from healthy and subclinical mastitic buffaloes. Lactoferrin levels were determined in a total of 475 mammary quarter milk samples, 391 of which were healthy and 84 suffering from subclinical mastitis. The mean milk lactoferrin level in logarithmic form for healthy and subclinical mastitic milk samples was 1.34+-0.27 and 1.85+-0.47, respectively. A significant difference (p LT 0.001) was determined between the two groups in terms of the lactoferrin levels. The milk lactoferrin levels in healthy buffaloes were significantly higher in the middle and late periods of lactation than the peak period (p LT 0.05). The mean milk lactoferrin levels in subclinical mastitic buffaloes infected with S. aureus or S. agalactiae were significantly higher than those from in buffaloes infected with Coagulase-Negative Staphylococci (CNS) or Candida spp. (p LT 0.05). An increase in milk lactoferrin levels was observed in parallel with the increase in somatic cell scores. In conclusion, milk lactoferrin levels in buffaloes were found to increase with the lactation stage, the presence of bacterial infection and the somatic cell count. This is the first study to determine the relationship between lactoferrin concentrations and bacterial infection in milk obtained from mammary quarters with subclinical mastitis in Anatolian buffaloes.

The objective of the present study was to identify the physico-chemical differences between experimentally fraudulent cheeses and those produced exclusively with cow and buffalo milk and to sensorially assess the consumer's perception of these products. Three types of cheese were produced using buffalo milk and cow milk as raw materials at different proportions, and total lipids were analysed; protein ash, moisture and carbohydrate contents were measured; energy values were determined; and affective sensory and purchase intent analyses were performed. The results indicated that 57 percent and 59 percent of the judges, respectively, reported liking the colour and texture of the mixed cheese very much and that they would certainly buy it. Regarding the composition, the fraudulent cheese had a lower energy value compared to the cow and buffalo cheeses and that the protein content of the fraudulent cheese was lower than that of the buffalo cheese. There were no significant differences in the carbohydrate or fat contents among the analysed products. Compared with the cow cheese, the mixed cheese and buffalo cheese had higher moisture contents. It was concluded that although the mixed cheese presented significant physico-chemical differences, it was considered acceptable product because consumers showed intent to purchase the mixed cheese.

The present study was conducted in buffaloes to determine the occurrence of subclinical mastitis and its diagnosis in relation to milk SCC and biochemical composition. The results showed that taking quarter foremilk (QFM) somatic cell count (SCC) threshold limit of LT 200*10**(3) cell for defining healthy quarters, 16.48 percent of buffaloes and 5.79 percent of quarters were positive for specific subclinical mastitis. In total, 57/363 (15.70 percent)of quarters were bacteriological positive with 21 (5.79 percent) representing specific and 36 (9.92 percent) latent infections. The major pathogen isolated were 28 (49 percent) coagulase-negative staphylococci followed by 16 (28 percent) S. aureus, 09 (16 percent) Streptococcus and 4(7 percent) Corynebacterium spp. The average QFM SCC was found as 52.50*10**(3) cells /ml for the quarter with no bacteria, and 356.51*10**(3) cell /ml for the quarters infected with one or the other bacteria (p LT 0.05). The milk electrical conductivity (EC), Fat and Lactoseshowed significant alterations with the quarter infection, the EC and Fat increased while Lactose decreased in infected quarters (p LT 0.05). The California mastitis test CMT) at cut off score of GT 0.5 for diseased quarters showed 68.75 percent sensitivity with 99.09 percent specificity. The EC with 87.50 percent sensitivity and 74.02 percent specificity and Lactose with 84.37 percent sensitivity and 66.16 percent specificity also showed significant (p LT 0.01) discrimination ability to differentiate healthy and mastitis quarters in buffaloes. The milk pH was not found to be a good parameter to identify diseased quarters.
