VERTICAL AND HORIZONTAL RESISTANCE OF F5:6 PROGENIES OF CAROTENOID-BIOFORTIFIED LETTUCE TO Bremia lactucae

ABSTRACT The use of resistant cultivars is one of the strategies for downy mildew management. The objective of this study was to evaluate the vertical and horizontal resistance of F5:6 progenies of carotenoidbiofortified lettuce to virulence phenotypes of Bremia lactucae 63/63/51/00, 63/31/19/00 and 63/63/19/00. The experimental design was completely randomized and subdivided into plots that were evaluated over time. In the plots, 12 genotypes of lettuce were used, and the subplots were monitored over time (7th to the 18th day after inoculation). For each virulence phenotypes of Bremia lactucae, a separate experiment was performed with three replicates. To select resistant genotypes, plants were inoculated with distilled water, sporangia removed from infected tissues and Tween 20. The genotypes were evaluated when sporulation appeared on cotyledonary leaves of the susceptible cultivar Solaris, verifying the proportion of necrotic and sporulated plants. There was a correlation between genotypes and times for all virulence phenotypes evaluated. Genotypes UFU-189#2, UFU-206#1, UFU-215#3 and UFU-215#14 showed vertical resistance to virulence phenotypes 63/63/51/00, 63/31/19/00, and 63/63/19/00 of B. lactucae. Horizontal resistance levels were observed in genotype UFU 206#1 for virulence phenotypes 63/63/51/00 and 63/31/19/00; in genotype UFU-66#7 for virulent phenotype 63/31/19/00; and in genotype UFU-215#10 for virulence phenotype 63/63/19/00.

ABSTRACT Loss of genetic variability in natural populations affects negatively the adaptive and evolutive process of plant species. In this context, the objective of this work was to evaluate the existing genetic variability between and within natural populations of P. platycephala, using biometric characters of fruits and seeds and physiological characters of seeds and seedlings, and, thus, assess the potential for selection of mother trees, indicate areas for seed collection and use, and generate support for conservation and pre-breeding of plant species. A completely randomized experimental design was used, with 45 treatments (progenies) and four replications of 25 seeds. The progenies were evaluated in laboratory, using seeds collected from 45 mother trees sampled in three natural populations. The following characters were evaluated: seed germination percentage, germination speed index, and mean germination time; shoot and main root lengths; and seedling shoot and root dry weights. The data were subjected to RELM/BLUP analysis for estimating genetic parameters and correlations and to analyses of genetic divergence (UPGMA). The populations present genetic differences to each other and a significant variation between progenies for all characters evaluated, and the variation was greater within the populations. The characters evaluated present high genetic control; therefore, they can be used in analysis of genetic variability of populations and progenies of P. platycephala, thus assisting in the identification of areas for seed collection, conservation programs, and pre-breeding of the species.

ABSTRACT In precision agriculture, determining management zones for soil and plant attributes is a complex process that requires knowledge of several variables, which complicates management and decisionmaking processes. This study evaluated the spatial variability of soybean yield and soil chemical properties using geostatistical and multivariate analyses to define management zones in an Oxisol. The soybean yield and soil chemical properties between 0 to 0.2 and 0.2 to 0.4 m soil depths were sampled at 70 points. Geostatistical and multivariate analyses were then performed on these data. The soil chemical properties showed higher variability at 0.2 to 0.4 m soil depth. The semivariogram parameters of the principal component analysis (PCA) data (PCA 1, PCA 2, and PCA 3) for both depths were more homogeneous than the original data. The maps of soil chemical properties showed high similarity to the soybean yield map. The PCA explained 65.34% (0 to 0.2 m) and 70.50% (0.2 to 0.4 m) of data variability, grouping the soybean yield, organic matter, pH, phosphorous, potassium, calcium, magnesium, and sodium. PCA spatialization allowed for the definition of management zones indicated by PCA 1, PCA 2, and PCA 3 for both depths. The result indicates that the area must be managed using different strategies of soil fertility management to increase soybean yield.

ABSTRACT Forage cactus (Opuntia ficus-indica Mill) is an important forage resource in the Northeast region of Brazil due to its adaptation to the edaphoclimatic conditions of the region, mainly to the drought periods. The objective of this work was to assess the effects of diets with forage cactus combined with different sources of fibers on dry matter and water intakes, digestibility, microbial protein production, balance of nitrogen compounds, and performance of Red Sindhi cows in the Semiarid region of Brazil. The silages used were prepared from plants of Atriplex nummularia Lindi., Sorghum bicolor (L.) Moench, Gliricidia sepium (Jacq.) Walp., and Manihot pseudoglaziovii Pax & K. Hoffm. They were included in forage cactus-based diets at a roughage to concentrate ratio of 58:42. The cows were distributed in a 4×4 double Latin square design with 12 days for each period. Water intake presented differences (p<0.05) when using the diet with A. nummularia silage (19.42 L d-1). The diet with S. bicolor silage resulted in lower (p<0.05) coefficient of digestibility of dry matter (48.71%) than the diet with A. nummularia silage (68.46%). The diet with A. nummularia silage resulted in higher (p<0.05) microbial synthesis than that with G. sepium silage, 115.01 and 80.07 g CPmic kg-1 TDN, respectively. The silages evaluated, combined with forage cactus, can be used as diets for lactating Red Sindhi cows without affecting the daily dry matter intake, milk production, fat content, microbial protein synthesis, and balance of nitrogen compounds.

ABSTRACT Considering the losses of phosphorus (P) in the soil by the adsorption process and that the use of biochar seems promising from the agricultural point of view, the aim of this research was to evaluate P adsorption by the Ultisol incubated with different biochar doses through kinetic and isothermal study. The experiment was carried out with poultry litter biochar pyrolyzed at 350 °C. Ultisol was incubated with increasing biochar doses, from 0.0 to 61.95 t ha-1. After 60 days, the phosphorus adsorption rate, kinetics and adsorption isotherms were evaluated. The results showed that the soil had the highest capacity to adsorb phosphorus (1.105 mg g-1). Biochar was not a good P adsorbent; regardless of the applied doses, it released P to the equilibration solution. Langmuir model was better at describing the adsorption of P. The pseudo-second order model fitted well to the adsorption kinetics of P, showing that the adsorption is chemically controlled. These results suggest that biochar can increase the availability of P, limiting adsorption; therefore, it could be used as a fertilizer and/or soil conditioner.

ABSTRACT This study aimed to validate the artificial hybridization without emasculation in lima beans, identifying F1 generation using microsatellite markers (SSR). We used four accessions from the Active Germplasm Bank of Phaseolus, Universidade Federal do Piauí (BGP-UFPI), with contrasting growth habits and days-to-maturity traits. The 241 crosses performed had a percentage of hybrid pods of 5.8%, obtaining 42 seeds in F1. Artificial cross-fertilization was confirmed by microsatellite markers (BM 211, BM 141, BM 154, and GAT 591), and four plants were identified as hybrids. Thus, the proposed method is effective in performing artificial crosses for lima beans.

ABSTRACT Handroanthus impetiginosus is a tree species with ecological and economic potential. Despite that, in the Brazilian market, its wood is heavily exploited in the illegal trade. Therefore, studies on genetic diversity are necessary in order to propose strategies for conservation of this species. Thus, the aim of this study was to select Inter Simple Sequence Repeat (ISSR) primers for genetic diversity studies applied to the forest population of H. impetiginosus. For this, 30 ISSR molecular primers were tested in 30 individuals, evaluating the total number of loci, polymorphism rate and polymorphic information content, as well as marker index and resolving power. Eight primers were selected for having a better amplification pattern, which provided 62 loci. The polymorphic information content of the primers ranged from 0.34 to 0.49, while the marker index (MI) averaged 3.20, with resolving power (RP) of 2.40, Nei’s diversity (He) of 0.35 and Shannon index (I) of 0.52. The results show that the primers UBC 807, 809, 818, 824, 857, 860, 873 and 881 are efficient for quantifying the genetic diversity of H. impetiginosus. These results can contribute to supporting strategies aimed at the conservation of this species and selection of parent trees.

ABSTRACT Family farming is important for the production of food for human consumption, but it lacks resources to invest in the soil fertility of its crops. The objective of this study was to analyze whether the use of green manures increases soil fertility in terms of the nutrients N, P and K with low environmental risk for family agricultural production. An experiment was set up in the State of Mato Grosso (Brazil), in randomized complete block design, with a 6 x 3 factorial scheme, four replications, corresponding to 6 types of plants and 3 doses of phosphorus. Leguminous green manure plants were: velvet bean, Crotalaria juncea, Crotalaria ochroleuca and dwarf pigeon pea, in addition to plots with pearl millet and weedy fallow control. Rock phosphate (i.e., reactive natural phosphate) was applied as a source of phosphorus at 0, 50 and 100 kg P2O5 ha-1. The species used as green manure were cut in full bloom and the levels of N, P, K and C in plants and soil were determined. Leguminous green manure plants, after mineralization, increased the stocks of N, P and K in the soil. Crotalaria juncea was the one that reached the highest N accumulation in its constitution, being recommended in the intercropping with species with C/N ratio > 30 to minimize risks of contamination with nitrate in the soil. This work concluded that the managements with green manure plants, especially Crotalaria ochroleuca, are recommended and environmentally safe technical alternatives for family farming.

ABSTRACT Morphoagronomic characterization is a basic requirement to identify a phenotypic profile of a population. The quantification of variability allows efficient selection of superior and divergent genotypes. Thus, this study aimed to estimate the variability among 490 genotypes and seven strains, from an F2 population of Ricinus communis L., in 35 morphoagronomic traits and 12 agronomic traits. For qualitative descriptors, the entropy technique was used in the percentage frequencies of each category, computing its level using the coefficient of Rényi (1961). Quantitative descriptors were subjected to analysis of variance by the F test, and Tukey test was performed at 1% probability level. Of the morphoagronomic traits used, 13 were related to plants, nine were related to inflorescence, six were related to fruits and seven were linked to seeds, in addition to 12 agronomic traits. The material was arranged in the field with families (strains of five families) interspersed with their respective parents (controls). Stem color, shape and number of racemes collected, main color, type of secondary color and hundred-seed weight have high variability in the population, with formation of 68 groups as a function of genetic similarity. The possibility of selection as to the number of racemes harvested is clear, so it is possible to identify genotypes with higher number, aiming to enhance crop yield.

ABSTRACT The knowledge of genetic diversity in plant population is essential to the success of plant breeding programs. Thus, previous characterization of landraces is one of the first steps before the indication of genotypes to these plant breeding programs. This study aimed to characterize, through morphological traits, and estimate the genetic diversity in landraces of lima bean (Phaseolus lunatus L.). Genetic diversity was estimated by using multivariate methods that selected individual landraces to be indicated for farmers and consumers. According to morphological traits proposed by Biodiversity International, the genotypes UFPI-667 and UFPI- 682 showed higher dissimilarity and high potential to be used in crosses since they are genetically distant and complementary. The principal component analysis identified positive and significant phenotypic associations between variables: pod length, pod width, and grain production. According to the multivariate methods used, the landraces UFPI-666, UFPI-650, UFPI-651, UFPI-687, UFPI-658, UFPI-673, UFPI-667, and UFPI-674 are potential candidates for selection since they have relevant features for lima bean breeding, such as shorter cycles and higher grain production. This study showed that multivariate analysis can be used as an effective tool to find potential traits in lima bean and can assist the lima bean breeders in the selection of potential landraces. Accessions UFPI-667 and UFPI-682 can be indicated as genitors for breeding programs, as they are genetically distant and complementary in their characteristics.
