Change in state of components of the Bg-rbg system of maize transposable elements and rapid genetic changes [Zea mays L.]

The Bg-rbg system of maize transposable elements controls instability of the mutable allele o2-m(r) at the Opaque2 (O2) locus. Reversion of the o2-m(r) allele to the normal O2 allele before the first division of the primary endosperm nucleus gives rise to phenotypically normal kernels (whole endosperm revertants or WER). Disruptive selection for WER frequency led to the isolation of changes in state of both the responsive allele o2-m(r) and the regulatory element Bg. While the state of the responsive alleles conditions the frequency of reversion to the normal allele, the state of the regulatory element affects the dosage dependence of the responsive allele on the copy number of Bg. Excision frequency of rbg (receptor of Bg) depends on the copy number of Bg-bf, the regulatory element present in lines with high WER; such dosage effect is not observed for the regulatory element Bg-bf present in lines with low frequency of WER. High WER frequency lines are characterised by higher kernel weight and volume, higher number of leaves and delayed flowering of male inflorescences in comparison with low WER frequency lines. We propose that transposable elements determine rapid genetic changes in organisms in presence of selection factors affecting the traits whose expression is controlled by transposable elements | [Il sistema Bg-rBg degli elementi trasponibili del mais controlla l'instabilita' dell'allele mutabile o2-m(r) al locus Opaco2 (O2). La reversione dell'allele o2-m(r) all'allele O2 normale prima della prima divisione del nucleo dell'endosperma primario da' luogo a cariossidi fenotipicamente normali (revertant per l'intero endosperma o WER. La selezione dirompente per la frequenza di WER portava all'isolamento di cambiamenti di stato tanto dell'allele rispondente o2-m(r) quanto dell'elemento regolatore Bg. Mentre lo stato degli alleli rispondenti condiziona la frequenza della reversione all'allele normale, lo stato dell'elemento regolatore influenza la dipendenza del dosaggio dell'allele rispondente sul numero di copie di bg. La frequenza di escissione di rbg (recettore di Bg) dipende dal numero di copie di Bg-bf, l'elemento regolatore presente in linee con elevate WER; tale effetto di dosaggio non si osserva per l'elemento regolatore Bg-bf presente in linee con bassa frequenza di WER. Le linee ad alta frequenza di WER sono caratterizzate da peso e volume piu' elevato delle cariossidi, da numero piu' alto di foglie e da fioritura ritardata per quanto concerne le infiorescenze maschili rispetto alle linee con bassa frequenza di WER. Noi ipotizziamo che gli elementi trasponibili determinino rapidi cambiamenti genetici negli organismi in presenza di fattori selettivi influenzanti i caratteri la cui espressione e' controllata da elementi trasponibili]
