Shoot bud formation and plantlet regeneration from the basal tissues of mulberry [Morus alba] leaves

Shoot bud formation was induced in the basal tissues of mulberry leaves using a two-step culture system: For achieving a high frequency of multiple shoot bud formation, it was essential that seedlings were cultured on a medium supplemented with a combination of 6-benzylaminopurine (BA) and 2, 3, 5-trilodobenzoic acid (TIBA), where after the basal leaf tissues were excised from the seedlings and cultured on a medium containing both thidiazuron (TDZ) and TIBA. It was clearly demonstrated that TIBA exerts a promotive effect on the induction of shoot buds when applied together with cytokinin. The shoot buds were formed as a mass on the entire surface of the midrib under the above mentioned conditions. From the shoot buds thus obtained, complete plantlets with profuse adventitious roots were produced in vitro
