Utilization of various methods of cytotoxicity detection in aquatic toxicology

Three assays were used for checking the cytotoxic effect of a model substance (K2Cr2O7) to the fish cell lines EPC, RTG and FHM. The assays were used as follows: NR assay based on the principle of accumulation of neutral red dye in lysozomes of animal cells, MTT assay detecting live cells by means of the activity of mitochondrial dehydrogenase; and Cell Death Detection ELISA for checking the apoptosis by determining the amount of DNA fragments in cell cytoplasm. Our results lead to the hypothesis that the first toxic effects of potassium dichromate affect the mitochodrial apparatus and trigger apoptotic changes.
