Relation between cell growth and phytosterols in cell cultures of Ginkgo biloba, 2: Effects of mevalonic acid metabolites

The relationships between metabolites formed by the mevalonic acid pathway and cell growth in suspension cell cultures of Ginkgo biloba were investigated. Treatment of zaragozic acid, a specific sterol synthesis inhibitor, reduced cell growth and beta-sitosterol production. Addition of beta-sitosterol restored cell growth. Treatment of compactin, a specific inhibitor of mevalonic acid synthesis, reduced both cell growth and beta-sitosterol production. The addition of mevalonic acid restored cell growth, although that of beta-sitosterol did not. Treatment of tunicamycin or chaetomellic acid sodium salt, inhibitors of the synthesis of modification proteins, caused a decrease in cell growth. These results indicate that beta-sitosterol is one of the indispensable factors for cell growth, while the other metabolites formed by the mevalonic acid pathway might participate in the cell growth.
