Preliminary assessment of correlation between T-lymphocyte responses and control of porcine reproductive and respiratory syndrome virus (PRRSV) in piglets born after in-utero infection of a type 2 PRRSV

A preliminary study into the protective mechanisms of adaptive immunity against porcine reproductive and respiratory syndrome virus (PRRSV) in piglets (n = 9) born to a gilt challenged intranasally with a type-2 PRRSV. Immune parameters (neutralizing antibodies, CD3+ CD4+ , CD3+ CD8+ , CD3+ CD4+ CD8+ T-lymphocytes, and PRRSVspecific interferon (IFN)-γ secreting T-lymphocytes) were compared with infection parameters (macro- and microscopic lung lesion, and PRRSV-infected porcine alveolar macrophages (CD172α+ PRRSV-N+ PAM) as well as with plasma and lymphoid tissue viral loads. Percentages of three T-lymphocyte phenotypes in 14-days post-birth (dpb) peripheral blood mononuclear cell (PBMC) had significant negative correlations with percentages of CD172α+ PRRSV-N+ PAM (p 0.05) as well as with macroscopic lung lesion (p 0.01). Plasma and tissue viral loads had significant (p 0.05) negative correlations with CD3+ CD4+ CD8+ T-lymphocyte percentage in PBMC. Cytogenetic and hematological analysis was performed in peripheral blood obtained from pigs bred in the high background radiation areas (HBRA) (Cheongwon-gun and Boeun-gun) and a control area. The frequencies of gamma-ray induced micronuclei (MN) in the cytokinesis-blocked (CB) lymphocytes at several doses were measured in three pigs. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 mGy to 1,969 mGy.

Cytogenetic and hematological analysis was performed in peripheral blood of cattle in the vicinity of Uljin nuclear power station and control area. The frequency of micronuclei(MN) in peripheral blood lymphocytes from cattle was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy.

A 4-year-old female Cocker spaniel was presented with respiratory distress and abdominal distension. Pleural effusion, ascites, hepatosplenomegaly, and superficial lymphadenopathy were observed and multicentric lymphoma was diagnosed by cytological examination. Immunophenotyping of lymph node and bone marrow using polymerase chain reaction for antigen receptor rearrangement identified a stage V lymphoma origination from T-cell. Despite of systemic chemotherapy using L-asparagenase, vincristine, cyclophoaphamide and prednisolone, neurologic deficits came out and progressed. Cerebrospinal fluid analysis revealed neoplastic lymphocytic pleocytosis indicating central nervous system involvement of lymphoma. The postmortem diagnosis was confirmed based on the histology and imunohistochemistry.

A 7-year-old Jeju native boar was requested to the Veterinary Pathology Laboratory of Cheju National University with a clinical signs of anorexia, melena, lethargy and sudden death. At necropsy, four coalescing firm masses were occupied in the abdominal cavity between ventral surface of stomach and pancreas. Individual mass was a yellowish white in color and up to 6 cm in diameter. These masses were not encapsulated and bulged from the cut surface. Liver and spleen were enlarged and pale nodules were presented in these tissues.

Hematologic investigations were made on the blood samples taken from bovine leukemia virus (BLV)-seropositive Holstein-Friesian cattle in Korea, and their absolute lymphocyte count was compared with that of BLV-seronegative cattle. The incidence of persistent lymphocytosis (PL) was also determined. The normal bovine lymphocyte count was established on the basis of studies of 656 blood samples taken three times from 297 seronegative animals aged from 0~6 months to over 5 years at 5~6-month intervals. The data were examined according to 7 age groups of samples placed into their respective age groups.

The alkaline single-cell gel electrophoresis (SCGE) assay, called the comet assay, has been applied to detect DNA damage induced by a number of chemicals and biological factors in vivo and in vitro. The DNA damage was analysed by tail moment (TM) and tail length (TL), which were markers of DNA strand breaks in SCGE. Human, mouse and rat peripheral blood lymphocytes (PBLs) were irradiated with different doses of ∨60Co γ-rays, e.g. 1, 2, 4, and 8 Gy at a dose rate of 1 Gy/min. A dose-dependent increase in TM (p less than 0.01) and TL (p less than 0.01) was obtained at all the radiation doses (1-8 Gy) in human, mouse and rat PBLs.

The purpose of the present experiment was to investigate the micronuclei(MN) frequency in cytokinesis-blocked(CB) cells after various doses of gamma-rays in pig (Landrace, male, 3-month-old) and so to contribute to the clarification of the question whether these species are suitable as a target organism in the test system. The frequencies of binucleated cells, and gamma-ray-induced MN in CB cells at several doses were measured in three donors. The peaks of binucleated lymphocyte formation(22%) were found at a concentration of 2% phytohaemagglutinin(PHA) and 4 ㎍/ml Cytochalasin B(Cyt-B) in pig at 72 hours after incubation.
