A Fluorescence resonance energy transfer (FRET) real-time reverse-transcription (rRT-PCR) assay was developed that distinguishes stains of South African and European highly pathogenic (HPAI) from low pathogenicity (LPAI) H5 avian influenza viruses in the absence of virus isolation, irrespective of the length of insertion at the hemagglutinin cleavage site (H0). The assay was used to pathotype H5-type viruses detected by rRT-PCR in ostrich tracheal swabs collected during the 2006 HPAI H5N2 outbreak in the Western Cape Province.

Onderstepoort Journal of Veterinary Research, T5:95-102 Sera from 19 wild caught vultures in northern Namibia and 15 (12 wild caught and three captive bred but with minimal histories) in North West Province, South Africa, were examined by an enzyme-linked immunosorbenats say( ELISA)f or antibodiesto the Bacillus anthracis toxin protective antigen (PA). As assessed from the baseline established with a control group of ten captive reared vultures with well-documented histories, elevated titres were found in 12 of the 19 (63%) wild caught Namibian birds as compared with none of the 15 South African ones. There was a highly significant difference between the Namibian group as a hole and the other groups (P < 0.001) and no significant difference between the South African and control groups (P > 0.05). Numbers in the Namibian group were too small to determine any significances in species-, sex- or age-related differences within the raw data showing elevated titres in four out of six Cape Vultures, Gyps coprotheress, six out of ten Whitebacked Vultures, Gyps africanus, and one out of three Lappet-faced Vultures, Aegypiust racheliotus, or in five of six males versus three of seven females, and ten of 15 adults versus one of four juveniles. The results are in line with the available data on the incidence of anthrax in northern Namibia and South Africa and the likely contact of the vultures tested with anthrax carcasses. lt is not known whether elevated titre indicates infection per se in vultures or absorption of incompletely digested epitopes of the toxin or both. The results are discussed in relation to distances travelled by vultures as determined by new tracking techniques, how serology can reveal anthrax activity in an area and the issue of the role of vultures in transmission of anthrax.

This studyw as carried out to establish whether cattle can develop resistance to re-infectionby Calicophoron microbothrium by assessing the response of intestinal mucosal globule leukocytese, osinophils, mast cells and basophils, and the establishment of the parasite in the host. A total of 241-year old Tuli steers were randomly divided into four groups of six animals each and infected with C. microbothriumm etacercariae. On the first day of the study, animals in Groups I and II were immunized with 5000 metacercariae and then challenged with 15000 metacercariae on Day 150 post immunization. Animals in Group III were immunized with 15000 metacercariae at the same time that Groups I and II animals were challenged to act as a positive control group Animals in Group IV were left uninfected and acted as a negative control group. Three animals from each group were slaughtered on Day 28 post-challenge and the remainder were slaughtered on Day 42 post-challenge. The established amphistomes were recovered and histopathological and cytological examinations were done on the jejunum, duodenuma, bomasum and the rumen. The establishment rates of the challenge infection in the immunized and challenged groups were lower and ranged from 0 to 0.2% as compared to 6% from naive animals infected as positive controls. Animals immunized and then challenged with C. microbothrium had significantly higher eosinophil, mast cell and globule leukocytes counts in the intestinal mucosa (P < 0.05) as compared to those of the control group. The study indicates that cattle can develop resistance to C. microbothrium re-infection and that eosinophils and mast cells may be important cells in the rejection of the parasite.

It is suggested that Haemophilus paragallinarum requires at least three haemagglutinins for adhesionduring infection. This paper reports the partial purification and characterization of the HA-L haemagglutininfrom H. paragallinarum strain 46-C3, a heat sensitive, trypsin sensitive haemagglutinin that hasbeen shown to be the serovar specific haemagglutinin in this organism. Using the pI and molecularmass obtained, it was shown that this protein shares similarities with other types of adhesins found in Gram-negative bacteria. The haemagglutination assay conditions were optimized at pH 7.5 at 37 °C. It was also shown that activity is enhanced by the addition of Ca2+ and Mn2+ ions.

Malignant theileriosis of sheep is a highly fatal, acute or subacute disease is caused by the tick-borne protozoan parasite, Theileria hirci. In this investigation ten healthy male lambs aged 5-6 months were randomly divided into two groups, A and B and were kept in isolated tick-proof pens. They were treated for internal and external parasite before commencement of the experiment. The lambs were experimentally infected with T. hirci by placing ticks Hyalomma anatolicum anatolicum infected with T. hirci on them. The ticks used in this survey had originally been isolated from sheep and colonies of them were established in an insectarium. Before and after infection rectal temperatures and clinical signs of the lambs were recorded, blood and prescapular lymph node smears were prepared and examined to determine the extent of the parasitaemia, and blood samples were analyzed to evaluate their haemoglobin (Hb) and packed cell volume (PCV) rates. Three days after the commencement of a febrile reaction and appearance of the schizonts in the lymph node smears, treatment of the lambs in Group A with an extract containing the alkaloids of Peganum harmala (wild rue) was commenced. Group B lambs were kept untreated controls. Before treatment there were no significant differences in the rectal temperature, parasitaemia rate, and the Hb and PCV values between animals in the two groups but after treatment significant differences in these values was detected (P < 0.05). After treatment, the clinical signs and parasites in the lymph node smears of the animals in Group A disappeared and they all animals recovered. These parameters in the animals of Group B progressed until their death. Pathological studies showed the characteristic lesions of theileriosis in lambs in Group B, but not in Group A. The results indicate a therapeutic effect of the alkaloids of P. harmala for treatment of ovine malignant theileriosis.

The species composition and geographic distribution of ixodid ticks infesting domestic dogs owned by people in rural communities and villages in Maputo Province was established by collecting ticks from dogs at each of 27 localities spread throughout the province. Ticks were collected from a total of 132 dogs, and nine species belonging to four genera were identified. One dog was infested withs six species, three with five and 13 with four species. Haemaphysalis elliptica followed by Rhipicephalus simus were present on dog sat most localities, and their geographic distribution in Maputo Province has been mapped for the first time.

Foot-and-mouth disease (FMD) is an economically important disease of cloven-hoofed animals that is primarily controlled by vaccination of susceptible animals and movement restrictions for animals and animal-derived products in South Africa. Vaccination using aluminium hydroxide gel-saponin (AS) adjuvanted vaccines containing the South African Territories (SAT) serotypes has been shown to be effective both in ensuring that disease does not spread from the endemic to the free zone and in controlling outbreaks in the free zone. Various vaccine formulations containing antigens derived from the SAT serotypes were tested in cattle that were challenged 1 year later. Both the AS and ISA 206B vaccines adjuvanted with saponin protected cattle against virulent virus challenge. The oilbased ISA 206B-adjuvanted vaccine with and without stimulators was evaluated in a field trial and both elicited antibody responses that lasted for 1 year. Furthermore, the ISA 206 adjuvanted FMD vaccine protected groups of cattle against homologous virus challenge at very low payloads, while pigs vaccinated with an emergency ISA 206B-based FMD vaccine containing the SAT 1 vaccine strains were protected against the heterologous SAT 1 outbreak strain.

The seroprevalence of Anaplasma antibodies in wildlife (eland, blue wildebeest, kongoni, impala, Thomson's gazelle, Grant's gazelle, giraffe and plains zebra) and domestic animal (cattle, sheep and goat) populations was studied in wildlife / livestock interface areas of Kenya. Serum samples were analyzed by competitive inhibition enzyme-linked immunosorbent assay (CI-ELISA), using a recombinant antigen (MSP-5) from Anaplasma marginale surface membrane. A monoclonal antibody, FC-16, was used as the primary antibody, while anti-mouse conjugated to horseradish peroxidase was used as the secondary antibody. The results indicate a high seroprevalence in both wildlife and livestock populations, in contrast to earlier reports from Kenya, which indicated a low seroprevalence. The differences are attributed to the accurate analytical method used (CI-ELISA), as compared with agglutination techniques, clinical signs and microscopy employed by the earlier workers.

2-Phenoxyethanol was used as an anaesthetic to translocate 102 species of fishes representing 30 families from the Sea World aquarium on Durban's beachfront to uShaka Marine World. Most fishes responded well to a final anaesthetic concentration of 0,150 mℓ / ℓ and there were no mortalities.

Body condition scoring (BCS) gives an indication of the nutritional status of an animal and it is thus an invaluable management tool in domestic livestock systems. It is, however, subjective. This study aimed at identifying biochemical indicators which could be objectively used as an indicator of undernutrition in ruminants. Blood samples were collected from 50 cattle with BCS ≤ 1 and from 50 cattle with BCS ≥ 2.5, using a 0-5 scale, and analysed for albumin, urea, creatinine, fructosamine, beta-hydroxybutyrate, non-esterified fatty acids, total serum protein and haematocrit. Rumpfat and ribfat thickness and marbling relative index were determined ultrasonographically in 15 of the low BCS group and 13 of the high BCS group. The laboratory measure with the best predictive ability for severe undernutrition was albumin, which correctly classified 94 % of cattle, using a cut-off of 31.5 g / ℓ. In contrast to a previous study, our study did not find the fructosamine : albumin ratio to be an accurate test to indicate undernutrition in cattle. Ultrasonic measurement of subcutaneous rumpfat and ribfat proved to reliably predict undernutrition, but may, however be impractical for routine use under most field conditions.
