Objective: A cross-sectional study was conducted to reveal the hematology, serum biochemistry, and reproductive behavior of free-roaming cats in the Sylhet Division of Bangladesh. Materials and Methods: Overall, 130 free-roaming cats were captured, of which 52 were Tom (male), 69 were Queen (female), and 9 were kittens. Regarding their ages, the cats were divided into three groups: <1 year as kittens, 1 to 2 years as young, and adults over two years. Thirty blood samples were drawn from the cephalic or saphenous veins of the hind leg for hematological and serum biochemical analysis. Results: The findings revealed that the average hemoglobin concentration was significantly higher in kittens than in young and adult cats (p <1 year as kittens, 1 to 2 years as young, and adults over two years. Thirty blood samples were drawn from the cephalic or saphenous veins of the hind leg for hematological and serum biochemical analysis. Results: The findings revealed that the average hemoglobin concentration was significantly higher in kittens than in young and adult cats (p >< 0.05). In comparison to young and adult cats, kittens had considerably greater red blood cell, packed cell volume, mean corpuscular volume, and platelet counts, whereas mature cats had much higher white blood cell counts. The number of lymphocytes and monocytes was shown to be non-significant, while other components, such as neutrophils, eosinophils, and basophils, varied considerably by age group. Rural cats showed significantly (p < 0.05) higher alanine transaminase and aspartate aminotransferase, whereas urban cats showed higher serum glucose (p < 0.001). The duration of the cat’s estrous cycle was (5.63 ± 1.75) days, as were the inter-estrous phase’s length (7 days), gestation time (66.6 days), and queening rate (65.2). There were 3.92 ± 0.55 kittens per litter. Males reached puberty at a young age of 9.3 months, while females did so at 8.3 months. Conclusion: These findings may contribute to the growing body of research on free-roaming cat populations, which is important for understanding the ecology and behavior of these cats and constructing strategies for their conservation and welfare. Objective: This study aims to explore mutation based on single nucleotide polymorphism (SNP) in the Cholecystokinin B receptor (CCKBR) gene of Pelung chickens. Materials and Methods: We collected DNA samples from 48 Pelung roosters that had won the crowing competition. The CCKBR target encompasses exon 3, intron 3, exon 4, and a part of intron 4, a long 601 bp. This target was replicated using PCR with specific primers that were designed by Primer-BLAST from NCBI. We generated the nucleotide sequence from the PCR product's sequencing results. The SNP analysis was done by BioEdit and MEGA. Genotyping and haplotyp¬ing were done based on nonsynonymous single nucleotide polymorphisms (SNPs) on exons 3 and 4. We calculated allele and genotype frequency, heterozygosity, and Hardy-Weinberg Equilibrium (HWE) using POPGENE 32 programs. Results: This study found three nonsynonymous single nucleotide polymorphisms. The nsSNP in exon 3 alters the coding for the 210th amino acid from serine to asparagine (g.1290 G > A/S210N), while the SNPs in exon 4 alter the coding for the 232nd amino acid from valine to phenylalanine (g.1423G > T/V232F) and the 243rd amino acid that changes the amino acid valine to glycine (g.1457T > G/V243G). The frequency of the mutated alleles is lower than the unmutated alleles. However, the mutation at position g.1457T > G/V243G produces a higher frequency than the unmutated allele. The allele and genotype frequency were not in HWE. It was caused by intensive selection in Pelung chickens, especially for growing capacity. Conclusion: Nonsynonymous mutation on CCKBR may cause variations in the crowing and other traits such as the growth of Pelung chickens. Further studies are needed to explore the CCKBR gene, including the relationship of the gene with the vigor and/or stress level of Pelung chickens. [J Adv Vet Anim Res 2025; 12(1.000): 141-148]

Objectives: The rise of antibiotic-resistant strains of Staphylococcus aureus in dairy milk products is a global concern, compromising treatment efficacy and highlighting the need for innovative solutions. Therefore, a study was conducted to isolate S. aureus strains (N = 21) from raw milk samples of cows infected with subclinical bovine mastitis. Additionally, the resistance of these strains against 12 different antibiotics was examined. Materials and Methods: Sixty raw cow milk samples, 20 from each of three separate dairy farms in Lahore city, were collected and screened for the presence of S. aureus. It was discovered that 70% of these milk samples were contaminated with this bacterium, indicating a widespread presence across the farms. Different isolation tests were employed in this study, including gram staining, capsule staining, catalase, mannitol salt fermentation, DNase, coagulase, and oxidase. Results: The obtained results revealed that the isolated strains of S. aureus showed % of their resistance against different antibiotics in the order of amoxicillin (85%), penicillin (71%), gentamicin (CN) (42%), carbenicillin and trimethoprim-sulfamethoxazole (33%), streptomycin, ciprofloxacin, and oxytetracycline (28%), cefotaxime (10%), and chloramphenicol (4%) in decreasing order, respectively. However, these strains showed no resistance against Bacitracin and Ampicillin. Conclusion: The existence of resistant strains of S. aureus has been attributed to various factors, such as poor milk hygiene, delayed milk transportation, subclinical bovine mastitis among dairy cows, and antibiotic-resistant genes. Thus, our present study will provide useful information about the resistant strains of S. aureus, which may transfer through cows into milk and then produce serious food-borne diseases in human beings. Objective: This study aimed to evaluate how well fresh broiler meat may be preserved in cold storage using chitosan–stearin as an edible coating. Materials and Methods: A completely randomized design with a 3 x 5 factorial arrangement and three replications was employed. Factor I represented the formula dosage (FD) (FD0 = 0% chitosan + 0% stearin; FD1 = 1.5% chitosan + 1% stearin; FD2 = 3% chitosan + 1% stearin), while Factor II represented storage time (ST) (ST0 = 0 days; ST1 = 3 days; ST2 = 6 days; ST3 = 9 days; ST4 = 12 days). Results: The results showed that the water content, cooking loss, protein content, and fat content of broiler meat were significantly affected (p < 0.01) by the FD and ST. Nonetheless, no significant difference (p > 0.05) was observed in the meat's ability to hold water. The broiler meat's pH was significantly affected (p < 0.01) by the FD, but it was not significantly affected (p > 0.05) by the ST. Furthermore, no treatment underwent testing, which revealed the presence of Escherichia coli and Salmonella sp. Conclusion: Chitosan–stearin edible coatings with different formula doses FD and ST consistently preserve the quality of fresh broiler meat during cold storage, with average values of water content ranging from 48.97% to 53.73%, water-holding capacity from 17.52% to 34.30%, cooking loss from 10.03% to 33.19%, pH levels from 4.93 to 5.53, protein content from 14.54% to 17.46%, fat content from 20.55% to 24.21%, and no detectable presence of E. coli and Salmonella sp. Objective: This study aimed to compare two approaches to the brachial plexus: the traditional blind method and an additional technique, both of which use anatomical landmarks to guide needle insertion. Materials and Methods: The traditional and additional approaches were performed on both thoracic limbs of 24 cadavers (24 for each approach). Methylene dye is used for injection and nerve staining. We counted the nerves that successfully stained (staining less than 1 cm). The Mann–Whitney U test was used to compare approaches. Results: The findings indicated that all cadavers were successfully used. The traditional approach and the additional approach revealed success rates of 45.83% and 54.17%, respectively. We found no significant differences between the two techniques (p > 0.05). Conclusion: The additional approach presents a viable alternative to the traditional method for performing the brachial plexus block in goats. Further research into the clinical differences between these techniques could lead to useful insights that help make them more accurate and useful. [J Adv Vet Anim Res 2025; 12(1.000): 64-69]

Objective: This study aimed to assess Aerodramus fuciphagus (Thunberg, 1812) nest's mineral content, cytotoxicity effects, and antioxidant activity. The Edible birds' nests (EBNs) were obtained from Tanah Merah (TM), Tumpat (TU), and Jeli (JE) districts in Kelantan, Malaysia. Materials and Methods: The mineral content was assessed via inductively coupled plasma mass spectrometry. The cytotoxicity effect was evaluated by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide and brine shrimp lethality assays. The antioxidant activity was investigated by 2,2´-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and ferric-reducing antioxidant power (FRAP) assays. Results: Valuable macro- and microelements, including sodium, potassium, aluminum, calcium, magnesium, zinc, iron, and selenium, were detected in the EBN samples. The cytotoxicity test revealed that JE hydrochloric acid (HCl) extract had promising efficacy against HeLa cell lines, with IC50 values of 20.00 ± 0.57 μg/ml. Similarly, TU sodium hydroxide (NaOH) extract inhibited the proliferation of MCF-7 cell lines (IC50 0.03 ± 0.01 μg/ml). Based on the BSL assay, the EBNs were considered safe for consumption as the LC50 values of all the samples were below the toxic level (>1,000 μg/ml). TM EBNs aqueous, TU aqueous, and HCl extracts showed promising antioxidant activity at IC50 values (10 μg/ml) in the DPPH assay. However, the NaOH extract from TU exhib¬ited the highest FRAP value (1.38 ± 0.00 mM gallic acid equivalents gm⁻¹) compared to all other extracts. Conclusion: This study's findings demonstrated the potential of EBN as an alternative anticancer agent and natural antioxidant. [J Adv Vet Anim Res 2025; 12(1.000): 132-140]

Objective: This study intended to figure out the effects of fixed effects and genes such as beta-lactoglobulin (β-LG), prolactin (PRL), annexin A9 (ANXA9), and acetyl-CoA acyltransferase 2 (ACAA2) on the structure of fatty milk acids in Awassi ewes, as well as any potential genotype-genotype interactions. Materials and Methods: Fatty acid (FA) profile and other milk components were examined from 116 Awassi ewes in total. Polymerase chain reaction (PCR) was used to extract and genotype their DNA, and either sequencing or restriction fragment length polymorphism (RFLP) analysis came next. Results: Saturated, medium-chain, and long-chain FA proportions were not significantly impacted by the genotypes of β-LG, PRL, ANXA9, and ACAA2. Conversely, ewes with the β-LG AA genotype displayed higher levels of monounsaturated fatty acids (MUFAs), while the β-LG AB genotype was linked to the highest levels of polyunsaturated fatty acids (PUFAs). It has been demonstrated that PUFA levels are impacted by single nucleotide polymorphisms (SNPs) of ANXA9, while VLCFA and PUFA levels are changed by SNPs of ACAA2. The PRL AA genotype showed the greatest influence on VLCFA. Furthermore, milk exhibited the highest levels of MUFA and PUFA due to the interac¬tion between β-LG and PRL. The interaction PRL-ANXA9 exhibited the greatest levels of VLCFA. Additionally, Dam lambing weight affected the levels of MUFA, MCFA, and long-chain fatty acids. Conclusion: To increase the levels of MUFA and PUFA constituents, the selection program should effectively harness and integrate the synergistic benefits of β-LG, PRL, ANXA9, and ACAA2 as well as their interaction. Objectives: The objective of this study was to assess the effects of varying glucose and trehalose concentrations on tris-citric acid-egg yolk-fructose (TCEF) diluents for the short-term cold storage of goat semen. Materials and Methods: The semen sample was collected, unwashed, and divided into the following groups: control (TCEF without glucose and trehalose), TCEF + glucose (75, 150 mm), and TCEF + trehalose (75, 150 mm). Each experimental sperm group (sperm concentration: 9×107/ ml) was kept at 4°C in a refrigerator. The impact of varying glucose and trehalose levels on the quality of the spermatozoa was assessed at different time points: after dilution and at 5, 24, 48, and 72 h of refrigeration. Results: After dilution, progressive motility (PM), total motility (TM), sperm viability (SV), functional integrity (FI), and acrosome integrity of G-75, G-150, T-75, and T-150 did not differ significantly from the control. The PM, TM, SV, FI, and acrosome integrity of sperm of T-150 were considerably lower than the control, G-75, G-150, and T-75 after 5 and 24 h of cool storage. The T-75 group showed superior PM, TM, and FI after 48 h of cool storage, with noticeably greater values than the other groups. Conclusion: This study indicates that trehalose is a more favorable sugar than glucose for 48 h cool storage of buck semen, providing greater advantages in PM, TM, and PMI. Objective: Investigation of Klebsiella pneumoniae in respiratory-diseased pet cats, estimation of antibiotic sensitivity, and molecular phylogeny of local K. pneumoniae to identify its identity to global isolates. Methods: Totally, 127 feline cases with various respiratory signs were selected for the collection of the nasal swabs that were cultured to isolate K. pneumoniae and detect the antibiotic sensitivity. Further molecular phylogeny of positive K. pneumoniae isolates was done. Results: Findings of culture media and biochemical tests showed that 26.77% of nasal swabs were positive samples for K. pneumoniae. The screening for the antibiotic susceptibility reported a higher sensitivity to ceftiofur, ciprofloxacin, cefepime, amikacin, gentamicin, cefotaxime, and meropenem, as well as ceftazidime, ceftriaxone, and doxycycline, imipenem, as well as clotrimazole and tetracycline. In contrast, the more significant resistant K. pneumoniae isolates were detected to clarithromycin, clindamycin, amoxicillin, cefixime, chloramphenicol, erythromycin, cephalexin, cefadroxil, azithromycin, and nalidixic acid, whereas, significant semi-sensitivity was shown to tylosin. Molecular testing by polymerase chain reaction demonstrated that all isolates were K. pneumoniae. The genetics-based analysis of local K. pneumoniae isolates recorded an overall similarity (95.47%–100%) and changes/mutations (0.0004%–0.0084%), in particular to the National Center for Biotechnology Information-Iraqi isolate (Lc732203.1). Conclusion: This study indicates the high prevalence of K. pneumoniae in respiratory-diseased cats with significant appearance of antibiotic resistance in study isolates. Sequencing data referred to the close related association of study isolates to human K. pneumoniae isolates, suggesting the increased prevalence of nosocomial infections in veterinary medicine. Objective: The objective of this study was to evaluate the integrity of the plasma membrane for human and other domestic animal spermatozoa by the hypo-osmotic swelling test (HOST). Materials and Methods: This study was conducted on spermatozoa of three species, including pigs, rabbits, and goats. Three experiments were conducted on three animal species (pigs, rabbits, and goats) to investigate the factors affecting sperm membranes: osmotic concentration (0, 50, 100, and 150 mOsmol) and treatment time (0, 15, 30, 45, 60, and 90 min) at 37°C. Results: The results showed that treatment time and osmotic concentration influenced the integrity of the sperm membrane in the hypo-osmotic solution. In general, the experiments showed a high rate of sperm reacting to HOST at 50 and 100 mOsmol for 30–45 min. According to this study, the hypo-osmotic solution resulted in a high rate of sperm with swollen tails and a high rate of viable sperm, indicating a correlation between these factors. Therefore, it is necessary to combine both conditions to assess sperm quality. Specifically, the positive rates for the HOST reaction in pig, rabbit, and goat sperm are 46.74%, 58.28%, and 61.95%, respectively. Conclusion: To sum up, the hypo-osmotic solution with a concentration of 100 mOsmol and an incubation time of 45 min is considered the optimal and most feasible condition that can be used for all three species: pigs, rabbits, and goats.
