Adaptation of locally isolated equine herpesvirus (EHV-1) on different cell lines 全文

Locally isolated EHV-1 (2002) was successfully adapted to different available cell lines (BHK, VERO, MDBK and MDCK). Characteristic cytopathic effect (CPE) was observed from the first passage and the virus growth became regular with constant incubation periods at 2nd day in BHK and VERO cells 4th day in both MDBK and MDCK cells post inoculation with maximum titers of 7, 8.5, 7.75, 6.2 log10 TCID50/ml in BHK6, VERO10, MDBK5 and MDCK5 cell lines; respectively. Growth kinetics of EHV-1 in VERO cells, showed initial increase in cell associated viruses (CAV) and cell free viruses (CFV) were at 8 and 48 hours post inoculation then reaching their peak at 72 and 96 hours post inoculation in CAV and CFV; respectively. Detection of viral protein in infected cells was done using IFT. It revealed that high fluorescence was shown at 4 till 72 hours post inoculation in CAV and 72 hours in CFV. In conclusion, EHV-1 is more cell associated and IFT is considered as a rapid and sensitive test for detection of viral protein

This study was conducted to evaluatethe degree of biosecurity level with especial reference to Salmonella Spp as an example to explain the expected causes and risk factors that leads to spread them in poultry flocks in Egypt then studied farms evaluated for the sensitivity of Salmonella isolates to the most common disinfectants used in Egypt. About 300 samples (100 cloacal swabs, 100 liver and intestinal samples, 100 litter samples) were collected from 10 broiler farms with different age (at 0 old day, one week, 2,4 and 6 weeks of age) then the samples were investigated for Salmonella Sppand subsequently identified based on biochemical and serological tests.The obtained results showed that 35 Salmonella species were isolated from 10 broiler poultry houses (25%);(6%) and (4%) from cloacal swab;liver and litter, respectively. Average prevalence of Salmonella spp. was 11.33 % in open broiler houses whether raised Cobb, Ross or Sasso breeds. Salmonella Typhimurium, S. enteritidis and S.Kentucky were the most serovars out of the 35 detected isolates. There was great statistical significant difference in the sensitivity of Salmonella isolates to the most common disinfectants(P<0.05)asVerkon- S® achieved 3 log reduction, after 5-minute,Formalin and Phenique were achieved 3 log reductions against S. entriditisafter one-minute Aldekol Des- Gda® achieved one log reduction after one minute, TH4®,Biosentry® 904 and Iodophoreachieved 2 log reductions after 5 minutes.

Fifty five milk samples were collected from dairy Friesian cows suffering from clinical mastitis in a farm at Quena Governorate. The samples were subjected to microbiological examination .The main isolated pathogenic bacteria were identified biochemically as Escherichia coli (36.3%), Staphylococcus aureus (23.6%) Streptococcus agalactiae (18.2%), Klebsiella pneumoniae (12.7%) and bacterial free samples (9.1%) . Sensitivity tests for the bacterial isolates revealed that the tested strains were sensitive to ciprofloxacin, nortril, gentamicin and lincospectin

50 specimens of the Mullets Mugil cephalus were collected from the Libian coastal waters near Misurata city in Libya. 7 different species of trematodes isolated from the intestine of the mullets were briefly described. Haploporus benedenii, Haploporus lateralis, Lecithobotryes putrescens, Saccocoelium tensum, Saccocoelium gohari, Haplosplanchnus caudatus and Haplosplanchnus pachysomus. The aim of the present work was to describe the way of creating chosen interactive and animated three-dimensional anatomic virtual models of particular parts of birds' skeleton. These models can be used for teaching purposes in anatomy courses to assist veterinary students, and veterinarians to know the different structures of birds’ bones without dissection. We used available multimedia technologies to develop a simple computer program for visualization and annotation of threedimensional duck skeleton. The entire program involves three-dimensional realistic objects, description texts, and images, in addition to quizzes for users' selfassessment. Addressing the anatomical structures made the program more useful to promote knowledge, enhance and facilitate teaching and learning of birds’ osteology for veterinary students.

In this study, neutralizing antibody response in Guinea pigs and mice inoculated by African horse sickness (AHS) bivalent inactivated oil adjuvant vaccine was similar when compared serologically with that in horse. Therefore, using G. pigs and mice may be a method of great value in potency test for routine immunization. Potency test of the prepared vaccine using an immunization challenge protocol in adult mice was studied, using challenge virus with 100, 50 and 25 LD50. Toxocariasis is an important cosmopolitan zoonotic disease mainlycaused by Toxocara spp., a type of soil transmitted helminth (STH) on cats and dogs. In this study, 80 soil samples were taken from four public playgrounds and six neighbourhood playgrounds in Ipoh, Perak between October and December 2016 to determine the status of soil contamination with the eggs of Toxocara spp. Results showed that 32.5% from the total soil samples were positive with Toxocara spp. eggs. Overall, five out of ten of the sampling sites were contaminated with Toxocara spp. eggs. Besides that, the relationship between the soil condition and the occurrence of the Toxocara spp. eggs in soils were also investigated. The findings showed that increase of moisture and pH of the soils contributed to the increase of contaminationwith Toxocara spp. eggs. Sandy soils were found significantly contaminated with the eggs of Toxocara spp. compared to theother types of soil. Therefore, appropriate preventive measures such as treatment of soil, regular monitoring and deworming of dogs and cats as well as awareness programmes to the public are important.

Tendon in horse is the most important type of connective tissue which connects muscle to bone, constituting a vital component of the musculoskeletal system, by enabling movement. Tendons suffer from a wide range of disorders, which includes different types of mechanical injuries and degenerative diseases. The sample population was a deep digital flexure tendon (DDFT) at mid-metacarpal region of thirty adult horses examined using scanning electron microscopy (SEM). Sex distribution was not taken into account. Changes in its structural organisation due to aging following tendon degeneration was unknown. The complex structures of tendon and its distinctive characteristics have been well demonstrated by SEM techniques. The tendon structure under SEM showed a dense, regular connective tissue arrangement thatreflects the mechanical requirement of this tissue. It is defined by thick regular bundles of longitudinal collagen fibres arrangedin a zig-zag conformation. All tendons examined from these horses had no history of deep digital flexure tendinitis and were allmacroscopically normal. The tendon showed a hierarchical structure, with collagen molecules forming fibrils (50 nm to 500 nm), which in turn comprise a fascicle (50 μm to 300 μm), and fascicles were aligned along the longitudinal axis of the tendon.

Malaysia has experienced four waves of H5N1 outbreak but no humandeaths were recorded which is in 2004, 2006, 2007 and latest outbreak in 2017 at Kelantan. The objective of this paper is tocollect, analyse and summarise the data of HPAI cases from the outbreak from 1 March to 22 June 2017. A total of 1,634 cases, 8,544 samples were received at the Registration Unit and Virology Section, Regional Veterinary Laboratory in Kota Bharu (RVLKB) for diagnosis and surveillance of H5N1 during this period. The samples received were pooled organs and intestines from post-mortem (41 cases) and cloaca swabs in tryptose phosphate buffer from surveillance (1,593 cases), which were sent to RVL, Kota Bharu and Veterinary Research Institute, Ipoh, Perak (VRI). They were processed and diagnosed using quantitative real-time reverse transcriptase PCR (RT qPCR) technique to detect and subtyping of the virus. Total positive cases for H5N1 were 53 out of 1,634 with 18 diagnostic cases and 35 surveillance cases. All positive cases during outbreak were detected from 43 locations from six districts of Kota Bharu, Bachok, Pasir Puteh, Pasir Mas, Tumpat and Tanah Merah, with 19 locations in Kota Bharu which is thehighest affected by H5N1, seven locations in Bachok, four in Pasir Putih, six in PasirMas, six in Tumpat and one in Tanah Merah. Many factors could have led to the acute spread of the virus between the districts like chicken smuggling, legal and illegal poultry trade, migration of infected wild birds and others. Understanding the source of outbreak and how it spread is important to control, eradicate and prevent the spread of the disease as it is zoonotic and infects human.

Stray dogs are carriers of several zoonotic diseases such as leishmaniasis and canine monocytic ehrlichiosis (CME) as a result of poor nutrition, low hygienic conditions and lack of veterinary care. Thus, the Veterinary Research Institute (VRI) conducted a survey to determine the parasite pathogens such as blood protozoans, gastrointestinal parasites and ectoparasites in stray dogs with the collaboration of the Kuala Lumpur City Council Pest Control Unit. Skin, organ, faecal and blood samples were analysed and results indicate that Babesia canis, Babesia gibsoni, Ehrlichia canis, Hepatozoon canis and microfilaria of Dirofilaria immittis are the common parasites species found in the blood and organ samples in 2014. The faecal floatation technique showed the presence ofhelminth ova such as Trichuris, Ancylostoma and Toxocara species. All skin samples were positive for Rhipicephalus sanguineus ticks. As strays are closely linked to human habitats such as market and housing areas, it is vital that stray population control is strategically implemented to safeguard these common zoonotic infections from spreading to humans.
