Purpose. To review publications relating to the key point of the genotyping technology that is competitive allele-specific polymerase chain reaction (which is called now Kompetitive Allele Specific PCR, KASPTM) and its use in various genetic-breeding researching (a study of maize). Results. The essence of KASP-genotyping, its advantages are highlighted. The requirements for matrix DNA are presented, since the success of the KASP-analysis depends on its qua­lity and quantity. Examples of global projects of plant breeding for increasing crop yields using the KASP genoty­ping technology are given. The results of KASP genotyping and their introduction into breeding and seed production, in particular, for determining genetic identity, genetic purity, origin check, marker-assisted selection, etc. are presented using maize as an example. It is demonstrated how geno­mic selection according to KASP genotyping technology can lead to rapid genetic enhancement of drought resistance in maize. Comparison of the effectiveness of creating lines with certain traits (for example, combination of high grain yield and drought resistance) using traditional breeding approaches (phenotype selection) and molecular genetic methods (selection by markers) was proved that it takes four seasons (two years in case of greenhouses) in order to unlock the potential of the plant genotype using traditional self-pollination, test-crossing and definitions), while using markers, the population was enriched with target alleles during one season. At the same time, there was no need for a stress factor. Conclusions. KASP genotyping technology is a high-precision and effective tool for modern genetics and breeding, which is successfully used to study genetic diversity, genetic relationship, population structure, gene­tic identity, genetic purity, origin check, quantitative locus mapping, allele mapping, marker-assisted selection, marker-assisted breeding. It is expedient and timely to introduce KASP genotyping technology in our country to solve a wide range of modern genetics, breeding, seed production tasks.

To establish the reasons of submitting not highly decorative lilac hybrids to state variety testing so that unpromising varieties and look-alikes can be sometimes included in the world collection, and focus on solving this situation. Methods. Analytical approach. Results. It was established why and how unoriginal hybrids were submitted to state variety testing that for several reasons can allow them to obtain the status of variety. Conclusions. In order to prevent obtaining the status of variety by some low decorative lilac hybrids, it is necessary that both a breeder and testing stations employer should use known presentable collections of identified varieties for evaluation of real value of a new hybrid. In Ukraine, there is such a collection at N. N. Gryshko National Botanical Garden of the National Academy of Sciences of Ukraine where more than 130 varieties and 21 species of the genus Syringa occupies the area of 2.35 ha. It is desirable to compare not only with the check variety that the breeder selected for his hybrid among such, but, if it is possible, with all varieties in the collection close to it by decorative effect. The problem is that the author of a hybrid, sometimes ignorantly or for some other reason, selects a check variety for his hybrid that is not the most decorative one among such, against which a candidate for variety status will be more attractive during its evaluation. In such a case, the breeder should change a check variety that permits to estimate really the decorative effect of a submitted hybrid.

Interspecific hybridization, laboratory test, analytical approach. Results. Based on the interspecific hybridization, the initial breeding material was created and the degree of its resistance to the above pathogens was determined by way of artificial infection of tubers with the inoculum of such fungi as Fusarium sambucinum Fuck and Phytophthora infestans (Mont.) De Bary. During interspecific hybridization based on schemes of saturating and enriching crosses, using forms of various species with a high phenotypic expression of resistance to Fusarium dry rot, the result of the cumulative effect of genes that control resistance to the pathogen was observed. Crossing combinations differed significantly for the degree of population average manifestation of resistance to the diseases. Conclusions. Combinations В54, В53, В61 with a mean resistance (above 7 grades) to Fusarium dry rot have been selected. Such combinations as B52, B50 and B54 had increased resistance to tuber late blight. It was found that the combination В54 is characterized by complex resistance to both diseases. For further work, the following samples with complex resistance to Fusarium dry rot and tuber late blight (7 grades or more) were selected: В59с42, В59с43, В50с16, В50с19, В50с44, В51с1, В51с26, В51с28, В52с11, В52с23, В52с24, В52с29, В53с1, В53с11, В53с17 , В53с23, В54с13, В54с14.

Purpose. To develop a method of propagation, stimulation of rhizomes growth in vitro culture for the genus Miscanthus representatives and their adaptation in the open field without the use of greenhouse complexes for acclimatization and completion of growing. Methods. Biotechnological procedures, mathematical and statistical analyses. Results. Prescription of nutrient medium was developed for explants inoculation, sprouts propagation, rhizomes growth stimulation in vitro. Such sterile explants as seeds, buds to be removed from rhizomes, parts of  stems with bud were placed on modified media with mineral portion by Murashige and Skoog (MS) that contained 0,5–1 dose of macroelements and one dose of microelements,  vitamins (10 mg/l of thia­minum, 1,0 mg/l of pyridoxine, 1,0 mg/l of nicotinic acid and 1,0 mg/l of ascorbic acid) supplemented with amino acids (250 mg/l of glutamic acid, 3 mg/l of tyrosine, 3 mg/l of arginine, 2 mg/l of hydroxyproline), plant growth regulators [0,5–1,0 mg/l of GA (gibberelline acid), 0,2 mg/l of 6-BAP (6-Benzylaminopurine, 0,1 mg/l of NAA (α-naphtylacetic acid)] in different variations. After seed germination, buds emerging and sprouts formation 1–2 cm in height, for propagation purpose they were passivated on the medium of other composition that differed from previous one by the content and ratio of growth regulators, especially by a high concentration of cytokinins [6-BAP (0,4–0,5 mg/l), kinetin (0,5 mg/l), adenine (0,5 mg/k)] in different variations in presence of GA (0,2 mg/l). In order to stimulate rhizomes growth, microclones were transferred on media with other composition and ratio growth regulators (6-BAP (0,2–0,3 mg/l) + GA (0,5–1,0 mg/l) or 6-BAP (0,2–0,3 mg/l) + GA (0,5–1,0 mg/l) + NAA (0,1 mg/l),  in other words,  with a high content of gibberellins. After the formation of rhizomes 10–15 cm in length, miscanthus plants were planted out in the open ground. Stimulation of rhizomes initiation and elongation on appropriate nutrient media before Miscanthus giganteus, M. sacchariflorus and M. sinensis planting in vivo resulted in 100% adaptation and 100% survival of plants in the winter period without the use of greenhouse complexes. Conclusions. The method of miscanthus propagation in vit­ro and adaptation in the open ground was developed that included stimulation of rhizomes growth and favoured the increase of their length on media supplemented with gibbe­relline that guaranteed 100% preservation of microplants to be propagated from in vitro culture during adaptation in the open ground and acclimatization in winter.

Виділення ДНК з проростків на колонках із сорбуючою мембраною; полімеразна ланцюгова реакція; горизонтальний гель-електрофорез; визначення розмірів продуктів ампліфікації за програмою «GelAnalyzer 2010a». Результати. За допомогою аналізу мікросателітного локусу Satt100, який є маркером гена Е7, проведено типування 15 сортів сої. Встановлено наявність продуктів ампліфікації розміром 133, 149 та 168 п.н., що маркують рецесивний та домінантний алелі гена Е7 відповідно. Усі досліджені сорти, крім сорту ‘Одеська 150А’, були гомогенними за мікросателітним локусом Satt100. Генотипи восьми сортів мали рецесивний алель е7, шести – домінантний алель Е7. Порівняли наявність певного алеля гена Е7 у генотипі сорту з належністю до групи стиглості. Відмічено зв’язок для сортів з вегетаційним періодом до 100 діб: усі чотири сорти цієї групи мали алель е7 та для сортів із вегетаційним періодом понад 110 діб: для всіх чотирьох сортів цієї групи характерні фрагменти ампліфікації локусу Satt100, що маркують домінантний алель. Із семи середньо­ранніх сортів чотири мали рецесивний алель е7, а три – домінантний Е7. Наведено інформацію про отримання різними науковцями фрагментів ампліфікації неоднакових розмірів, але при цьому виявлено тенденцію щодо зв’язку більш високомолекулярних фрагментів з домінантним, а низькомолекулярних – з рецесивним алелем. Висновки. За результатами типування за мікросателітним маркером Satt100 гена Е7 фотоперіодичної чутливості для 15 сортів сої визначено наявність рецесивного або домінантного алеля гена Е7. Typingsoybean (Glycinemax (L.) Merr.) culti­vars of Ukrainian and foreign breeding for microsatellite mar­ker Satt100 of E7 gene of photoperiod sensitivity. Methods.DNA recover from seedlings on columns with sorbing membrane; polymerase chain reaction (PCR); horizontal gel electro­phoresis; amplification products size determination using “GelAnalyzer 2010a” program. Results. PCR analysis of the microsatellite locus Satt100 to be the marker of the E7 gene was used for typing 15 soybean cultivars. Availability of the 133, 149 and 168 bp amplification products has been established that marked recessive and dominant alleles of Е7 gene correspondingly. All varieties, except ‘Odeska 150A’, were homogeneous for the microsatellite locus Satt100. Genotypes of eight varieties has the recessive allele e7, six ones – the dominant allele E7. Availability of the certain allele of the E7 gene was compared in variety genotype with belonging to the maturity group. The association was found for varieties with vegetation period up to 100 days: all four varieties of this group had the e7 allele, for varieties with vegetation period more than 110 days: for all four varieties of this group the Satt100 locus amplification fragments that mark the dominant allele are specific. Amongsevenmiddle-agedvarieties, fourof them hadtherecessiveallelee7, threeofthem – dominantalleleЕ7. According to thepresented information, some scientistsobtainedamplificationfragments of different size, butthetendency was revealed concerning the association of the higher molecular fragments witht hedominant gene, and the lowmolecular fragments with therecessive gene. Conclusions. Based on the results of typing of 15 soybean varieties for the microsatellite marker Satt100 of E7 gene of photoperiod sensitivity, it was determined availability of recessive or dominant allele of E7 gene. For most samples, the concordance of availability of the certain allele of E7 gene and the duration of the vegetation period was established. Availability of amplification fragments of different size to be specific for recessive allele testified the needfor sequencing and bioinformatic analysis of the Satt100 locus amplification fragments. | Цель. Осуществить типирование сортов сои культурной (Glycine max (L.) Merr.) украинской и зарубежной селекции по микросателлитному маркеру Satt100 гена Е7 фотопериодической чувствительности. Методы. Выделение ДНК из ростков на колонках с сорбирующей мемб­раной; полимеразная цепная реакция; горизонтальный гель-электрофорез; определение размеров продуктов амплификации с использованием программы «GelAnalyzer 2010a». Результаты. С помощью анализа микросателлитного локуса Satt100, являющегося маркером гена Е7, проведено типирование 15 сортов сои. Установлено наличие продуктов амплификации размером 133, 149 и 168 п.н., которые маркируют рецессивный и доминантный аллели гена Е7 соответственно. Все исследуемые сорта, кроме сор­та ‘Одесская 150А’, были гомогенными по микросателлитному локусу Satt100. Генотипы восьми исследованных сор­тов имели рецессивный аллель е7, шести – доминантный аллель Е7. Сравнили наличие определенного аллеля гена Е7 в генотипе сорта с принадлежностью к группе спелости. Отмечено связь для сортов с вегетационным периодом до 100 суток: все четыре сорта этой группы имели аллель е7, и для сортов с вегетационным периодом более 110 суток: для всех четырех сортов этой группы характерны фрагменты амплификации локуса Satt100, которые маркируют доминантный аллель. Среди семи среднеранних сортов четыре имели рецессивный аллель е7, а три – доминантный Е7. Представлено информацию о получении разными учеными фрагментов амплификации неодинаковых размеров, но при этом выявлено тенденцию связи более высокомолекулярного фрагмента с доминантным геном, а низкомолекулярных – с рецессивным геном. Выводы. По результатам типирования по микросателлитному маркеру Satt100 гена Е7 фотопериодической чувствительности для 15 сортов сои определено наличие рецессивного или доминантного аллеля гена Е7. Для большинства образцов установлено соответствие наличия определенного аллеля гена Е7 и продолжительности вегетационного периода. Наличие фрагментов амплификации разного размера, характерных для рецессивного аллеля, свидетельствует о необходимости секвенирования и биоинформатичного анализа фрагментов амплификации локуса Satt100.

Мета. Проаналізувати сортовий потенціал селекції, оптимізацію сортової структури виробництва та періодичність сортових змін тютюну Nicotiana tabacum L. у Республіці Болгарія. Результати. Ретроспективний аналіз селекції тютюну (Nicotiana tabacum L.) свідчить, що за період 1950–2010 рр. у Болгарії створено 158 сортів, серед яких 130 сортів з підвиду Східний (ssp. Orientalis), 28 сортів підвиду Американський (ssp. Americana), з яких 17 сортів типу Вірджинія і 11 сортів типу Берлі. На основі цього сортового потенціалу проведено відповідні сортозміни у виробничому секторі країни. За 50–60 років (1950–2010 рр.) здійснено 5–7 сортозмін вітчизняними сортами. Висока ефективність селекції тютюну з отриманням якісних сортів і порівняно часті сортозміни створюють сприятливі можливості для щорічного оптимального формування сортової структури виробництва залежно від вимог і попиту відповідного сировинного сегменту ринку та кон’юнктури ринку, що швидко змінюється. Висновки. Із загального сортового потенціалу селекції сортів тютюну підвиду Східний (130 сортів) за 13-річний період (1995, 2000–2011 рр.) увійшли до виробничого обороту 47,7% (62 сорти), приблизно 25% (24–30 сортів) формують основний обсяг Східного тютюну, який щорічно виробляють у Болгарії. Сортовий потенціал селекції великолистового тютюну у виробництві використано повністю – 17 сортів типу Вірджинія та 11 сортів типу Берлі. Тютюн є єдиною сільськогосподарською культурою в Болгарії, яка включає в структуру виробництва майже 100% сортів вітчизняної селекції.

Aspergillus flavus and its main secondary metabolite AFB1 pose a serious threat to several important crops worldwide. Recently, it has been reported that some PHD family transcription factors are involved in the morphogenesis and AFB1 biological synthesis in A. flavus, but the role of Cti6, a PHD domain containing protein in A. flavus, is totally unknown. The study was designed to reveal the biological function of Cti6 in the fungus by deletion of cti6, and its two domains (PHD and Atrophin-1) through homologous recombination, respectively. The results showed that Cti6 might up-regulate the mycelium growth, conidiation, sclerotia formation and AFB1 biological synthesis of A. flavus by its PHD domain, while Atrophin-1 also improved the conidiation of the fungus. The qRT-PCR analysis showed that Cti6 increased the conidiation of the fungus through AbaA and BrlA mediated conidiation pathway, triggered the formation of sclerotia by orthodox sclerotia formation pathway, and improved the production of AFB1 by orthodox AFB1 synthesis pathway. Crops models analysis showed that A. flavus Cti6 plays vital role in colonization and the production of AFB1 on the host grains mainly via PHD domain. Bioinformatics analysis showed Cti6 is conservative in Aspergillus spp., and mCherry mediated subcellular localization showed that most Cti6 accumulated in the nuclei, which reflected that Cti6 performed its important biological function in the nuclei in Aspergillus spp.. The results of the current study elucidate the roles of PHD domain containing proteins in the mechanism of the infection of crops by A. flavus, and provided a novel target for effectively controlling the contamination of Aspergillus spp. to crops.

Purpose. To reveal breeding and genetic peculiarities of modern spring barley varieties for the “number of grains per main ear” trait and identify genetic sources of increased combining ability for involving in hybridization. Methods. Investigations were carried out at the V. M. Remeslo Myronivka Institute of Wheat of NAAS of Ukraine. Modern varieties of domestic (‘Virazh’, ‘Talisman Myronivskyi’, ‘Komandor’) and foreign (‘KWS Aliciana’, ‘KWS Bambina’, ‘Zhana’, ‘Explorer’) breeding were involved in crossing for a full (7´7) diallel scheme. Parents and F1 were studied in field conditions during 2014–2016. Results. The analysis of variance of combining ability has shown a significant advantage in varying of general combining ability (GCA) effects. The mean square of specific combining ability (SCA) was significantly less than the GCA, but reliable throughout the years. The reciprocal effect was reliable only in 2014. Stably high effects of GCA during all years of investigations were noted in the varieties ‘KWS Aliciana’ (1.18–1.62) and ‘Virazh’ (1.33–1.48). The variety ‘KWS Bambina’ was characterized by lower but reliable positive effects of GCA (0.43–0.99) as compared to mentioned above. Non-allelic gene interaction was not found, that allowed to calculate the basic parameters of genetic variation. During all years of investigations, dominant effects of genes (H1 and H2) prevailed over the additive (D) ones in phenotypic expression of grain number per main ear. Mean degree of dominance in the experiment (H1/D) has shown overdominance. The same pattern was also distinctive for the index of mean degree of dominance in the loci . The dominance was reliably directed. Dominant effects of genes increased grain content, and recessive ones reduced it. At least 3–4 genes (groups of genes) have been revealed which determined the effects of dominance. At the same time, recessive genes (F<0) or gene effects were prevailed quantitatively in the varieties investigated. A high coefficient of heritability in broad sense (H2 = 0.98) has shown a significant determination of phenotypic variability with genetic factors. The coefficient of heritability in narrow sense (h2 = 0.66–0.68) confirmed that despite the advantage of dominant effects over the additive ones, the contribution of the latter was also significant. Conclusions. The prevalence of dominant effects of genes in the phenotypic expression of the number of grains per main ear causes the need for sufficient sample size of hybrid material and points to the expediency of conducting a more “rigid” selection for phenotype in later generations. At the same time, the considerable contribution of additive effects and high values of heritability indices give reason to predict the efficiency of selections aimed at increasing the trait in created hybrid material. The varieties ‘Virazh’, ‘KWS Aliciana’, ‘KWS Bambina’ should be used as effective genetic sources to increase grain content in combination breeding.

The studied samples were grown in the nurseries of field selection crop rotation. Accordingly, the initial material with the highest drought resistance value has been defined. Among the evaluated material, eight hybrids of the competitive and ecological test have been selected (drought resistance coefficient was ranging from 59.4% to 84.8%) and five biotechnological lines of ‘Hlazurna’ and ‘Dorohin’ varieties (drought resistance coefficient was in the range of 55.5% to 67.5%). As for wild species, almost half of the samples (47.8%) were characterized by a high coefficient of drought resistance (from 55 to 78%). Selected samples with high values of drought resistance were recommended to use as a source and drought resistance donors when creating new potato varieties. Conclusions. The initial potato material (hybrids, varieties, biotechnological lines and wild species) with high values of drought resistance (55.0–84.8%) has been selected. These samples are recommended to use in the breeding process when creating new drought resistance potato varieties.

Оцінити та провести добір гібридів, сортів батьківського розсадника селекційного процесу, біотехнологічних ліній та диких видів картоплі за фізіологічними показниками посухостійкості.Методи. Фізіолого-біохімічні, селекційні, статистичні.Результати. Наведено дані оцінювання водоутримувальної та водовідновлювальної здатності листків картоплі перспективних гібридів конкурсно-екологічного випробування, сортів батьківського розсадника, біотехнологічних ліній і диких видів та їх інтегрального показника посухостійкості. Досліджувані зразки вирощували в розсадниках польової селекційної сівозміни. Відповідно визначено вихідний матеріал з найвищим показником посухостійкості. З-поміж оцінюваного матеріалу виділено: вісім гібридів конкурсно-екологічного випробування – коефіцієнт посухостійкості коливався від 59,4 до 84,8%, п’ять біотехнологічних ліній сортів ‘Глазурна’ та ‘Дорогинь’ – коефіцієнт посухостійкості у межах від 55,5 до 67,5%. Серед диких видів майже половина зразків (47,8%) мала високий коефіцієнт посухостійкості – від 55 до 78%. Зразки з високими показниками посухостійкості рекомендовано використовувати як джерело та донори стійкості до посухи під час створення нових посухостійких сортів картоплі.Висновки. Виділено вихідний матеріал картоплі (гібриди, сорти, біотехнологічні лінії та дикі види) з високим коефіцієнтом посухостійкості (55,0–84,8%). Рекомендується використовувати ці зразки у селекційному процесі під час створення посухостійких сортів картоплі. | Цель. Оценить и провести отбор гибридов, сортов родительского питомника селекционного процесса, биотехнологических линий и диких видов картофеля по физиологичес­ким показателям засухоустойчивости. Методы. Физиолого-биохимические, селекционные, статистические. Результаты. Приведены данные оценивания водоудерживающей и водообновляющей способности листьев картофеля перспективных гибридов конкурсно-экологического испытания, сортов родительского питомника, биотехнологических линий, диких видов и их интегрального показателя засухоустойчивости. Исследуемые образцы выращивали в питомниках полевого селекционного севооборота. Соответственно определен исходный материал с наивысшим показателем засухоустойчивости. Среди оцениваемого материала выделено: восемь гибридов конкурсно-экологического испытания – коэффициент засухоустойчивости колебался от 59,4 до 84,8%, пять биотехнологических линий сортов ‘Глазурна’ и ‘Дорогинь’ с коэффициентом засухоустойчивости в пределах от 55,5 до 67,5%. Среди диких видов почти половина образцов (47,8%) имели высокий коэффициент засухоустойчивости – от 55 до 78%. Образцы с высокими показателями засухоустойчивости рекомендовано использовать как источники и доноры устойчивости к засухе при создании новых засухоустойчивых сортов картофеля. Выводы. Выделен исходный материал картофеля (гибриды, сорта, биотехнологические линии и дикие виды) с высоким коэффициентом засухоустойчивости (55,0–84,8%). Рекомендовано использовать эти образцы в селекционном процессе при создании засухоустойчивых сор­тов картофеля.
