Historical review was done concerning the formation and development of scientific research of the Experimental Station of Medicinal Plants over the period 1916–2016. The main stages of experimenting in the direction of “medicinal plant cultivation’’ in Ukraine were covered. Role and importance of the Experimental Station of Medicinal Plants in the scientific and methodological support of such scientific directions as breeding, seed production, cultivation technologies, pharmacognosy in medicinal plant cultivation were shown. For the needs of the branch, over 100 species of introduced medicinal plants were studied at the Station and agricultural techniques of their growing were developed, in addition, more than 50 varieties of medicinal plants were created, a number of machines and devices for growing, harvesting and initial treatment of raw materials and seeds of the most important medicinal plants was designed. Other scientific achievements of the Station were mentioned and promising areas of medicinal plant research in Ukraine were outlined.

High-throughput sequencing (HTS) enables the generation of large amounts of genome sequence data at a reasonable cost. Organisms in mixed microbial communities can now be sequenced and identified in a culture-independent way, usually using amplicon sequencing of a DNA barcode. Bulk RNA-seq (metatranscriptomics) has several advantages over DNA-based amplicon sequencing: it is less susceptible to amplification biases, it captures only living organisms, and it enables a larger set of genes to be used for taxonomic identification. Using a model mock community comprising 17 fungal isolates, we evaluated whether metatranscriptomics can accurately identify fungal species and subspecies in mixed communities. Overall, 72.9% of the RNA transcripts were classified, from which the vast majority (99.5%) were correctly identified at the species level. Of the 15 species sequenced, 13 were retrieved and identified correctly. We also detected strain-level variation within the Cryptococcus species complexes: 99.3% of transcripts assigned to Cryptococcus were classified as one of the four strains used in the mock community. Laboratory contaminants and/or misclassifications were diverse, but represented only 0.44% of the transcripts. Hence, these results show that it is possible to obtain accurate species- and strain-level fungal identification from metatranscriptome data as long as taxa identified at low abundance are discarded to avoid false-positives derived from contamination or misclassifications. This study highlights both the advantages and current challenges in the application of metatranscriptomics in clinical mycology and ecological studies.

Purpose. To analyze peculiarities of Ukrainian cereals and oilseeds trade, the situation on the world market, and determine future prospects of its development. Methods. Analysis and synthesis, comparative evaluation, graphic procedure. Results. The role and place of Ukraine in a total grain supply to the world market was determined. Ukraine is a world’s top ten grain producer. Among domestic agricultural products, cereals, oilseeds and sunflower oil are in the highest demand in the world. In recent years, our state has reinforced its status as one of the leading exporters of cereals. The commodity pattern of cereals and oilseeds export was analyzed with specifying most in-demand positions and the main countries purchasing these products. According to the results of 2015, Ukraine obtained the highest foreign currency revenue from export of corn, wheat and barley (in grain structure) including soybeans and rapeseed (among oil crops). Key domestic and multinational operators are the main exporters of cereals and oilseeds in Ukraine and still hold their leading position. It was found a significant excess of import price of seeds as compared with export price of crops grown in Ukraine. Assortment of maize and sunflower seeds offered by major companies-producers in Ukraine was studied. Main trends of the world grain market development are considered. Conclusions. It was established that Ukraine is one of the major exporters of cereals and oilseeds. However, volatility of their prices significantly affects the export revenue that was decreasing even with increasing export quantities in kind. The dependence of domestic grain industry development on high-quality imported seed of maize and sunflower hybrids was recorded. It is expected that in the years to come Ukraine will maintain its strong positions in the world’s grain market.

Для оцінки стійкості до фузаріозу колоса в 2009 році 41 сорт твердої та м’якої пшениці, переважно з Італії, пройшов сортовипробування у CIMMYT (Міжнародний центр поліпшення кукурудзи та пшениці). Крім того, виконано оцінку впливу одного з основних QTL стійкості до фузаріозу колоса (Qfhs.ndsu-3BS QTL), вперше виявленого у китайського сорту пшениці м’якої ‘Sumai 3’, на хромосомі 3В, у 125 удосконалених ліній пшениці твердої BC4F6, отриманих шляхом схрещування з вихідним сортом пшениці м’якої ‘Sumai 3’ (68 ліній з ‘Sumai 3’ QTL та 57 ліній без цього QTL), були досліджені в однакових умовах штучного зараження. Для обох груп ділянки заражували під час цвітіння суспензією односпорових культур F. graminearum, підтримуючи вологість до 100%, щоб сприяти розвитку захворювання за допомогою системи дрібнодисперсного зволоження. Через тридцять днів після зараження підрахували кількість колосків, інфікованих F. graminearum, на колосі десяти рослин на кожній ділянці; пошкодження виразили показником зараження фузаріозом (кількість випадків ураження ступінь ураження / 100, де ступінь ураження = кількість інфікованих колосків / загальна кількість випадків ураження 100 та кількість інфікованого колосся / загальна кількість колосся 100). В обох випадках пізнє цвітіння було ключовим чинником, здатним обмежити ураженість хворобою. Попередні дані стосовно впливу Qfhs.ndsu-3BS QTL не виявили відмінності між двома групами вдосконалених ліній.

Purpose. To establish the level of formation of winter barley yield depending on weather conditions during the growing season in the Forest-Steppe zone of Ukraine. Methods. Field investigations, ANOVA, variance and correlation analysis. Results. Positive correlation was revealed between yield and the duration of the vegetation (r = 0.54) and especially “heading–maturation” period (r = 0.76). Moderate positive correlation was established between yield and mean daily air temperature during the period from termination to resumption of vegetation (r = 0.44), and strong negative one – between yield and mean daily air temperature during heading to maturation period (r = -0.77). Positive correlation was observed between yield and rainfall amount during the period from vegetation resumption to heading (r = 0.30). In general, during the growing season the correlation was weak (r = 0.16). For the period of “heading–maturation” low negative value (r = -0.11) was fixed. This was due to heavy precipitations, especially torrential rains, which in some years have caused lodging of plantings resulted in yield decrease. Conclusions. Under the conditions of Forest-Steppe zone of Ukraine, fluctuations in temperature and water regimes are responsible for significant variability in growing season duration and yield of winter barley. Optimal amount of precipitation (close to long-time annual average) and their uniform distribution throughout growing season is the most favorable for obtaining high yield. “Heading–maturation» period is responsible for the formation of winter barley yield level.

Purpose. To study flowering productivity and reproductive ability of multiple-aged plants of introduced Astilba varieties, assess varieties for these characters and investigate the effectiveness of vegetative propagation. Methods. Field experiment, introduction and statistics evaluation. Results. The analysis of published data and author’s own observations revealed that Astilba varieties are highly tolerant for winter and frost conditions as well as pest and disease resistant, therefore we defined flowering productivity and reproductive ability as main economic and biological characters that are subject to assessment. According to the results of flowering efficiency study, it was found that the highest value of this index for the most studied varieties was observed on the fourth year of cultivation. Whereas the majority of varieties rating was relatively low, Astilbe flowering productivity on the whole was sufficiently high. In order to arrange collection plots, flower beds and implement the selection process, the most effective method of vegetative propagation can be used – the division of rhizomes into pieces with three to five renovation buds, and for mass reproduction – with one or two buds. It turned out that for Astilbe the indicator of reproductive ability is specific. For most of the investigated five-year varieties it was quite high. We have noted a high positive correlation between the index of reproductive ability and flowering productivity. Varieties with optimal flowering had high value of planting material yield. Conclusions. Based on the experimental data, the flowering productivity and reproductive ability of multiple- aged plants of Astilbe introduced varieties was studied, varieties was assessed according to these characters, and the effectiveness of vegetative propagation was investigated.

Dialectical, field, laboratory and statistical ones. Results. Medium-grown (‘Amos’, ‘Nikanor’, ‘Rarytet’, ‘Yaroslava’) and low-stem (‘HAD 69’, ‘HAD 86’, ‘HAD 110’, ‘Timofei’) multiline varieties of winter and alternate hexaploid triticale were developed with higher adaptability, potential yield of 9–12 tons per ha and high bread-making properties. Among the most drought resistant genotypes, such varieties as ‘Amos’, ‘Buket’, ‘Harne’, ‘Markiian’, ‘Kharroza’, ‘Shalanda’, ‘Nicanor’ and ‘Yaroslava’ showed high values of yield, plasticity and stability. Conclusions. The use of interspecific hybridization instead of intergeneric one in hexaploid triticale breeding, together with systemic testing of the hybrid material in contrasting agro-ecological zones, ensured the creation of multiline competitive varieties with an optimal combination of yield and adaptive properties

Purpose. Providing information on the formation, composition, maintenance of the collection of Buckwheat (Fagopyrum Mill.) genus at Ustymivka Experimental Station of Plant Production (Poltava region), and areas of its use: introduction, preservation and study of the material, for the set of index, formation of special collections and providing research and education institutions of Ukraine with a valuable original material. Methods. Methodical developments dealing with formation and maintenance of crop collections, methodical literature on the study of buckwheat collection for breeding and agronomic performance. Results. The author represented results of the collection material study for the main breeding and economically valuable traits including yield, productivity, resistance to abiotic and biotic factors of the environment. The results of work are considered aimed on creating special collections of the gene pool – base, trait (for yield and large seeds; for productivity, drought and heat resistance; for suitability for mechanical cultivation), educational and core ones. Conclusions. All formed and registered collections are a valuable base material for solving a number of breeding goals to increase the quantity and improve the quality of plant production.

Purpose. To select promising high productive maize hybrids of middle-early maturity group in terms of stability and plasticity of main economic characters. Methods. Field study, laboratory test, analytical procedure and statistical evaluation. Results. 14 maize hybrids recorded in the State Register of Plant Varieties Suitable for Dissemination in Ukraine in 2015 were studied for plasticity and stability of such traits as productivity, protein and starch content. Intensive highly-plastic hybrid ‘SI Tiptop’ was selected among the studied ones for productivity trait that can respond properly to changes of growing conditions. It was defined that for the starch content such hybrids as ‘SI Tiptop’, ‘SI Enigma’, ‘SI Arioso’, ‘Svich 38’, ‘Svich 35’, ‘HU 8653’, ‘Zdobutok’ and ‘SI Contrakt’ belonged to the intensive type and combined rather high values and the stability of the studied trait under variable conditions. The following hybrids as ‘NS 2642’, ‘DK S3016’, ‘Svich 38’, ‘NS 2632’ were qualified as intensive for protein content and appeared to be highly-plastic but stability values of this trait were low. ‘Svich 38’ hybrid was intensive simultaneously for two traits such as protein and starch content and showed rather high values of plasticity. ‘SI Tiptop’, ‘SI Enigma’ and ‘Svich 35’ were defined as hybrids of extensive type that provided stable protein content in adverse cultivation conditions. Conclusions. On the condition that intensive crop growing technologies should be used, for obtaining stable yields it is advisable to sow only highly-plastic hybrids that can adapt to unfavorable environmental factors, including ‘SI Tiptop’ – for productivity trait, ‘Zdobutok’ and ‘SI Kontrakt’ – for starch content, ‘MAC 24N‘, ‘NA 2642‘ and ‘Danubio’ – for protein content.

Purpose. To develop a conceptually new method for determination of varietal purity (typicality), hybridity, sterility of seed lots. Methods of molecular biology (genomic DNA extraction, PCR with SSR markers application, capillary electrophoresis), genetic, statistical, mathematical analysis. Results. New method for determining the varietal qualities of seed lot was developed that consists of the following steps: simultaneous DNA extraction from a representative sample of aggregated seeds; PCR and further analysis of the amplification products by determination of the qualitative and quantitative composition of SSR-markers’ alleles; calculation of values of varietal seed lot quality using experimentally derived allele ratios. Conclusions. The developed method for determining varietal qualities of seed lots allows to reduce significantly the consumption of materials, time and labor during the analysis. Consistent qualification and quantification of alleles in the total sample of a seed lot is a conceptually new approach to establish varietal purity (typicality), hybridity, sterility.

ß-tubulin, calmodulin, internal transcribed spacer and partial Isu-rDNA, RNA polymerase 2, DNA replication licensing factor Mcm7, and pre-rRNA processing protein Tsr1 were amplified and sequenced from numerous isolates belonging to Aspergillus sect, versicolor. The isolates were analyzed phylogenetically using the concordance model to establish species boundaries. Aspergillus austroafricanus, A. creber, A. cvjetkovicii, A. fructus, A. jensenii, A. puulaauensis, A. subversicolor, A. tennesseensis and A. venenatus are described as new species and A. amoenus, A. protuberus, A. sydowii, A. tabacinus and A. versicolor are accepted as distinct species on the basis of molecular and phenotypic differences. PCR primer pairs used to detect A. versicolor in sick building syndrome studies have a positive reaction for all of the newly described species except A. subversicolor.

Purpose. To investigate special aspects of accumulation of inorganic elements including heavy metals in seeds of white and black mustard to be grown for obtaining drugs. Methods. Field experiments, microwave digestion, ICP-MS and statistical analysis. Results. The content of inorganic elements including heavy metals was determined in the seeds of white and black mustard grown in Kiev Oblast. It was revealed that during the growing season plants of white mustard were able to accumulate such elements as aluminum, barium, strontium, zinc in seeds in concentrations that exceed their content in black mustard seeds, while compounds of calcium, cesium, iron, magnesium, manganese, potassium in a greater degree were accumulated in black mustard seeds. Conclusions. As legal and regulatory documents for important chemical elements don’t contain the maximum permissible limits of their content in medicinal plants, it would make sense to launch a comprehensive research with the involvement of specialists of relevant profiles in order to establish such a gradation. Plants of white and black mustard in Kiev Oblast have accumulated high levels of such metals as Ba, Cu, Fe, Mn, Mo, Sr, Zn that exceed the known limits of accumulation, indicating a partial contamination of soils in the region. Consequently, these plants can be used for phytoremediation of soils. Considering the fact that in the pharmaceutical practice refined mustard seed oil is used, revealed alterations of metal accumulation in seeds will not affect the quality of the final drugs. According to the research results, white and black mustard is promising for cultivation in Kiev Oblast with a view to obtain raw material that can be processed into drugs.

Purpose. To investigate special aspects of accumulation of inorganic elements including heavy metals in seeds of white and black mustard to be grown for obtaining drugs. Methods. Field experiments, microwave digestion, ICP-MS and statistical analysis. Results. The content of inorganic elements including heavy metals was determined in the seeds of white and black mustard grown in Kiev Oblast. It was revealed that during the growing season plants of white mustard were able to accumulate such elements as aluminum, barium, strontium, zinc in seeds in concentrations that exceed their content in black mustard seeds, while compounds of calcium, cesium, iron, magnesium, manganese, potassium in a greater degree were accumulated in black mustard seeds. Conclusions. As legal and regulatory documents for important chemical elements don’t contain the maximum permissible limits of their content in medicinal plants, it would make sense to launch a comprehensive research with the involvement of specialists of relevant profiles in order to establish such a gradation.
