Biodegradation of 4-nitroaniline by novel isolate Bacillus sp. strain AVPP64 in the presence of pesticides 全文

In this study, Bacillus sp. strain AVPP64 was isolated from diuron-contaminated soil. It showed 4-nitroaniline (4-NA) degradation, pesticide tolerance, and self-nutrient integration via nitrogen (N)-fixation and phosphate (P)-solubilization. The rate constant (k) and half-life period (t₁/₂) of 4-NA degradation in the aqueous medium inoculated with strain AVPP64 were observed to be 0.445 d⁻¹ and 1.55 d, respectively. Nevertheless, in the presence of chlorpyrifos, profenofos, atrazine and diuron pesticides, strain AVPP64 degraded 4-NA with t₁/₂ values of 2.55 d, 2.26 d, 2.31 d and 3.54 d, respectively. The strain AVPP64 fixed 140 μg mL⁻¹ of N and solubilized 103 μg mL⁻¹ of P during the presence of 4-NA. In addition, strain AVPP64 produced significant amounts of plant growth-promoting metabolites like indole 3-acetic acid, siderophores, exo-polysaccharides and ammonia. In the presence of 4-NA and various pesticides, strain AVPP64 greatly increased the growth and biomass of Vigna radiata and Crotalaria juncea plants. These results revealed that Bacillus sp. strain AVPP64 can be used as an inoculum for bioremediation of 4-NA contaminated soil and sustainable crop production even when pesticides are present.

The rapid determination of the bioaccessibility of polycyclic aromatic hydrocarbons (PAHs) in soils is challenging due to their slow desorption rates and the insufficient extraction efficiency of the available methods. Herein, magnetic poly(β-cyclodextrin) microparticles (Fe₃O₄@PCD) were combined with hydroxypropyl-β-cyclodextrin (HPCD) or methanol (MeOH) as solubilizing agents to develop a rapid and effective method for the bioaccessibility measurement of PAHs. Fe₃O₄@PCD was first validated for the rapid and quantitative adsorption of PAHs from MeOH and HPCD solutions. The solubilizing agents were then coupled with Fe₃O₄@PCD to extract PAHs from soil-water slurries, affording higher extractable fractions than the corresponding solution extraction and comparable to or higher than single Fe₃O₄@PCD or Tenax extraction. The desorption rates of labile PAHs could be markedly accelerated in this process, which were 1.3–12.0 times faster than those of single Fe₃O₄@PCD extraction. Moreover, a low HPCD concentration was sufficient to achieve a strong acceleration of the desorption rate without excessive extraction of the slow desorption fraction. Finally, a comparison with a bioaccumulation assay revealed that the combination of Fe₃O₄@PCD with HPCD could accurately predict the PAH concentration accumulated in earthworms in three field soil samples, indicating that the method is a time-saving and efficient procedure to measure the bioaccessibility of PAHs.

Hexabromocyclododecanes (HBCDDs) are common chemical additives in expanded polystyrene foam (EPS). To evaluate the bioaccumulation potential of endogenous HBCDDs in EPS microparticles by earthworms, two ecologically different species of earthworms (Eisenia fetida and Metaphire guillelmi) were exposed to soil added with EPS microparticles of different particle sizes (EPS2000, 830–2000 μm and EPS830, <830 μm). To clarify the accumulation mechanisms, leaching experiments using EPS microparticles in different solutions were conducted. After exposure to EPS microparticles-amended soils (S-EPS) for 28 d, the total concentrations of HBCDDs reached 307–371 ng g−1 dw in E. fetida and 90–133 ng g−1 dw in M. guillelmi, which were higher than those in earthworms exposed to the soil that was artificially contaminated with a similar level of HBCDDs directly (ACS). The accumulation of HBCDDs in earthworms was significantly influenced by EPS microparticles' size and earthworms' species. The total concentrations of HBCDDs in earthworms' cast were significantly higher than the theoretical concentration of HBCDDs in S-EPS, which suggested that EPS microparticles can be ingested by earthworms. The release rate of HBCDDs from EPS5000 (2000–5000 μm) into water-based solutions (<1%) after a 3.5-h incubation was far lower than that into earthworm digestive fluid (7%). These results illustrated that the ingestion of EPS microparticles and consequent solubilization of HBCDDs by digestive fluid play an important role in the accumulation of HBCDDs contained in EPS microparticles in earthworms. After a 28-d incubation with the soil solution, 4.9% of the HBCDDs was accumulatively leached from the EPS5000, which indicated that HBCDDs can be released from EPS microparticles to soil environment, and then accumulated by earthworms. Moreover, similar to those exposed to ACS, the diastereoisomer- and enantiomer-specific accumulation of HBCDDs in earthworms occurred when exposed to S-EPS. This study provides more evidence for the risk of microplastics to the soil ecosystem.

Surfactants are known to enhance the degradation of halogenated organics by nanoscale zerovalent iron (n-ZVI) or n-ZVI-based bimetallic particles, but the mechanism of the promotion is not well understood. In this study, we used nanoscale Ag/Fe particles (n-Ag/Fe) to degrade 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) in different surfactant solutions. The results show that the nonionic surfactant TX-100 had the best promoting effect, which might be attributed to the decrease in particle agglomeration and improvement of mass transfer efficiency after the adsorption of TX-100 on n-Ag/Fe. The distribution analysis of BDE-47 in solid and liquid phases indicates that when the concentration of TX-100 in aqueous solution was above critical micelle concentration, BDE-47 started to dissolve in the liquid phase. Thus, TX-100 micelles can enhance the mass transfer efficiency of BDE-47. However, a too high concentration of TX-100 (above 1.0 mM) would influence the promotion effect of BDE-47 degration, which might be attributed to the excessive and thicker micelles of TX-100 hindering the contact between BDE-47 and n-Ag/Fe. We also studied the degradation pathway of BDE-47 and its products, and found that surfactants did not change the degradation pathway of BDE-47.

In this report, we investigated how the presence of a polymer shell (poly(styrene-co-butyl acrylate) alters the toxicity of CuO NPs in Lemna gibba. Based on total Cu concentration, core–shell CuO NPs were 10 times more toxic than CuO NPs, inducing a 50% decrease of growth rate at 0.4 g l−1 after 48-h of exposure while a concentration of 4.5 g l−1 was required for CuO NPs for a similar effect. Toxicity of CuO NPs was mainly due to NPs solubilization in the media. Based on the accumulated copper content in the plants, core–shell CuO NPs induced 4 times more reactive oxygen species compared to CuO NPs and copper sulfate, indicating that the presence of the polymer shell changed the toxic effect induced in L. gibba. This effect could not be attributed to the polymer alone and reveals that surface modification may change the nature of NPs toxicity.

This study investigated the simultaneous desorption of trace metal elements and polychlorinated biphenyl (PCB) from mixed contaminated soil with a novel combination of biosurfactant saponin and biodegradable chelant S,S-ethylenediaminedisuccinic acid (EDDS). Results showed significant promotion and synergy on Pb, Cu and PCB desorption with the mixed solution of saponin and EDDS. The maximal desorption of Pb, Cu and PCB were achieved 99.8%, 85.7% and 45.7%, respectively, by addition of 10 mM EDDS and 3000 mg L−1 saponin. The marked interaction between EDDS and saponin contributed to the synergy performance. The sorption of EDDS and saponin on soil was inhibited by each other. EDDS could enhance the complexation of metals with the saponin micelles and the solubilization capabilities of saponin micelles for PCB. Our study suggests the combination of saponin and EDDS would be a promising alternative for remediation of co-contaminated soils caused by hydrophobic organic compounds (HOCs) and metals.

Solubilization of particulate Cu by different solutions, mimicking digestive fluids of deposit-feeders, was quantified in stable isotope ⁶⁵Cu-spiked sediments (with 3 days-2 months Cu-sediment contact time or aging). Copper solubilization generally decreased with prolonged aging. However, such decrease became less evident after 1 month and equilibrium of Cu in sediments could be reached after 2 months. Aging effects on Cu solubilization can be explained by the changes in Cu geochemical fractionation with aging: Cu generally transferred from more mobile phases (carbonate and Fe–Mn associated) to more refractory phases (organic associated and residual phase). Besides Cu geochemical fractionation, digestive fluid composition and different Cu solubilization pathways involved, as well as sedimentary organic content, could all affect the digestive solubilization of Cu and its change with aging. Our results emphasize the necessity of considering Cu aging in laboratory sediment toxicity experiments, and in risk assessment of Cu contaminated sediments.

To determine if trinitrotoluene (TNT) forms nonextractable residues in earthworms and to measure the relative degree of accumulation as compared to TNT and its deaminated metabolites, Eisenia fetida was exposed to ¹⁴C-TNT using dermal contact to filter paper or exposure to soil. Nonextractable residues made up 32–68% of total body burden depending on exposure media and depuration time. Parent TNT accounted for less than 3% of radioactivity, while ADNTs accounted for 7–38%. Elimination half-lives were 61–120h for TNT, ADNTs, and DANTs, which was significantly lower than the half-lives found for nonextractable residues, 201–240h. However, over 80% of the nonextractable residue was solubilized using weak acid (pH 2). Based on our findings that TNT accumulation occurs primarily as nonextractable residues, which have a longer half-life, and that nonextractable residues can be solubilized, we propose that nonextractable residues could be used as a selective biomarker for assessing TNT contamination.

In this study, two proficient Cadmium (Cd) resistant and plant growth-promoting actinobacterial strains were isolated from metal-polluted soils and identified as Streptomyces sp. strain RA04 and Nocardiopsis sp. strain RA07. Multiple abiotic stress tolerances were found in these two actinobacterial strains, including Cd stress (CdS), drought stress (DS) and high-temperature stress (HTS). Both actinobacterial strains exhibited multifarious plant growth-promoting (PGP) traits such as phosphate solubilization, and production of indole-3-acetic acid, siderophores and 1-aminocyclopropane-1-carboxylate deaminase under CdS, DS and HTS conditions. The inoculation of strains RA04 and RA07 significantly increased Sorghum bicolor growth and photosynthetic pigments under CdS, DS, HTS, CdS + DS and CdS + HTS conditions as compared to their respective uninoculated plants. The actinobacterial inoculants reduced malondialdehyde concentration and enhanced antioxidant enzymes in plants cultivated under various abiotic stress conditions, indicating that actinobacterial inoculants reduced oxidative damage. Furthermore, strains RA04 and RA07 enhanced the accumulation of Cd in plant tissues and the translocation of Cd from root to shoot under CdS, CdS + DS and CdS + HTS treatments as compared to their respective uninoculated plants. These findings suggest that RA04 and RA07 strains could be effective bio-inoculants to accelerate phytoremediation of Cd polluted soil even in DS and HTS conditions.

Uranium (U) contamination often occurs in the topsoil (arable layer), and is a serious threat to crop growth. However, conventional microbial reduction methods are sensitive to oxygen and cannot be used to treat aerobic topsoils. In this study, phosphate-solubilizing microorganisms (PSM) were isolated from U-contaminated topsoil and used for soil remediation. Microbial metabolites and products were analyzed, and the pathways and mechanisms of PSM immobilization were revealed. The results showed that strain PSM8 had the highest phosphate-solubilizing capacity (dissolved P was 208 ± 5 mg/L) and the highest U removal rate (97.3 ± 0.1%). Multi-technical analyses indicated that bacterial surface functional groups adsorbed (UO₂)²⁺ ions on the cell surface, glycolysis produced 3–10 mg/L of lactic acid (pH 4.7–6.0), and lactic acid solubilized Ca₃(PO₄)₂ to form stable chernikovite (a type of uranyl phosphate) on the cell surface. The coupled application of Ca₃(PO₄)₂ and strain PSM8 significantly reduced the bioavailability of soil U (62 ± 11%), converting U from the exchangeable to the residual phase and P from the steady to the available form. In addition, pot experiments showed that soil remediation promoted crop growth and significantly reduced U uptake and toxicity to photosynthetic systems. These findings demonstrate that PSM and Ca₃(PO₄)₂ are good coupled fertilizers for U-contaminated agricultural soil.
