The bacterial microbiota in florfenicol contaminated soils: The antibiotic resistome and the nitrogen cycle 全文

Soil antibiotic resistome and the nitrogen cycle are affected by florfenicol addition to manured soils but their interactions have not been fully described. In the present study, antibiotic resistance genes (ARGs) and nitrogen cycle genes possessed by soil bacteria were characterized using real-time fluorescence quantification PCR (qPCR) and metagenomic sequencing in a short-term (30 d) soil model experiment. Florfenicol significantly changed in the abundance of genes conferring resistance to aminoglycosides, β-lactams, tetracyclines and macrolides. And the abundance of Sphingomonadaceae, the protein metabolic and nitrogen metabolic functions, as well as NO reductase, nitrate reductase, nitrite reductase and N₂O reductase can also be affected by florfenicol. In this way, ARG types of genes conferring resistance to aminoglycosides, β-lactamases, tetracyclines, colistin, fosfomycin, phenicols and trimethoprim were closely associated with multiple nitrogen cycle genes. Actinobacteria, Chlorobi, Firmicutes, Gemmatimonadetes, Nitrospirae, Proteobacteria and Verrucomicrobia played an important role in spreading of ARGs. Moreover, soil physicochemical properties were important factors affecting the distribution of soil flora. This study provides a theoretical basis for further exploration of the transmission regularity and interference mechanism of ARGs in soil bacteria responsible for nitrogen cycle.

Like their parent polycyclic aromatic hydrocarbons (PAHs), substituted polycyclic aromatic hydrocarbons (SPAHs), including methyl PAHs (MPAHs), oxygenated PAHs (OPAHs), and chlorinated PAHs (ClPAHs), exist ubiquitously in urban and agricultural rivers. Although laboratory studies have found the biological toxicities of certain SPAHs to be higher than that of their parent PAHs, the ecological risk of SPAHs in rivers has been largely ignored. Here, we studied the distribution, source and transport of PAHs and SPAHs as well as ecological risks in the Chaobai River System, which experiences a high level of anthropogenic activity. The results show that the concentration of ΣOPAHs (321 ± 651 ng/L) was higher than that of ΣPAHs (158 ± 105 ng/L), ΣMPAHs (28 ± 22 ng/L), and ΣClPAHs (30 ± 12 ng/L). We also found that (S)PAHs in Chaobai River mainly originated from Beiyun River (53%–65%), which receives considerable municipal wastewater treatment plant effluent from Beijing. The major transport pathway of (S)PAHs from Chaobai River was likely for irrigation (83%–86%) and transportation into Yongdingxin River (13%–16%), which finally merged into the Bohai Sea. The mixed chronic risk of (S)PAHs (risk quotient = 45 ± 53) was higher than the mixed acute risk (risk quotient = 1.9 ± 1.4), with all sites facing chronic risk and 90% of sites experiencing acute risk. Although the chronic and acute risks of (S)PAHs to plants, invertebrates, and vertebrates were mainly from PAHs (97.5% to chronic risk and 96.5% to acute), SPAHs still posed a chronic risk to invertebrates and vertebrates (risk quotient > 1). Accordingly, the ecological risk of (S)PAHs in Chaobai River should be taken into consideration for ecosystem protection. The transmission of PAHs and SPAHs from Chaobai River may also pose potential risks to farmland through irrigation, as well as to the Bohai Sea via river water discharge.

Chinese dark tea is widely enjoyed for its multiple health-promoting effects and pleasant taste. However, its production involves fermentation by microbiota in raw tea, some of which are filamentous fungi and thus potential mycotoxin producers. Accordingly, whether mycotoxins pose health risk on dark tea consumption has become a public concern. In this study, a cleaning method of multi-functional column (MFC) and immunoaffinity column (IAC) in tandem combined to HPLC detection was developed and validated for determining ten mycotoxins of six groups (i.e., aflatoxins of B₁, B₂, G₁ and G₂, ochratoxin A, zearalenone, deoxynivalenol, fumonisins of B₁, B₂, and T-2) in dark teas. The interferences from secondary metabolites were effectively reduced, and the sensitivities and recoveries of the method were qualified for tea matrices. Six groups mycotoxins were determined in 108 samples representing the major Chinese dark teas by using the new method. Subsequently, the dietary exposure and health risks were evaluated for different age and gender groups in Kunming and Pu’er in China and Ulan Bator in Mongolia. The occurrence of zearalenone was 4.63% and that of ochratoxin A was 1.85%, with the other four groups mycotoxins were below the limits of quantification. The hazard index values for the five groups’ non-carcinogenic mycotoxins were far below 1.0. The deterministic risk assessment indicated no non-carcinogenic risks for dark tea consumption in the three areas. Probabilistic estimation showed that the maximum value of 95th percentile carcinogenic risk value for the aflatoxins was 2.12 × 10⁻⁸, which is far below the acceptable carcinogenic risk level (10⁻⁶). Hereby, six groups mycotoxins in Chinese dark tea showed no observed risk concern to consumers.

A novel amino-functionalized hydrochar material (referred to NH₂–HCs) was prepared and used as the soil amendment to immobilize multi-contaminated soils for the first time. The results showed that the application of NH₂–HCs significantly improved (P < 0.05) soil properties (i.e., pH value, cation exchange capacity and organic content). By introduction of NH₂–HCs, the contaminated soil showed the highest value of 96.2%, 52.2% and 15.5% reductions in Cu, Pb and Cd bioavailable concentrations and the leaching toxicity of Cu, Pb and Cd were remarkably reduced by 98.1%, 31.3% and 30.4%, respectively. Most of exchangeable Cu, Pb and Cd reduced were transformed into its less available forms of oxidizable and residual fractions. Potential ecological risk assessment indicated that the element Cd accounted for the most of total risks in NH₂–HCs amended soils. The mechanism study indicated that surface complexation, chemical chelating and cation-pi interaction of NH₂–HCs played a vital role in the immobilization of heavy metals. Pot experiments further verified that the application of NH₂–HCs significantly improved plant growth and reduced metal accumulations. The present study offered a novel approach to prepare amino-functionalized hydrochars with great potential as the green and alternative amendments for efficiently immobilizing heavy metals in multi-contaminated soil.

The establishment of a screening index would be a powerful tool to decide whether abandoned uranium mining areas should be rehabilitated or decommissioned. Thus, in this work we established a radiological index which uses the activity concentrations of different groups of gamma emitters from the natural radioactive series of ²³⁸U, ²³⁵U, and ²³²Th, as well as ⁴⁰K and ¹³⁷Cs. These activity concentrations were calculated by using the absorbed gamma radiation dose value of 175 nGy h⁻¹ specified in the U.S. Code of Federal Regulations. We studied our index in an abandoned uranium mining area in Salamanca, Western Spain, and found that the most influential factors in this area were the presence of organic matter in the soil and the possible effect that plants and fungi may have on the retention of these aforementioned radionuclides. In addition, the results showed that contaminants are migrating in an easterly direction in line with the prevailing wind direction and we were able to identify areas in which the radiological risk is likely high. The mean effective dose rate was 2.51 ± 0.98 mSv y⁻¹ which was equivalent to the levels obtained in previous works.

Firefighting foams contain per- and polyfluoroalkyl substances (PFAS) – a class of compounds widely used as surfactants. PFAS are persistent organic pollutants that have been reported in waterways and drinking water systems across the United States. These substances are of interest to both regulatory agencies and the general public because of their persistence in the environment and association with adverse health effects. PFAS can be released in large quantities during industrial incidents because they are present in most firefighting foams used to suppress chemical fires; however, little is known about persistence of PFAS in public waterways after such events. In response to large-scale fires at Intercontinental Terminal Company (ITC) in Houston, Texas in March 2019, almost 5 million liters of class B firefighting foams were used. Much of this material flowed into the Houston Ship Channel and Galveston Bay (HSC/GB) and concerns were raised about the levels of PFAS in these water bodies that have commercial and recreational uses. To evaluate the impact of the ITC incident response on PFAS levels in HSC/GB, we collected 52 surface water samples from 12 locations over a 6-month period after the incident. Samples were analyzed using liquid chromatography–mass spectrometry to evaluate 27 PFAS, including perfluorocarboxylic acids, perfluorosulfonates and fluorotelomers. Among PFAS that were evaluated, 6:2 FTS and PFOS were detected at highest concentrations. Temporal and spatial profiles of PFAS were established; we found a major peak in the level of many PFAS in the days and weeks after the incident and a gradual decline over several months with patterns consistent with the tide- and wave-associated water movements. This work documents the impact of a large-scale industrial fire, on the environmental levels of PFAS, establishes a baseline concentration of PFAS in HSC/GB, and highlights the critical need for development of PFAS water quality standards.

Microcystis blooms and their secondary metabolites microcystins (MCs) occurred all over the world, which have damaged aquatic ecosystems and threatened public health. Techniques to reduce the Microcystis blooms and MCs are urgently needed. This study aimed to investigate the algicidal and inhibitory mechanisms of a red pigment prodigiosin (PG) against the growth and MC-producing abilities of Microcystis aeruginosa (M. aeruginosa). The numbers of Microcystis cells were counted under microscope. The expression of microcystin synthase B gene (mcyB) and concentrations of MCs were determined by quantitative real-time polymerase chain reaction (qRT-PCR) and enzyme linked immunosorbent assay (ELISA) methods, respectively. The inhibitory effects of PG against M. aeruginosa strain FACHB 905 with 50% algicidal concentration (LC50) at 120 h was 0.12 μg/mL. When M. aeruginosa cells exposed to 0.08 μg/mL, 0.16 μg/mL, 0.32 μg/mL PG, the expression of mcyB of M. aeruginosa was down-regulated 4.36, 8.16 and 18.51 times lower than that of the control at 120 h. The concentrations of total MC (TMC) also were 1.66, 1.72 and 5.75 times lower than that of the control at 120 h. PG had high algicidal effects against M. aeruginosa, with the activities of superoxide dismutase (SOD) initially increased and then decreased after 72 h, the contents of malondialdehyde (MDA) increase, the expression of mcyB gene down-regulation, and MCs synthesis inhibition. This study was first to report the PG can simultaneously lyse Microcystis cells, down-regulate of mcyB expression and inhibit MCs production effectively probably due to oxidative stress, which indicated PG poses a great potential for regulating Microcystis blooms and MCs pollution in the environment.

Excess fluoride in drinking water is an environmental issue of increasing worldwide concern, because of its adverse effect on human health. Skeletal fluorosis caused by chronic exposure to excessive fluoride is a metabolic bone disease characterized by accelerated bone turnover accompanied by aberrant activation of osteoblasts. It is not clear whether Wnt/β-catenin signaling, an important signaling pathway regulating the function of osteoblasts, mediates the pathogenesis of skeletal fluorosis. A cross-sectional case-control study was conducted in Tongyu County, Jilin Province, China showed that fluoride stimulated the levels of OCN and OPG, resulting in accelerated bone turnover in patients with skeletal fluorosis. To investigate the influence of fluoride on Wnt/β-catenin signaling pathway, 64 male BALB/c mice were allotted randomly to four groups and treated with deionized water containing 0, 55, 110 and 221 mg/L NaF for 3 months, respectively. The results demonstrated that fluoride significantly increased mouse cancellous bone formation and the protein expression of Wnt3a, phospho-GSK3β (ser 9) and Runx2. Moreover, partial correlation analysis indicated that there was no significant correlation between fluoride exposure and Runx2 protein levels, after adjusting for β-catenin, suggesting that β-catenin might play a crucial role in fluoride-induced aberrant osteogenesis. In vivo, viability of SaoS2 cells was significantly facilitated by 4 mg/L NaF, and fluoride could induce the abnormal activation of Wnt/β-catenin signaling, the expression of its target gene Runx2 and significantly increased Tcf/Lef reporter activity. Importantly, inhibition of β-catenin suppressed fluoride-induced Runx2 protein expression and the osteogenic phenotypes. Taken together, the present study provided in vivo and in vitro evidence reveals a potential mechanism for fluoride-induced aberrant osteoblast activation and indicates that β-catenin is the pivot molecule mediating viability and differentiation of osteoblasts and might be a therapeutic target for skeletal fluorosis.

Strategies for reducing cadmium (Cd) content in polluted farmland soils are currently limited. A type of composite with nanoparticles incorporated into a hydrogel have been developed to efficiently remove heavy metals from sewage, but their application in soils faces challenges, such as organic hydrogel degradation due to oxygen exposure and slow Cd²⁺ release from soil constituents. To overcome these challenges, a composite with superior stability for long-term application in soil is required. In this study, ferrous sulfide (FeS) nanoparticle@lignin hydrogel composites were developed. The lignin-based hydrogels inherited lignin’s natural mechanical and environmental stability and the FeS nanoparticles efficiently adsorbed Cd²⁺ and enhanced Cd²⁺ desorption from soils by producing H⁺. The high sorption capacity (833.3 g kg⁻¹) of the composite was attributed to four proposed mechanisms, including cadmium sulfide (CdS) precipitation via chemical reaction (84.06%), lignin complexation (13.19%), hydrogel swelling (0.61%), and nanoparticle sorption (2.15%). In addition, Fe²⁺ displaced from the composite was gradually oxidized to form solid iron oxide hydroxide, which increased Cd²⁺ sorption. The composite significantly reduced the total, surfactant-soluble, and fixed Cd in heavily and lightly polluted paddy soils by 22.4–49.6%, 13.5–68.6%, and 40.1–16.6%, respectively, in 7 days.

In this study, the degradation of eight bromophenols (BPs), including monobromophenols (2-BP, 3-BP, and 4-BP), dibromophenols (2,4-DBP, 2,6-DBP, and 3,5-DBP), a tribromophenol (2,4,6-TBP) and a pentabromophenol (PBP), by a Fe(VI) reaction process at a pH of 8.0 was systematically studied. It was concluded that their degradation rates increased with increasing Fe(VI) concentrations in solution. The removal of 2,4,6-TBP, 2-BP, and 2,6-DBP was faster than that of the other five BPs, which could be attributed to the position of the substituting Br atom. Moreover, the direct oxidation and coupling reactions greatly influenced the reactivity of the bromophenols with Fe(VI). The electron paramagnetic resonance (EPR) analysis confirmed the presence of hydroxyl radicals in present system. The oxidation reaction products of PBP and 2-BP were recognized by an electrospray time-of-flight mass spectrometer; hydroxylation, hydroxyl substitution, the cleavage of the C–C bond, direct oxidation and polymerization via an end linking mechanism were noticeably found in the reaction process, resulting in the formation of polymerization products and causing hydroxylation to occur. Theoretical calculations further determined the possible oxidation sites of 2-BP and PBP. This study may provide comprehensive and important information on the remediation of BPs by Fe(VI).
