High-frequency assessment of air and water quality at a concentration animal feeding operation during wastewater application to spray fields 全文

Air and water quality at a concentrated animal feeding operation (CAFO) in Eastern North Carolina that uses a covered lagoon and anaerobic digester was evaluated for 2 weeks in August 2020. Real-time PM₂.₅ mass concentrations were determined using a reference ADR-1500 nephelometer and high-frequency measurements of dissolved inorganic nitrogen (DIN) were evaluated using autonomously logging sensors. Air and water quality parameters were assessed before, during and after wastewater from the lagoon was irrigated onto adjacent spray fields. Reference measurements were conducted alongside a HOBO weather station to collect real-time wind speed and direction, temperature, and humidity measurements. PM₂.₅ concentrations varied between 0 and 159 μg/m³ with an average concentration of 11 μg/m³, below EPA standard for secondary aerosols of 15 μg/m³. Higher PM₂.₅ concentrations were observed when wind originated from swine barns but not from covered lagoons. Water quality data showed that DIN concentrations downgradient from the CAFO were elevated relative to upstream concentrations. A groundwater seep that drains a spray field contained the highest average DIN concentration (31.0 ± 12.8 mg L⁻¹), which was 25 times greater than upstream DIN concentrations (1.2 ± 0.8 mg L⁻¹). Average DIN concentration at the downstream station was lower than the seep concentration (8.6 ± 16.2 mg L⁻¹), but approximately 8 times greater than upstream. Air quality data show that the lagoon cover was effective at mitigating air quality degradation, whereas DIN concentrations in water were similar to previous studies on CAFOs using open lagoons. In addition, air and water quality parameters were significantly (p < 0.001) higher after irrigation, indicating possible influence due to ammonia and nitrate elevation. Additional research is needed to compare high-frequency data collected from swine CAFOs using capped and uncapped lagoon systems to better understand spatiotemporal air and water quality trends of this practice.

Global climate change (GCC) significantly affects aquatic ecosystems. Continual use of pyrethroid insecticides results in contamination of these ecosystems and concurrent GCC raises the potential for synergistic effects. Resistance to pyrethroids has been documented in Hyalella azteca, a common epibenthic amphipod and model organism. Resistant H. azteca can bioconcentrate elevated amounts of pyrethroids and represent a threat to consumers via trophic transfer. In the present study, a predator of H. azteca, the inland silverside (Menidia beryllina), was used to examine the impacts of GCC on pyrethroid bioaccumulation via trophic transfer from resistant prey organisms. M. beryllina were fed ¹⁴C-permethrin dosed pyrethroid-resistant H. azteca for 14 days at three salinities (6, 13 and 20 practical salinity units (PSU)) and two temperatures (18 and 23 °C). Fish were analyzed for total body residues, percent parent compound and percent metabolites. Gene expression in liver and brain tissue were evaluated to assess whether dietary bioaccumulation of permethrin would impact detoxification processes, metabolism, and general stress responses. M. beryllina bioaccumulated significant amounts of permethrin across all treatments, ranging from 39 to 557 ng g⁻¹ lipid. No statistically significant effect of temperature was found on total bioaccumulation. Salinity had a significant effect on total bioaccumulation, owing to greater bioaccumulation at 6 PSU compared to 13 and 20 PSU, which may be due to alterations to xenobiotic elimination. Permethrin bioaccumulation and the interaction with temperature and salinity elicited significant transcriptional responses in genes relating to detoxification, growth, development, and immune response. Given the increased prevalence of pesticide-resistant aquatic invertebrates, GCC-induced alterations to temperature and salinity, and the predicted increase in pesticide usage, these findings suggest trophic transfer may play an important role in pesticide bioaccumulation and effects in predatory fish.

Exposure of females to fine particulate matter ≤2.5 μm in diameter (PM2.5) prior to pregnancy could produce adverse impact on fertility and enhances susceptibility of the offspring to a variety of diseases. In the current study, female C57BL/6 mice (6 weeks of age) were exposed to either concentrated PM2.5 or filtered air (average PM2.5 concentration: 115.60 ± 7.77 vs. 14.07 ± 0.38 μg/m⁻³) using a whole-body exposure device for 12 weeks. Briefly, PM2.5 exposure decreased anti-Müllerian hormone level (613.40 ± 17.36 vs 759.30 ± 21.90 pg mL⁻¹, P<0.01) and increased reactive oxygen species (ROS) level (45.39 ± 0.82 vs 24.20 ± 0.85 arbitrary unit in fluorescence assay, P<0.01) in oocytes. The exposure increased oocyte degeneration rate (21.5% vs 5.1%, respectively (P<0.01) and decreased the 2-cell formation rate (71.9% vs 86.0%, P < 0.01). Transcriptome profiling using RNA sequencing showed wide spectrum of abnormal expression of genes, particularly those involved in regulating the mitochondrial respiratory complex in oocytes and metabolic processes in blastocysts. The exposure decreased litter size (6 ± 0.37 vs 7 ± 0.26, P<0.05) and weight (1.18 ± 0.02 vs 1.27 ± 0.02 g, P<0.01). In summary, PM2.5 exposure decreased female fertility, possibly through increased ROS production in oocytes and metabolic disturbances in developing embryos. The cause-effect relationship, however, requires further investigation.

Toluene is a highly flammable and commonly used industrial chemical with severe health consequences on humans upon exposure and ingestion. In this study, multispecies bioassays were conducted using a species sensitivity distribution approach to determine acute and chronic hazardous concentrations of toluene in soil. Acute and chronic toluene toxicity tests were conducted with seven soil species from four taxonomic groups. The results from the toxicity tests were used to estimate the acute and chronic HC₅ (hazardous concentration for 5 % of species) of toluene in the terrestrial environment at 58.9 (5.4–639.6) mg kg⁻¹ and 2.2 (0.2–19.8) mg kg⁻¹, respectively. To the best of our knowledge, this is the first study to estimate the hazardous concentration of toluene in soil by conducting a battery of bioassays. These values can be used as references for the environmental risk assessment of chemical accidents involving toluene and estimating its impact on soil to protect the terrestrial environment.

Heavy metal pollution in paddy fields has caused widespread concerns due to the threat to food safety. The present study used low-cost sugarcane bagasse (SB) and two sugarcane bagasse materials modified with citric-acid (SSB) and citric-acid/Fe₃O₄ (MSB) to investigate their effects on the bioavailability of Cd in soil and Cd accumulations in rice in a pot experiment. The three organic amendments significantly decreased the Cd accumulation in plants by limiting its mobilization in soil. The MSB and SSB but not SB increased the soil pH and immobilized the Cd in soil significantly during the 120-day experiment. The amendments decreased Cd bioavailability through transforming to the stable fraction throughout the whole growth stage. The functional groups in the amendments (-OH, –COOH, C–O, -COO⁻ and Fe–O) and precipitates [Cd(NO₂)₂K(NO₂)₂, Cd(OH)₂ and Cd₇₅Zn₂₅Fe₂O₄] played active roles in Cd immobilization. Moreover, the three organic materials increased the content of Fe–Mn plaque on rice roots, which prevented its transport from soil to rice roots further. We also found that Fe competed with Cd for transporters and reduced potential Cd uptake and translocation in rice tissues. The addition of MSB and SB but not SSB inhibited the rice growth compared to the unamended control, indicating the potential of SSB in situ remediation. These results provide valuable information to use organic amendments for Cd passivation in soil and food safety.

Landfill mining and reclamation is a new strategy for addressing the lack of space available for new landfills and realizing the sustainable development of landfills. A gas-water joint bioreactor landfill is regulated by injecting water and/or recirculating leachate, and a blasting aeration system to optimize waste stabilization. In this study, four landfill reactors were constructed to investigate the effects of ventilation methods, including continuous (20 h d⁻¹) and intermittent aeration (4 h d⁻¹ in continuous or 2-h aeration per 12 h, twice a day), on the degradation of organic matter and volatile organic compound (VOC) emissions in comparison with traditional landfills. A total of 62 VOCs were detected in the landfill reactors. Among them, halogenated compounds had the highest abundance (39.8–65.4 %), followed by oxygenated compounds, alkanes and alkenes, and aromatic compounds. Both intermittent and continuous aeration could accelerate the degradation of landfilled waste and increase the volatilization rate of VOCs. Compared with intermittent aeration, the degradation of landfilled waste was more quickly in the landfill reactor with continuous aeration. However, intermittent aeration could create anaerobic-anoxic-aerobic conditions, which were conducive to the growth and metabolism of anaerobic and aerobic microorganisms in landfills and thereby reduced more than 63.4 % of total VOC emissions from the landfill reactor with continuous aeration. Moreover, intermittent aeration could reduce the ventilation rate and decrease the cost of aeration by 80 % relative to continuous aeration. Firmicutes, Bacteroidetes, Proteobacteria and Tenericutes predominated in the landfill reactors. The environmental variables including organic matter and VOCs concentrations had significant influences on microbial community structure in the landfilled waste. These findings indicated that intermittent aeration was an effective way to accelerate the stabilization of landfilled waste and reduce the cost and environmental risks in bioreactor landfills with gas-water joint regulation.

Combining hydrothermal treatment and composting is an effective method to dispose of penicillin fermentation residue (PFR), but the safety and related mechanism are still unclear. In this study, penicillin solution was hydrothermally treated to decipher its degradation mechanism, and then hydrothermally treated PFR (HT-PFR) was mixed with bulking agents at ratios of 2:0 (CK), 2:1.5 (T1), and 2:5 (T2) to determine the absolute abundance of antibiotic resistance genes (ARGs) and the succession of bacterial community. Results showed that penicillin was degraded to several new compounds without the initial lactam structure after hydrothermal treatment. During composting, temperature and pH of the composts increased with the raising of HT-PFR proportion, except the pH at days 2. After 52 days of composting, the absolute copies of ARGs (blaTEM, blaCMY2, and blaSFO) and the relative abundance of bacteria related to pathogens were reduced significantly (P < 0.05). Especially, the total amount of ARGs in the samples of CK and T1 were decreased to equal level (around 5 log₁₀ copies/g), which indicated that more ARGs were degraded in the latter by the composting process. In the CK samples, Bacteroidetes and Proteobacteria accounted for ~69.8% of the total bacteria, but they were gradually replaced by Firmicutes with increasing proportions of HT-PFR, which can be caused by the high protein content in PFR. Consisting with bacterial community, more gram-positive bacteria were observed in T1 and T2, and most of them are related to manganese oxidation and chitinolysis. As composting proceeded, bacteria having symbiotic or pathogenic relationships with animals and plants were reduced, but those related to ureolysis and cellulolysis were enriched. Above all, hydrothermal treatment is effective in destroying the lactam structure of penicillin, which makes that most ARGs and pathogenic bacteria are eliminated in the subsequent composting.

Bioinsecticides based on the bacterium Bacillus thuringiensis subsp. israelensis (Bti) are increasingly being applied directly into aquatic compartments to control nuisance mosquitoes and blackflies and are generally considered environmentally friendly alternatives to synthetic insecticides. Bti-based insecticides are considered highly selective, being Diptera-specific, and supposedly decompose rapidly in the environment. Nevertheless, their safety to non-target species and freshwater ecosystems has been questioned by recent studies, which in fact document possible indirect effects in aquatic food webs such as the decrease of prey availability to predators. This work aimed to evaluate the potential effects of a Bti-based insecticide (VectoBac® 12AS) on a freshwater macroinvertebrate community and on stream ecological functions by using artificial microcosm streams. Artificial microcosm streams were colonized with a macroinvertebrate community plus periphyton collected in a stream together with Alnus glutinosa leaf packs. They were exposed for 7 days to different Bti treatments (0, 12, 120, 1200 μg/L), which are within the recommended concentrations of application in aquatic compartments for blackfly and mosquito control. Besides invertebrate community structure and abundance, effects were evaluated regarding leaf decomposition and primary production as measures of ecosystem functioning. Community structure was significantly altered in all Bti treatments after 7 days of exposure, mostly due to a decline in chironomids, followed by oligochaetes, which both belong to the deposit-feeders’ functional group. Direct effects on oligochaetes are surprising and require further research. Also, reductions of leaf decomposition due to Bti-induced sublethal effects on shredders (reduced feeding) or mortality of chironomids (that can also feed on coarse organic matter) observed in our study, represent potential indirect effects of Bti in aquatic ecosystems. Our short-exposure experiment evidenced some negative effects on stream benthic invertebrate communities and on ecosystem functioning that must be considered whenever Bti is used in water bodies for blackfly or mosquito control programs.

Bats are poorly understood as a reservoir of multidrug-resistant strains; therefore, the aim of this study was to determine molecular characterization of multidrug-resistant Enterococcus strains isolated from bat species from Poland. A multi-stage analysis based on targeted isolation of drug-resistant strains (selective media with tetracycline, chloramphenicol, gentamicin, streptomycin, and vancomycin), determination of the phenotypic profile of drug-susceptibility using the disc diffusion method, and amplification of DNA fragments surrounding rare restriction sites (ADSRRS-fingerprinting) was used for the isolation and differentiation of strains. The applied strategy finally allowed identification of E. faecalis resistant to at least one antimicrobial in 47.2% of the single-animal group and in 46.9% of the pooled samples of bat’s guano. Out of the 36 distinct isolates, 69% met the criteria of multi-drug resistance, with a dominant combination of resistance to tetracycline, erythromycin, and rifampicin. Simultaneously, 41.6% of the strains were high-level aminoglycoside resistant (HLAR). In most strains, phenotypic resistance was reflected in the presence of at least one gene encoding resistance to a given drug. Moreover, our research results show that some genes were detected simultaneously in the same strain statistically significantly more frequently. This may confirm that the spread of some genes (tetM and ermB or aph (3′)-IIIa as well as gelE and aac (6′)-Ie-aph (2″)-Ia or ant (6)-Ia) is associated with their common occurrence on the same mobile genetic element.To our knowledge, this is the first analysis of multidrug-resistance among E. faecalis isolated from bats. Our research demonstrates that the One Health concept is not associated exclusively with food-producing animals and humans, but other species of wildlife animals should be covered by monitoring programs as well.We confirmed for the first time that bats are an important reservoir of multi-resistant E. faecalis strains and could have a great impact on environmental resistance.

High ammonia (NH₃) emissions from fertilized soil in China have led to various concerns regarding environmental safety and public health. In response to China's blue skies protection campaign, effective NH₃ reduction measures need to consider both mitigation efficiency and food security. In this context, we conducted a meta-analysis (including 2980 observations from 447 studies) to select effective measures based on absolute (AV) and yield-scaled (YSAV) NH₃ volatilization reduction potential, with the aim of establishing a comprehensive NH₃ mitigation framework covering various crop production sectors, and offering a range of potential solutions. The results showed that manipulating crop density, using an intermittent irrigation regime for paddy field rice, applying N as split applications or partially substituting inorganic fertilizer N with organic N sources could achieve reductions in AV and YSAV reduction of 10–20 %; adopting drip irrigation regimes, adding water surface barrier films to paddy fields, or using double inhibitor (urease and nitrification), slow-release or biofertilizers could achieve 20–40 % mitigation; plastic film mulching, applying fertilizer by irrigation or using controlled-release fertilizers could yield 40–60 % reduction; use of a urease inhibitor, fully substituting fertilizer N with organic N, or applying fertilizer by deep placement could decrease AV and YSAV by over 60 %. In addition, use of soil amendments, applying suitable inorganic N sources, or adopting crop rotation, intercropping or a rice-fish production model all had significant benefits to control AV. The adoption of any particular strategy should consider local accessibility and affordability, direct intervention by local/government authorities and demonstration to encourage the uptake of technologies and practices, particularly in NH₃ pollution hotspot areas. Together, this could ensure food security and environmental sustainability.
