Atualização sobre a epidemiologia de Dirofilaria immitis na América do Sul e no México: revisão de literatura

Dirofilaria immitis é um nematoide de ampla distribuição geográfica, que ocorre com maior frequência em áreas quentes e úmidas do planeta. O primeiro registro de sua ocorrência na América do Sul foi realizado em 1878, no Brasil. Naquela época os registros eram poucos e raramente de fácil obtenção, razão pela qual reuni-los facilitará a recuperação da memória ao longo dos anos. Quatro bases de dados (Scopus, MEDLINE, LILACS e PubMed) foram estudadas utilizando-se as palavras-chave “Dirofilaria” ou “heartworm”, os nomes dos países da América do Sul e o México. Nenhum registro foi encontrado para quatro países (Bolívia, Equador, Guiana Francesa e Uruguai) e para outros três (Suriname, Guiana e Paraguai) os registros eram antigos. Apenas o Chile é o território onde houve estudos registrados com ausência do parasita. Os outros países (México, Peru, Colômbia, Venezuela, Argentina e Brasil) apresentam registros com frequência variável no tempo ou no espaço. Assim, as informações reunidas indicam que infecções por D. immitis ocorrem na maior parte da América do Sul e no México e que os médicos veterinários devem instituir programas preventivos para garantir cuidados médicos de qualidade aos pacientes e para proteger a saúde destes e de suas famílias.

In this study ELISA was standardized for detecting antibodies against Hydropericardium Syndrome Virus. Factors like Antigen, Conjugate and incubation time were studied. Standardization was carried out by checker board titration method. Optimum incubation time for colour development was found to be 15 minutes while 50 mug/ml of antigen gave better results than 100 mug/ml. The conjugate dilution of 1:500 was observed to give satisfactory results as determined by regression analysis. Cut off value was determined by adding 2SDs and 3SDs in the mean optical density (OD) values of negative serum samples. It was found that mean OD+2SD (0.0684) resulted in better diagnostic sensitivity) and diagnostic specificity than mean OD+3SDs (0.0741).

Clinical and epidemiological characterization of leishmaniasis in dogs in São Paulo State | Caracterização clínica e epidemiológica das leishmanioses em cães no Estado de São Paulo

<!--[if gte mso 9]><xml> <o:OfficeDocumentSettings> <o:AllowPNG/> </o:OfficeDocumentSettings> </xml><![endif]--><p class="MsoNormal"><span style="mso-ansi-language: EN-US;" lang="EN-US">The occurrence of visceral or cutaneous canine leishmaniasis cases among dermatophatic dogs admitted to the veterinary teaching hospital School of Veterinary Medicine and Animal Husbandry - University of São Paulo from 1997 to 2008 was retrospectively characterized. For the diagnosis of 427 animals, serological (Enzyme-Linked Immunosorbent Assay, Indirect Immunofluorescence and/or Complement Fixation), parasitological and molecular (Polymerase Chain Reaction) tests were performed, which resulted in 117 (27.4%) confirmed positive cases. Of these, 96 (82.1%) were pure breed dogs and the others 21 (17.9%) were cross breed, being 57 (48.7%) females and 60 (51.3%) males. In terms of age, 26 (22.2%) were younger than 36 months, 48 (41%) were between 36 and 84 months, and 43 (36.8%) were older than seven years old. Sixty-four (54.7%) dogs were considered oligosymptomatic, 44 (37.6%) symptomatic, and the remaining nine (7.7%) asymptomatic. The main clinical findings were, in descending order, dermal lesions, lymphadenopathy, anemia, dysorexia, prostration and splenomegaly. As for hematological disorders, relative and absolute lymphopenia was the most important findings. Ninety-six (82.1%) of the positive animals correspond to allochthonus cases from São Paulo municipality.</span></p> | <!--[if gte mso 9]><xml> <o:OfficeDocumentSettings> <o:AllowPNG/> </o:OfficeDocumentSettings> </xml><![endif]--><p class="MsoNormal">Caracterizou-se retrospectivamente, a partir do ano de 1997 até março de 2008, a ocorrência de casos de leishmaniose, visceral ou tegumentar, dentre os cães dermatopatas atendidos em serviço especializado de hospital-escola veterinário de Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo. Para o diagnóstico em 427 animais, foram utilizados testes sorológicos (Ensaio Imunoenzimático Indireto, Imunofluorescência Indireta e/ou Fixação de Complemento), parasitológicos diretos e moleculares (Reação em Cadeia da Polimerase), que resultaram na confirmação de 117 (27,4%) casos positivos. Da amostragem, 96 (82,1%) eram cães de raça definida, os demais 21 (17,9%) não dispunham de plena definição racial, sendo 57 (48,7%) fêmeas e 60 (51,3%) machos. Quanto à faixa etária, 26 (22,2%) apresentavam idade inferior a 36 meses, 48 (41%) entre 36 e 84 meses e 43 (36,8%) ultrapassavam o sétimo ano de vida. Considerou-se 64 (54,7%) cães como oligossintomáticos, 44 (37,6%) sintomáticos e os nove (7,7%) remanescentes como assintomáticos. As principais alterações clínicas observadas foram, em ordem decrescente, lesões tegumentares, linfonodomegalia, anemia, disorexia, prostração e esplenomegalia. Quanto às alterações hematológicas, a mais importante foi a linfopenia relativa e absoluta. Dos animais positivos para a enfermidade, 96 (82,1%) correspondeu a casuística alóctone ao Município de São Paulo.</p><!--[if gte mso 9]><xml> <w:WordDocument> <w:View>Normal</w:View> O objetivo do presente estudo foi pesquisar a ocorrência de infecção por Leishmania sp. em cães (N = 491) domiciliados no município de Florianópolis, Santa Catarina, considerada uma região indene para leishmaniose visceral até o ano de 2011, quando foram notificados casos autóctones da doença canina. A soroprevalência na população foi avaliada por ELISA (0,4%; 2/491) e RIFI (4,09%; 24/491). Somente um cão apresentou sororeatividade em ambos os métodos sorológicos, totalizando 25 (5,3%) animais sororeagentes. O DNA de Leishmania sp., obtido de uma amostra do sangue total desse animal, foi amplificado por PCR convencional e PCR em Tempo Real. Não foi possível realizar o sequenciamento do DNA amplificado e, deste modo, determinar a espécie de Leishmania envolvida. Os nossos resultados sugerem a necessidade de uma investigação epidemiológica minuciosa em Florianópolis. | The aim of the present study was to investigate the occurrence of Leishmania sp. infection in dogs (N = 491) living in the municipality of Florianópolis, Santa Catarina (SC), Brazil, which was considered a disease-free region for visceral leishmaniasis until 2011, when autochthonous cases of canine disease were notified. Seroprevalence in this population was assessed by ELISA (0.4%; 2/491) and IFAT (4.09%; 24/491). Only one dog exhibited seroreactivity in both serological methods, comprising a total of 25 (5.3%) seroreagent animals. Leishmania sp. DNA, obtained from a sample of whole blood of this animal, was amplified by both conventional and Real-Time PCR. Sequencing of the amplified DNA and, thereby, determination of the Leishmania species involved, was not possible. Our results suggest the necessity of a thorough epidemiological investigation in Florianópolis.

Nineteen kittens divided into four groups were fed with brains of mice infected with rabies viruses. Each four kittens (group I) received four brains infected with the PV fixed strain; nine kittens (group II) ingested 4-5 brains infected with the field isolate T-9/95, isolated from the Desmodus rotundus vampire bat; two kittens (group III) fed ten T-9/95-infected brains, and four cats consumed 32-37 PV strain-infected brains. One adult male, inoculated into masseter muscle with a 20% T-9/95-infected brain suspension, presented rabies after an incubation period of six days, followed with 8 days of clinical evolution, and died thereafter and this cat was considered as the rabies "positive standard". After observing for 20-230 days, all the cats feeding the rabid brains were submitted to euthanasia, by using Acepran®, Zoletil®, and T-61®. At necropsy, samples of brain, heart, lung, kidney, submaxillary salivary gland, and cervical medulla were collected from all the cats and further submitted to the direct fluorescence antibody test (dFA), mouse inoculation test (MIT) and to the reverse transcriptase-polymerase chain reaction (RT-PCR) technique. Brain, cervical medulla, and the submaxillary salivary gland of the positive standard cat were dFA-positive, and brain and cervical medulla were positive for MIT. All specimens of this cat tested by the RT-PCR were found positive. No animals ingesting PV or T-9/95 virus-infected brains developed clinical signs and all materials tested were negative by dFA and MIT. Several specimens, however, showed positive reactions by the RT-PCR technique, but cats were resistant to rabies through the viruses administered orally. | Dezenove gatos, divididos em quatro grupos, foram alimentados com cérebros de camundongos infectados com vírus de raiva. Cada um dos quatro gatos (grupo I) receberam quatro cérebros infectados com vírus fixo PV; nove gatos (grupo II) ingeriram 4-5 cérebros infectados com uma amostra de campo T-9/95, isolada do morcego Desmodus rotundus; dois gatos (grupo III) ingeriram 10 cérebros infectados com T-9/95 e quatro gatos (grupo IV) ingeriram 32-37 cérebros infectados com vírus PV. Um macho adulto, inoculado no músculo masséter, com uma suspensão cerebral a 20% da amostra T-9/95, desenvolveu raiva após período de incubação de seis dias, seguidos por oito dias de evolução clínica, morrendo em seguida. Este gato foi denominado de "padrão positivo". Após observação por um período de 20-230 dias, todos os gatos que receberam cérebros foram submetidos à eutanásia, utilizando Acepran®, Zoletil® e T-61®. À necropsia, foram colhidas amostras do cérebro, coração, pulmão, rim, glândula salivar submaxilar e medula cervical e submetidas à prova de imunofluorescência direta (IFD), inoculação em camundongos (IC), e reação em cadeia pela polimerase-transcriptase reversa (RT-PCR). No "padrão positivo", cérebro, medula cervical e glândula salivar foram positivos à IFD e à IC, cérebro e medula cervical foram os positivos. Todos os espécimes do "padrão positivo" foram positivos à RT-PCR. Nenhum animal que ingeriu cérebros contendo amostras de vírus PV ou T-9/95 apresentou sinais clínicos e todos os espécimes testados foram negativos à IFD e IC, no entanto, alguns espécimes reagiram positivamente à RT-PCR, porém, os gatos foram resistentes à raiva com vírus administrados oralmente.

The agar gel immunodiffusion test (AGID), agar gel immunodiffusion test in sera treated by 2-mercaptoethanol (ME-AGID) and complement fixation test (CFT), applied to diagnosis of the canine brucellosis due to Brucella canis, were compared. For this purposes, 80 blood samples from dogs were collected during the rabies vaccination campaign of Santana de Parnaíba municipality, State of São Paulo, in August 1999. The antigens used in the AGID and ME-AGID tests were lipopolysaccharides and proteins from Brucella ovis, strain Reo 198, and in the CFT, the antigen employed was Brucella ovis, strain 63/290. The serological tests were compared two by two by Mc Nemar test, and the agreement was analyzed by the Kappa indicator. The agreement between the CFT and ME-AGID was moderate (Kappa = 0.54), between the AGID and CFT was fair (Kappa = 0.36) and between the AGID and ME-AGID was fair (Kappa = 0.39). | Foram comparados os testes de imunodifusão em gel de ágar (IDGA), imunodifusão em gel de ágar em soros tratados pelo 2-mercaptoetanol (IDGA-ME) e reação de fixação de complemento (CFT), aplicados ao diagnóstico da infecção de caninos por Brucella canis. O antígeno utilizado nas técnicas de IDGA e IDGA-ME constituiu-se de lipopolissacarídeos e proteínas de Brucella ovis, amostra Reo 198, e na CFT, o antígeno empregado foi a Brucella ovis, amostra 63/290. Foram examinadas 80 amostras de soro sanguíneo de cães colhidas durante a campanha de vacinação anti-rábica animal do Município de Santana de Parnaíba-SP, realizada em agosto de 1999. As provas sorológicas foram comparadas, duas a duas, pelo teste de Mc Nemar, e a concordância foi analisada pelo indicador Kappa. A concordância entre as técnicas de CFT e IDGA-ME foi regular (Kappa = 0,54), entre as técnicas de IDGA e CFT foi sofrível (Kappa = 0,36) e entre as provas de IDGA e IDGA-ME também foi sofrível (Kappa = 0,39).

Bovine serums were evaluated by the plate agglutination, tube agglutination, buffered plate antigen and 2-Mercaptoethanol tests for bovine brucellosis diagnosis. All presented a good concordance when considering the official Ministry of Agriculture interpretation. High concordance was verified for the buffered plate antigen and 2-Mercapthoetanol tests. This study suggests the use of the buffered plate antigen test as a screening test for bovine brucellosis diagnosis. | Avaliaram-se comparativamente as provas de soroaglutinação rápida, soroaglutinação em tubo, 2-mercaptoetanol e antígeno tamponado acidificado no diagnóstico da brucelose bovina. Todas as provas apresentaram boa concordância quando considerada a interpretação preconizada pelo Ministério da Agricultura do Brasil. As provas do antígeno tamponado acidificado e do 2-mercaptoetanol apresentaram alta concordância. Neste sentido, o presente estudo propõe o uso da prova do antígeno tamponado acidificado como triagem para o diagnóstico da brucelose bovina.

The presence of antibodies against BHV-1 was evaluated in young bovines from fifteen different counties distributed along North, South and Central regions of the state of Bahia. Bovines with age ranged from 1 to 4 years old, non-vaccinated against BHV-1 and without diagnostic of BHV-1 infection were chosen for blood collection. A total of 558 serum samples were collected equivalent to 10% of the population in each farm. The sera were submitted to ELISA (HerdCheck-IDDEX) and Microserum neutralization (MSN) tests. The MSN was performed in monolayers of MDBK cells in microplates using Los Angeles BHV-1 strain. The results of serum samples by ELISA showed that 56% (314/558) were positive and 44% (244/558) were negative. The sera assayed by MSN showed that 48% (269/558) were negative and 52% (289/558) were positive. These positive sera demonstrated that 54% had serumneutralization titer between 4-16, 35% between 32-64 and 11% >; or = 64. Our results suggest that the virus is circulating actively in herds. Furthermore, the high diffusion of the BHV-1 in young bovines could represent a constant source of viral infection in the herd. | A presença de anticorpos contra Herpesvírus Bovino tipo 1 (HBV-1) foi avaliada em bovinos jovens provenientes de quinze municípios diferentes, distribuídos nas regiões Norte, Sul e Central do estado da Bahia. Foram coletadas amostras de sangue de bovinos de 1 a 4 anos de idade, não vacinados contra HBV-1 e sem prévio diagnóstico de infecção pelo vírus HBV-1. O total de amostras correspondeu a 558 soros, equivalente a 10% da população bovina de cada fazenda. Os 558 soros foram submetidos à técnica de Microssoroneutralização (MSN) em microplacas, utilizando-se monocamadas de células MDBK e a cepa de HBV-1 Los Angeles, e ao teste de ELISA comercialmente adquirido: HerdCheck-IDDEX. Os resultados da amostragem feito pelo teste de ELISA mostraram que 56% (314/558) dos soros foram positivos e 44% (244/558), negativos. A técnica de MSN mostrou que 48% (269/558) dos soros foram negativos e 52% (289/558) foram positivos. Destes soros positivos, 54% tiveram títulos soroneutralizantes entre 4-16, 35% entre 32-64, e 11% >; ou = 64. Os resultados sugerem que o HBV-1 esteja circulando ativamente na população bovina jovem. Por outra parte, a alta difusão do vírus em animais jovens representaria uma fonte contínua de infecção viral para o rebanho.

Film test was performed in fecal samples of Eurytrema sp infected and non infected cattle but both groups showed similar results. So, it was concluded that film test is an unreliable one for screening Eurytrema sp infection in cattle. | Amostras fecais de bovinos comprovadamente parasitados e outras de animais possuidores de pâncreas macroscopicamente normais (controle) foram submetidas à prova do filme. Os resultados mostraram que tanto os animais parasitados quanto aqueles do grupo controle comportaram-se de modo semelhante à prova do filme. Com isto, concluiu-se que o referido teste deve ser desconsiderado para os fins propostos de diagnóstico presuntivo de infecções por Eurytrema sp em bovinos.
