Objectives: The objective of this study was to isolate and identify Staphylococcus aureus and Escherichia coli from raw milk samples of cattle and buffalo, and to evaluate the antibiotic sensitivity pattern. Materials and methods: A total of 34 milk samples were collected twice from 17 different healthy cattle (n=14) and buffaloes (n=3) at one-month interval, and analyzed in laboratory by staining, cultural and biochemical characteristics followed by polymerase chain reaction targeting nuc gene of S. aureus and 16 S rRNA of E. coli. Antibiotic sensitivity pattern of the isolated bacteria was assessed using the disc diffusion method. Results: Confirmation of the isolates as S. aureus and E. coli were carried out by PCR using nuc gene, 16S rRNA gene specific primers specific for S. aureus and E. coli respectively. A total of 12 samples (35.29%; 11 from cattle, 1 from buffalo) were found to be positive for S. aureus; 5 and 7 during first and second month, respectively. The E. coli were found in three samples (2 from cattle, 1 from buffaloe); one in first month and two in the second month. The antibiotic sensitivity test using 4 commonly used antibiotics indicated that the most of the isolates were resistant to Gatifloxacin and one isolate showed intermediate resistance to Ofloxacin while sensitive to Ciprofloxacin and Levofloxacin. Conclusion: Two different species of bacteria i.e., S. aureus and E. coli are contaminating with milk samples. The pathogenic bacteria can be controlled effectively by using Ciprofloxacin and Levofloxacin in the case of mastitis in cattle and buffaloes in Bangladesh. [J Adv Vet Anim Res 2016; 3(1.000): 62-67]

Objectives: Here we determined the prevalence of Salmonella in cloacal swabs and pharyngeal swabs of apparently healthy pigeons sold in the live bird markets and villages in and around Bangladesh Agricultural University Campus, Mymensingh, Bangladesh. Materials and methods: A total of 50 samples, comprised of cloacal swabs (n=24) and pharyngeal swabs (n=26) were collected. The samples were processed, and Salmonella was isolated through a series of conventional bacteriological techniques and biochemical tests followed by polymerase chain reaction (PCR). Results: The prevalence rate of Salmonella was found to be 37.5% (n=9/24) in cloacal swabs and 30.77% (n=8/26) in pharyngeal swabs with an overall prevalence rate of 34% (n=17/50). The prevalence rate of Salmonella pigeon varied slightly among locations; 34.62% (n=9/26) in live bird markets, and 33.33% (n=8/24) in villages. Molecular detection of 17 Salmonella isolates obtained from biochemical test was performed by genus specific PCR, where all of them amplified a region of 496-bp segment of the histidine transport operon gene. Antibiogram study revealed multi-drug resistant traits in most of the isolates tested. The highest resistance was found against Ampicillin (88.23%) followed by Cephalexin (82.35%). The rate of sensitivity of the isolates to Ciprofloxacin was 100% followed by Azithromycin (82.35%), Gentamicin (76.47%) and Nalidixic acid (76.47%). Conclusion: Our findings suggest that pigeons carry multi-drug resistant Salmonella that may transfer to the humans and animals. [J Adv Vet Anim Res 2016; 3(1.000): 51-55]

Objective: An attempt was undertaken for the detection and characterization of Avibacterium paragallinarum from clinically sick broiler chickens during field outbreaks. Materials and methods: Nasal and ocular discharges (n=6), tracheal swab (n=6), tracheal washing (n=4) and infraorbital sinus exudates (n=4) were collected aseptically from broiler chickens (n=10). To isolate A. paragallinarum, the clinical samples were cultured onto blood agar and chocolate agar enriched with Nicotinamide Adenine Dinucleotide (NAD) and feeder organism (Staphylococcus aureus). Identification of A. paragallinarum was performed by Gram staining reaction, sugar fermentation profiles using five basic sugars (Dextrose, Maltose, Sucrose, Lactose and Mannitol) and biochemical tests (Indole, Voges Proskauer and Methyl red tests). Antibiogram of the bacterial isolates of infected chicken was performed against five antibiotics namely Ciprofloxacin, Azithromycin, Gentamicin, Ampicillin and Cefalexin using disk diffusion method. Results: Results of colonial morphology, Gram staining reaction, sugar fermentation and biochemical tests confirmed one isolate as A. paragallinarum. The overall prevalence of IC in broiler chicken was 10% (1 of 10). This isolate was found to be sensitive to Ciprofloxacin, Azithroycin and Gentamicin and resistant to Ampicillin and Cefalexin. Objective: The study was conducted to determine the prevalence of Staphylococcus aureus in cockroaches (Periplaneta americana), and to assess the antimicrobial resistance profiles of the isolated bacteria. Materials and methods: A total of 150 cockroaches (P. americana) were randomly captured from three households and four restaurants in Chittagong City Corporation, Bangladesh during July to December 2014. The cockroaches were transported to the bacteriology laboratory at the Poultry Research and Training Centre (PRTC), Chittagong Veterinary and Animal Sciences University. The isolation and identification of Staphylococcus spp. from the external surface wash and gut homogenates by pooling cockroaches were done by following conventional bacteriological examinations followed by biochemical characterization. The antibiotic susceptibility profiles of the isolates were determined using disc diffusion method. Results: In this study, the overall prevalence of S. aureus was 38% (n=57/150). Higher prevalence of Staphylococcus spp. was observed among the cockroaches from restaurant (49.3%; n=37/75) as compared to those of households (26.7%; n=20/75) having a significant difference (P<0.05). Highest level of resistance by the Staphylococcus spp. was found to Penicillin (68%) followed by Erythromycin (60%), Oxacillin (46%) and Clindamycin (31%). On the other hand, the Staphylococci isolates were highly sensitive to Cephalothin (84%) and Kanamycin (65%). Conclusion: The rational use of antibiotics needs to be adopted in both human and animal medicine practices to prevent the emergence of drug resistant Staphylococcus spp. [J Adv Vet Anim Res 2016; 3(3.000): 221-228]

Enumeration of nematode eggs in fecal samples using the McMaster technique and morphological identification of third stage larvae from fecal cultures have been extensively used with satisfactory results in the diagnosis of the gastrointestinal nematode infections in ruminants. In order to improve sensitivity and accuracy, other approaches for quantification of eggs have been employed, like the FLOTAC and Mini-FLOTAC techniques. Results obtained in different studies indicate that fecal egg counts are a reliable measure of the size of the worm burden. However, the immunological status of the animals should be taken into consideration to interpret the results of the fecal examination. Molecular techniques have also been useful in the diagnosis of parasitic diseases. The ultimate in diagnosis has been the development of robotic platforms that enable separation of eggs from feces. Because manipulation is minimal, good quality DNA from eggs is obtained, which is used for amplification, and finally, produces a result indicating the degree of the infection by the different parasite species in mix infections. The ideal method should be reliable, friendly to non-experts and quick to perform. With the advance in robotics, bioinformatics and molecular biology, methods with such characteristics are expected to become available and affordable to be used in laboratories for the routine diagnosis of gastrointestinal nematodes of ruminants.

A quantificação de ovos de nematoides em amostras de fezes e a identificação morfológica das larvas de terceiro estágio a partir de culturas de fezes têm sido amplamente utilizadas, com resultados satisfatórios para o diagnóstico das infecções por nematoides gastrintestinais em ruminantes. Com o intuito de melhorar a sensibilidade na detecção dos ovos, outras técnicas, tais como Flotac e Miniflotac, têm sido empregadas. Resultados obtidos em diferentes estudos indicam que a contagem de ovos nas fezes é uma medida confiável e representativa da magnitude da carga parasitária. No entanto, o estado imunológico dos animais deve ser considerado na interpretação dos resultados coproparasitológicos. As técnicas moleculares também têm sido úteis no diagnóstico das enfermidades parasitárias. Os maiores avanços no diagnóstico têm sido propiciados pelo desenvolvimento de plataformas robóticas, que separam os ovos das fezes com pouca manipulação, propiciando a obtenção de DNA de ovos de boa qualidade, o que resulta em uma amplificação que possibilita indicar o grau da infecção pelas diferentes espécies de parasitas em infecções mistas. O método ideal deve ser preciso, rápido e de fácil execução por não-especialistas. The aim of the present study was to evaluate the effect of two crude protein contents (CP) and two main nitrogen sources in the diet of dairy cows, using sugarcane as forage, on intake, total apparent digestibility, milk yield and composition, and nitrogen balance. Twelve Holstein cows in late lactation were assigned in three contemporary square, in a Latin Square design 4 × 4, with 2 × 2 factorial treatment arrangement, in 21-day trial by period, 14 for diet adaptation and the last seven for sampling and analysis. Cows were housed in individual stalls and fed a total mixed ration (TMR) "ad libitum" composed of two main nitrogen sources (cottonseed meal or whole raw soybean grain) and two levels of CP (130g or 148g/kg dry matter - DM) in the diet. In the present study, there was an interaction effect between nitrogen source and crude protein on DM intake (DMI). Cows fed whole raw soybean had higher DMI when the CP content of the diet was 130 g CP/kg DM, while cows fed cottonseed meal had higher DMI when the CP content of the diet was 148 g CP/kg DM. A similar result of DMI was observed for organic matter intake, whereas there was no interaction effect between dietary nitrogen source and crude protein content on intake of other nutrients. Total apparent digestibility of NDF and TDN was higher when soybean was used as the main diet nitrogen source; whereas the digestibility of dietary CP was not changed by the nitrogen sources. Although there were changes in the DMI and the apparent digestibility coefficient, in the current study, there was no effect of dietary crude protein content and nitrogen source on milk yield. In conclusion, the use of low dietetic concentrations of crude protein (130 g/Kg of DM) does not change the lactating performance of dairy cows and may reduce diet cost, while the choice between whole raw soybean grain or cottonseed meal as a protein ingredient may depends only on the availability and/or price, as both ingredients result in similar performance.

O objetivo do presente estudo foi avaliar o efeito de dois teores proteicos (PB) e duas fontes nitrogenadas na dieta de vacas leiteiras, utilizando cana-de-açúcar como forragem, sobre o consumo, digestibilidade aparente total, produção e composição do leite, e o balanço de nitrogênio. Doze vacas leiteiras Holandesas em estágio intermediário de lactação foram distribuídas em três quadrados contemporâneos, em um delineamento em quadrado latino 4 × 4, com arranjo fatorial de tratamentos 2 × 2, em períodos de 21 dias, onde os primeiros 14 dias foram destinados para a adaptação às dietas e os últimos sete para as coletas de dados e análises. As vacas foram alocadas em baias individuais em um sistema free-stall, e alimentadas com dietas totalmente misturada (TMR) “ad libitum” compostas por duas fontes nitrogenadas principais (farelo de algodão ou grão de soja crú integral) e dois níveis de proteína (130 g ou 148 g/kg de matéria seca- MS). No presente estudo, houve efeito de interação entre a fonte nitrogenada e o teor de PB sobre o consumo de MS (CMS). As vacas alimentadas com grão de soja crú integral tiveram maior CMS quando o teor de PB da dieta foi de 130 g CP/kg MS, enquanto que as vacas alimentadas com farelo de algodão apresentaram maior CMS quando o teor de PB da dieta foi de 148 g PB/kg MS. Resultado semelhante ao CMS foi observado para o consumo de matéria orgânica, enquanto que não houve efeito de interação entre fonte nitrogenada e teor de PB sobre o consumo dos demais nutrientes dietéticos. A digestibilidade aparente total da FDN foi maior quando grão de soja crú integral foi utilizado como principal fonte nitrogenada da dieta; enquanto que a digestibilidade da PB não foi alterada pelas fontes nitrogenadas. Apesar de haver alterações no CMS e na digestibilidade aparente total, no presente estudo, não houve efeito de teor de PB e fonte nitrogenada sobre a produção de leite. Como conclusão, o uso de baixo teor dietético de PB (130 g PB/kg de MS) não altera o desempenho produtivo de vacas leiteiras e pode reduzir os custos da dieta, enquanto que a escolha entre grão de soja cru integral e farelo de algodão como principais fontes nitrogenadas da dieta pode depender apenas da disponibilidade e custo, uma vez que ambos ingredientes apresentam desempenhos semelhantes. | The aim of the present study was to evaluate the effect of two crude protein contents (CP) and two main nitrogen sources in the diet of dairy cows, using sugarcane as forage, on intake, total apparent digestibility, milk yield and composition, and nitrogen balance. Twelve Holstein cows in late lactation were assigned in three contemporary square, in a Latin Square design 4 × 4, with 2 × 2 factorial treatment arrangement, in 21-day trial by period, 14 for diet adaptation and the last seven for sampling and analysis. Cows were housed in individual stalls and fed a total mixed ration (TMR) "ad libitum" composed of two main nitrogen sources (cottonseed meal or whole raw soybean grain) and two levels of CP (130g or 148g/kg dry matter - DM) in the diet. In the present study, there was an interaction effect between nitrogen source and crude protein on DM intake (DMI). Cows fed whole raw soybean had higher DMI when the CP content of the diet was 130 g CP/kg DM, while cows fed cottonseed meal had higher DMI when the CP content of the diet was 148 g CP/kg DM. A similar result of DMI was observed for organic matter intake, whereas there was no interaction effect between dietary nitrogen source and crude protein content on intake of other nutrients. Total apparent digestibility of NDF and TDN was higher when soybean was used as the main diet nitrogen source; whereas the digestibility of dietary CP was not changed by the nitrogen sources. Although there were changes in the DMI and the apparent digestibility coefficient, in the current study, there was no effect of dietary crude protein content and nitrogen source on milk yield. In conclusion, the use of low dietetic concentrations of crude protein (130 g/Kg of DM) does not change the lactating performance of dairy cows and may reduce diet cost, while the choice between whole raw soybean grain or cottonseed meal as a protein ingredient may depends only on the availability and/or price, as both ingredients result in similar performance.

Toxoplasmosis and neosporosis are diseases that may affect production animals and cause significant economic losses. Given the importance of this fact, risk factors and occurrence of antibodies anti-Toxoplasma gondii and anti-Neospora caninum were determined for 81 sheep in seven farms of the Guarapuava region, state of Paraná, Brazil. Indirect fluorescence antibody test (IFAT) showed antibody frequency of 40.74% for T. gondii and 3.70% for N. caninum. The risk factors evaluated were: for T. gondii, direct contact of sheep with cats and for N. caninum, with dogs; abortion in the herd; and access of other animal species to the same water tank used by the sheep. CI (95%), Odds Ratio, and p value were determined by Fisher Exact Test. No statistical difference was observed between the occurrence of antibodies and risk factors associated with the presence of antibodies against both parasites. It may be stated that the protozoa T. gondii and N. caninum are found in the sheep herds of this region.

A toxoplasmose e neosporose são doenças que podem afetar os animais de produção causando perdas econômicas significativas. Sabendo disso, foram determinados os fatores de risco e a ocorrência de anticorpos anti-Toxoplasma gondii e anti-Neospora caninum em 81 ovinos de sete propriedades da região de Guarapuava, Paraná, Brasil. A pesquisa de anticorpos foi realizada por meio da Reação de Imunofluorêscencia Indireta (RIFI) e foi observada uma frequência de 40,74% para T. gondii e de 3,70% para N. caninum. Os fatores de risco pressupostos para ambas as infecções foram: o contato direto dos ovinos com felinos, para T. gondii, e com cães para N. caninum, presença de aborto no rebanho e o acesso de outras espécies animais ao mesmo bebedouro dos ovinos. Por meio do Teste Exato de Fisher foram calculados o IC (95%), Odds ratio e o valor de p, os quais não revelaram associação entre a ocorrência de anticorpos e os fatores de riscos analisados. Some Organochlorine Pesticides (OCPs) can pose numerous adverse effects on biota. Marine turtles face numerous threats, in particular those related to anthropogenic activities. Therefore, development and improvement methodologies for monitoring chemical compounds are a relevant task. In this work, we developed a methodology based on the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) extraction for detection of twelve OCPs, by gas chromatography with electron capture detector, in fat and liver samples of green sea turtles. Quantification limits were lower than 5.3 ng g-1; acceptable recovery rates for most compounds; medium matrix effect; matrix-calibration with linearity at the range from 1.0 to 200 ng g-1. This methodology provides contributions for the study of pesticide residues with adverse effects on sea turtle health, important skills for new directions in conservation issues.

Alguns Pesticidas organoclorados (OCPs) podem causar numerosos efeitos adversos na biota. As tartarugas marinhas enfrentam diversas ameaças, em especial aquelas relacionadas às atividades antropogênicas, por isso o desenvolvimento de melhorias nos métodos para monitorar compostos químicos são tarefas importantes. Neste trabalho foi desenvolvida uma metodologia baseada na extração QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) para a detecção de doze OCPs, por cromatografia gasosa com captura de elétrons, em amostras de gordura e fígado de tartarugas verdes. Os limites de quantificação ficaram abaixo de 5.3 ng g-1; com taxas de recuperação aceitáveis para a maioria de compostos; efeito matriz médio; calibração da matriz com linearidade variando de 1.0 a 200 ng g-1. Serum samples from 83 free living wild pigs (Sus scrofa) from the Nhecolândia region, Pantanal do Mato Grosso, Brazil, were examined for the presence of antibodies against Neospora caninum by means of the indirect fluorescence antibody test (IFAT) with a cut off of 50. Antibodies were present in 10.8%, with titers of: 1:200 in one animal, 1:400 in four and 1:800 in four. Analysis using the χ2 test showed an association between sex and presence of antibodies, with females showing occurrence of 20.5% and males 2.3% (p = 0.017). No association was found between age and occurrences of antibodies against this parasite. This was the first observation of N. caninum antibodies in Brazilian wild pigs from Pantanal.

Quando analisados pelo teste do χ2, foi observada a existência de associação entre sexo e presença de anticorpos, com as fêmeas apresentando ocorrência de 20,5% e os machos 2,3% (p = 0,017). Não houve associação entre idade e ocorrência de anticorpos contra este parasito.
