Use of partially immobilized membrane ATPase in heavy metals determination

Previous studies clearly indicated that membrane enzymes - ATPases i.e. Na, K-ATPase and Mg-ATPase, responded to presence of various organic and inorganic pollutants. In this work effect of mercury and cadmium on these enzymes activities was investigated in synaptic plasma membranes partially immobilized on microliter plate. Comparing those activities with the control enzyme activities obtained with native and partially immobilized mambranes it was concluded that: a. both metals exerted a concentration-dependent inhibition of investigated enzymes, b. for partially immobilized membranes estimated half maximum inhibition (IC50) values for Na, K-ATPase were IC50 (Hg) = 0.9 micromol/cubic cm, IC50 (cd) = 35 micromol/cubic cm and for Mg - ATPase IC50 (Hg) = 3.5 micromol/cubic cm, IC50 (Cd) = 36 micromol/cubic cm; for native membranes IC50 for Na,K-ATPase were IC50 (Hg) =3.3 micromol/cubic cm, IC50 (Cd) = 2 micromol/cubic cm and for Mg ATPase IC50 (Hg) = 2.3 micromol/cubic cm, IC50 (Cd) = 0.2 mmol/cubic cm. Obtained results indicate avaibility of microtitar plates for partially immobilization of membranes with aim to form a new biosensor for heavy metals detection.

During the period 1990-1997, the content of 11 trace metals (Fe, Cr, Co, Al, Mn, Cd, Cu, Ni, Pb, Zn, Sr) was studied in muscle and other organs of fish from DTD (Danube-Tisza-Danube) channel system (Yugoslavia). According to obtained and analysed data, the trace metal bioaccumulation can possibly act as enviromental indicator in biomonitoring. DTD (Danube-Tisza-Danube) Channel System can, therefore, be classified as the 3rd class, concerning analysed trace metal content.

One of the very dangerous pollution of environment are heavy metals. They are "indestructible" at the condition which leads at the environment. Pollution of heavy metals is irreversible process, except if they are physical removed from the environment which they are polluting. We determinated heavy metals (Pb, Cd, Cu, Zn, Cr, Ni and Co) in the mud of three fish ponds by means of sequential extraction, because of possible mobility in the mud and its concentration increase in the water. This method was selected because we wanted to determine distribution of heavy metals by substrates, and analyse was worked by the method of AAS. The results are showing that the concentration of all metals is in the extent of "natural phone" of lakes sediment.

The last decade of 20th century in Serbia (Yugoslavia) was marked with economy recession, war surroundings, and finally bombing in the period March-June 1999. Global increment of environmental pollution was one of the catastrophic consequences. War actions caused destroying of big oil refineries and reservoirs, chemical complexes and industrial objects. Afterwards, toxic and carcinogenic substances reached out to atmosphere, soil and hydrographic system. Also, ammunition with depleted uranium was used. Heavy metals are very dangerous, so that, in this paper are presented results of investigation of heavy metals contents in surface soil layers. Heavy metals are movable, by erosion processes, with possibility of reaching hydrographic network and water supply systems. Investigation was carried out at a few localities: Fruska Gora, Ivanovo, Goc and Crni Vrh. Total contents of heavy metals (Zn, Cu, Pb, Cd, Mn) was determined by AAS (atomic spectrophotometry), by usage "Varian-Spectra AA-10" apparatus.

The paper deals with the results of the phytoplankton investigations carried out in the 7 localities along the Yugoslav section of the Tisza river in the period of 13 Februar - 3 March 2000, after accidental pollution with the cyanide and heavy metals. Although certain decrease in the density of the phytoplankton and variation in the qualitative and quantitative composition were recorded, there are no significant differences in comparison with the data before accident. The short-term and possible long-term consequences were recognized, having in mind ability of algae to accumulate pollutants into their cells, and that algae are the first link on food chains. Since the content of pollutants increases with every next member in the chain, it is easy to assume cumulative effect of the all previous members.

In this work, we were investigated level and source of trace elements in industrial and sea waters in Bokakotorska Bay (Adriatic sea, Montenegro, Yugoslavia). The ranges concentration in (microgram/cubic dm) in seawater are as follows: Hg (0.1-0.4); As (0.1-0.9); Pb (2.3-10.0); Zn (3.0-12.0); Cu (2.3-8.4); Cr (0.8-3.0); Cd (0.1-1.0); Ni (0.9-3.0); Mn (0.2-0.5). The average levels of trace elements in the samples collected from industrial effluents of factory ILK were 110 - 16500 times higher than concentrations of trace elements in other industrial effluents.

Over the last several years, the investigations of the natural zeolites application in the sorption processes have been intensified. Purification of waste waters in order to remove lead, cadmium, copper and other heavy metals, is one significant example of such application. In this paper, the investigations results on characteristics of the natural and chemically activated zeolites from the region of Vranje (Serbia, Yugoslavia), are presented. The experiments with zeolites were performed after determination of their physico-chemical characteristics. Adsorptive characteristics were investigated under laboratory conditions, in a liquid medium, depending on granulation and concentration of the heavy metals. As the obtained results show, these natural materials can be used to remove heavy metals from the waste waters.

The paper provides experimental results of testing the content of heavy metals, and other harmful and dangerous matters of the Sava river sludge (Serbia, Yugoslavia). The testing carried out by the Republic Hydrometeorological Service of Serbia (Yugoslavia), has been in compliance with the program of testing the Sava river water and sludge quality along the river flow. The establishing of sludge pollution range is not based upon values of maximum allowable concentrations. The most complex is the issue of heavy metals contents, since these also represent the natural contents of soil and minerals. However, considering the process of migration and distribution of heavy metals, and other harmful and dangerous matters discharged from utilities, industrial and power supply facilities, provides the basis for estimating the environmental pollution as well.

Spectrophotometric method for determination of inorganic phosphate liberated in hydrolysis of ATP catalyzed by ATPase was modified in order to obtain faster procedure, which could also be used in none laboratory conditions. The modification has some advantages compared to the most used Pennial method: a) the reagents are stable for several months; b) the 45 min procedure of phosphomolibdate extraction by isobutanole-benzene is ommited, and the method is not dangerous for the analyst; c) color develops after 20 min. The method was tested on the determination of inorganic phosphate in the presence of cadmium nitrate as inhibitor of ATPase activity. The results were compared to the results obtained by Pennial method. The results obtained have shown some good agreements.

In our previous papers, it is concluded that wood sawdust can be applied as adsorbent for heavy metals removing from water. All earlier investigations are done in batches, while it is confirmed that different heavy metals are adsorbing with various efficacy on sawdust of different kind of wood. Adsorption of copper ions from model water is investigated in this paper, by its passing throw column filled with poplar and fir sawdust, granulation in the range 0.5-1 mm. Starting concentration of copper ions was the same in all experiments, and flowrates of model water and column geometry are varied. Certain volumes of water are taken on the column outlet, and copper ions concentration are determinated in this samples. On the basis of these results breakthrough curves are obtained, and adsorption parameters in dynamic conditions are determinated.
