Arsenic accumulation in Pteris vittata: Time course, distribution, and arsenic-related gene expression in fronds and whole plantlets

In this work, arsenic (As) accumulation and distribution over time in Pteris vittata young fronds from adult plants and in whole plantlets, grown on a highly contaminated As-soil, was determined by μ-XRF. A linear increase in As content up to 60 days was found in young fronds at different times, and a progressive distribution from the apex to the base of the fronds was observed. In whole plantlets, As signal was detectable from 9 to 20 days in the apex of a few fronds and fiddleheads. Later, up to 60 days, As was localized in all fronds, in the rhizome and in basal part of the roots. The dynamics of expression of As-related genes revealed a good correlation between As content and the level of the As (III)-antiporter PvACR3 transcript in plantlets roots and fronds and in young fronds. Moreover, the transcription of As (V)-related gametophytic genes PvGAPC1, PvOCT4 increases over time during As accumulation while PvGSTF1 is expressed only in roots. Here, we demonstrate the suitability of the μ-XRF technique to monitor As accumulation, which allowed us to propose that As is initially directly transported to fiddleheads and apex of fronds, is later distributed to the whole fronds and simultaneously accumulated in the rhizome and roots. We also provide indications on the expression of candidate genes possibly involved in As (hyper)accumulation.

Selenium (Se) is an essential micronutrient for animals with a relatively narrow margin between essentiality and toxicity. To evaluate Se removal efficiency by a constructed wetland treatment system and its potential eco-risk, a floating-leaved macrophyte system was constructed, consisting of three main trophic levels. Over 21-d treatment, water Se concentration was gradually reduced by 40.40%, while 24.03% and 74.41% of the removed Se were found in the plant Nymphoides sp. and sediment, respectively. Among plant tissues, roots accumulated the highest Se level, although the greatest total Se was found in stems, followed by leaves, roots and rhizomes. X-ray absorption spectroscopy revealed that 82.65% of the absorbed selenite by the plants was biotransformed to other forms, as organo-Se species accounted for 45.38% of the Se retained in the sediment, which was primarily responsible for the entry of Se into the detritus food chain. The proportion of organo-Se compounds increased with trophic levels from sediments to fish, indicating, instead of direct uptake of selenite, the food chain transfer and biotransformation of Se may serve as a key exposure route for Se in aquatic organisms. When exposed to organo-Se compounds, i.e., SeCys and SeMet, the plants, shrimp and fish tended to accumulate more Se. However, the greater trophic transfer factor was obtained for selenate, leading to higher Se levels accumulated in fish. Overall, in addition to key mechanisms involved in Se removal, our research also provides a much better understanding of the potential eco-risk that may be posed by the floating-leaved plant system for bioremediation of Se via food chain transfer and biotransformation, paving the way for a low eco-toxic treatment system for Se remediation.

Dietary consumption of contaminated vegetables may contribute to polycyclic aromatic hydrocarbon (PAH) exposure in humans; however, this exposure pathway has not been examined thoroughly. This study aims to characterize the concentrations of PAHs in six types of vegetables grown near industrial facilities in Shanghai, China. We analyzed 16 individual PAHs on the US EPA priority list, and the total concentration in vegetables ranged from 65.7 to 458.0 ng g−1 in the following order: leafy vegetables (romaine lettuce, Chinese cabbage and Shanghai green cabbage) > stem vegetables (lettuce) > seed and pod vegetables (broad bean) > rhizome vegetables (daikon). Vegetable species, wind direction, and local anthropogenic emissions were determinants of PAH concentrations in the edible part of the vegetable. Using isomer ratios and principal component analysis, PAHs in the vegetables were determined to be mainly from coal and wood combustion. The sources of PAHs in the six types of vegetables varied. Daily ingestion of PAHs due to dietary consumption of these vegetables ranged from 0.71 to 14.06 ng d−1 kg−1, with contributions from Chinese cabbage > broad bean > romaine > Shanghai green cabbage > lettuce > daikon. The daily intake doses adjusted by body weight in children were higher than those in teenagers and adults. Moreover, in adults, higher concentrations of PAHs were found in females than in males. For individuals of different age and gender, the incremental lifetime cancer risks (ILCRs) from consuming these six vegetables ranged from 4.47 × 10−7 to 6.39 × 10−5. Most were higher than the acceptable risk level of 1 × 10−6. Our findings demonstrate that planting vegetables near industrial facilities may pose potential cancer risks to those who consume the vegetables.

The potential risk to human health of antibiotics that pass through the food chain has become an important global issue, but there are few reports on the response of ginger (Zingiber officinale) to antibiotic pollution. In this study, we investigated the enrichment characteristics and biological response of ginger to sulfamethoxazole (SMZ) and ofloxacin (OFL) residues, which are common in the environment. Lower levels of SMZ, OFL and their combined duplex treatment (SMZ+OFL) promoted the growth of ginger, but the critical doses necessary to stimulate growth differed among treatments: 10 mg L⁻¹ SMZ, 1 mg L⁻¹ OFL and 1 mg L⁻¹ (SMZ+OFL) had the strongest stimulating effects. At higher dosages, the root growth and light energy utilization efficiency of ginger were impaired, and (SMZ+OFL) had the strongest inhibitory effect. Treatments with lower levels of antibiotics had no significant effect on reactive oxygen species and antioxidant enzyme activities. However, when SMZ, OFL and SMZ+OFL concentrations exceeded 10 mg L⁻¹, the contents of H₂O₂, O₂⁻ and MDA continued to increase, while the activities of SOD, POD, CAT first increased and then decreased, especially in SMZ+OFL. Ginger accumulated more SMZ and OFL in rhizomes and less in leaves, and accumulation increased significantly as antibiotic concentration increased. When SMZ concentration was 1 mg L⁻¹, the SMZ concentrations in rhizomes, roots, and leaves were 0.23, 0.15, and 0.05 mg kg⁻¹, respectively, and the residual SMZ in the rhizome was 2.3 times higher than the maximum residue limit. The abundance of the resistance genes sul1, sul2, qnrS, and intI1 increased with increasing antibiotic concentrations, and intI1 abundance was the highest. OFL induced higher levels of intI1 expression than did SMZ.

Large amounts of non-biodegradable plastics are currently deposited on beach-dune systems, and biodegradable plastics could enter these already declining habitats in coming years. Yet, the impacts of plastics on vegetative recruitment, a plant strategy playing a key role in dune stabilization, are unknown. Whether these pollutants interact with increased atmospheric nitrogen (N) deposition, a major global driver of plant biodiversity loss, in affecting plant communities of such nutrient-poor habitats, and how plant-plant interactions mediate their effects need to be explored. In a one-year field experiments, we examined individual and combined effects of plastic (non-biodegradable, biodegradable), N deposition (ambient, elevated) and biotic condition (no interaction, interaction with a conspecific or with a hetero-specific) on the colonization success and growth of vegetative propagules of dune plants. Thinopyrum junceum and Sporobolus pumilus were chosen as models because they co-occur along Mediterranean dunes and differ in ecological role (dune- vs. non dune-building) and photosynthetic pathway (C3 vs. C4). For both species, survival probability was reduced by non-biodegradable plastic and elevated N by up to 100%. Thinopyrum junceum survival was also reduced by S. pumilus presence. Elevated N and biodegradable plastic reduced T. junceum shoot biomass when grown alone and with a conspecific, respectively; these factors in combination mitigated their negative individual effects on root biomass. Biodegradable plastic increased S. pumilus shoot and root biomass, and in combination with elevated N caused a greater biomass investment in belowground (root plus rhizome) than aboveground organs. Non-biodegradable plastic may be a further threat to dune habitats by reducing plant colonization. Biodegradable plastic and increased N deposition could favour the generalist S. pumilus and hinder the dune-building T. junceum. These findings highlight the urgency of implementing measures for preventing plastic deposition on beaches and reducing N input.

Understanding Pb removal from the translocation stream is vital to engineering Pb hyperaccumulation in above ground organs, which would enhance the economic feasibility of Pb phytoextraction technologies. We investigated Cu, Pb, Sb and Zn distributions in Hay-scented fern (Dennstaedtia punctilobula) rhizomes on shooting range soils by laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS), analyzing digested rhizomes, stems, and fronds using ICP-MS. Nutrients Cu and Zn concentrated in fronds while toxic elements Pb and Sb did not, showing potential Pb and Sb sequestration in the rhizome. Frond and rhizome concentration of Pb was 0.17 ± 0.10% and 0.32 ± 0.21% of dry biomass, respectively. The ²⁰⁸Pb/¹³C and ¹²¹Sb/¹³C determined by LA-ICP-MS increased from inner sclerotic cortex to the epidermis, while Pb concentrated in the starchy cortex only in contaminated sites. These results suggest that concentration dependent bioaccumulation in the rhizome outer cortex removes Pb from the vascular transport stream.

Ozone and atmospheric nitrogen are co-occurring pollutants with adverse effects on natural grassland vegetation. Plants of the rhizomatous sedge Carex arenaria were exposed to four ozone regimes representing increasing background concentrations (background-peak): 10–30, 35–55, 60–80 and 85–105 ppb ozone at two nitrogen levels: 12 and 100 kg N ha−1 yr−1. Ozone increased the number and proportion of senesced leaves, but not overall leaf number. There was a clear nitrogen × ozone interaction with high nitrogen reducing proportional senescence in each treatment and increasing the ozone dose (AOT40) at which enhanced senescence occurred. Ozone reduced total biomass due to significant effects on root biomass. There were no interactive effects on shoot:root ratio. Rhizome tissue N content was increased by both nitrogen and ozone. Results suggest that nitrogen mediates above-ground impacts of ozone but not impacts on below-ground resource translocation. This may lead to complex interactive effects between the two pollutants on natural vegetation. Nitrogen alters threshold of ozone-induced senescence, but not below-ground resource allocation.

Existing literature illustrates inconsistent responses of seagrasses to oil exposure, both in the field and in the laboratory. Here, we add a new study that combined morphometric, demographic and photophysiology assessments to determine the potential oiling impacts to eelgrass (Zostera marina) from the 2007 Cosco Busan event in San Francisco Bay. Shoot densities, reproductive status, and rhizome elongation of Z. marina were examined at sites with pre-spill data, and eelgrass photosynthetic efficiency was measured post-spill. Shoot densities and percent elongation of rhizome internodes formed after the oil spill varied but with no consistent relationship to adjacent shoreline cleanup assessment team (SCAT) oiling categories. Similarly, differences in seagrass photosynthetic efficiency were not consistent with SCAT oiling categories. While thresholds for negative impacts on seagrass in general remain to be defined, conclusive oiling indicators for degree and duration of exposure would be important considerations and need examination under controlled study.

Population reconstruction techniques was used to assess for the first time the population dynamics of a seagrass, Cymodocea nodosa, exposed to long-term elevated CO₂ near three volcanic seeps and compared them with reference sites away from the seeps. Under high CO₂, the density of shoots and of individuals (apical shoots), and the vertical and horizontal elongation and production rates, were higher than at the reference sites. Nitrogen limitation effects on rhizome elongation and production rates and on biomass were more evident than CO₂ as these were highest at the location where the limitation of nitrogen was highest. At the seep where the availability of CO₂ was highest and nitrogen lowest, density of shoots and individuals were highest, probably due to CO₂ effects on shoot differentiation and induced reproductive output, respectively. At the three seeps, there was higher short- and long-term shoot recruitment than at the reference sites, and growth rates was around zero, indicating that elevated CO₂ increases the turnover of C. nodosa shoots.

We studied the recovery of the fast-growing seagrass Cymodocea nodosa from disturbances of different intensities (shoots removal or the entire plant), plot sizes (from 0.04 to 1 m²) and in different seasons (spring and autumn) in a shallow coastal bay. We monitored recovery over 27 months and measured plant traits at the end. Shoot density and canopy height recovered faster (1 month) when only shoots were removed compared to when the entire plant was removed (10–25 months). Small areas took longer to recover than large ones, probably due to limited light availability or the accumulation of detritus. Plants disturbed in autumn took 9 months longer to recover than those disturbed in spring. After the 27-month, all plant traits were similar to those of control plots, except rhizome biomass, which was lower. Our results suggest that mechanical disturbances might exert a negative effect on the long-term resilience of seagrasses.
