Environmental co-exposure to TBT and Cd caused neurotoxicity and thyroid endocrine disruption in zebrafish, a three-generation study in a simulated environment

Although the coexistence of heavy metals and environmental hormones always occur in aquatic environment, the information of the combined impacts remains unclear. To explore the multi-generational toxicity of cadmium (Cd) and tributyltin (TBT), adult zebrafish (Danio rerio) (F0) were exposed to different treated groups (100 ng/l Cd, 100 ng/l TBT and their mixture) for 90 d, with their offspring (F1 and F2) subsequently reared in the same exposure solutions corresponding to their parents. Both developmental neurotoxicity and thyroid disturbances were examined in the three (F0, F1, and F2) generations. Our results showed that co-exposure to Cd and TBT induced the developmental neurotoxicity in F1 and F2 generations, reflected by the significant lower levels of neurotransmitters (dopamine and serotonin) and the inhibited acetylcholinesterase (AChE) activities. And the thyroid endocrine disruption were observed in the two-generations larval offspring by parental exposure to Cd and/or TBT, including the significantly decreasing levels of thyroid hormones and the down-regulated the expression of genes involved in the hypothalamus-pituitary-thyroid axis, compared to the control. Additional, the embryonic toxicity and growth inhibition were also determined in the fish larvae. Overall, this study examined the impacts of parental co-exposure to Cd and TBT, with regard to developmental inhibition, nervous system damage and endocrine disruption, which highlighted that co-exposure influences are complicated and need to be considered for accurate environmental risk assessment.

The strict environmental management has been implemented in Taihu Basin to reduce the surface water contamination; however, the effectiveness of the management actions has not been comprehensively evaluated. In the present study, 364 samples were collected during four campaigns over a span of one year from surface water, municipal wastewater treatment plants (MWWTPs), industrial wastewater treatment plants (IWWTPs), industrial enterprises, and aquaculture in a typical region in the Upper Taihu Basin. Overall concentration, temporal variation and spatial distribution of 16 PAHs in surface water and various pollution sources were evaluated and the potential pollution sources were identified. Results showed that concentrations of individual PAHs in the surface water ranged from less than the limit of quantification (LOQ) to 949 ng L⁻¹, indicating a reduction of PAH contamination level after the implementation of environmental management actions. Influent of MWWTPs and wastewater from industrial enterprises exhibited relatively high ∑PAHs concentrations (mean: 880 ng L⁻¹ and 642 ng L⁻¹, respectively); these samples also exhibited a similar seasonal variation as well as composition of PAH congeners to those found in surface water, and therefore were designated as the main emission sources of PAHs in the studied region. Additional source apportionment using principal component analysis was also conducted to verify the proposed sources and diagnose other pollution sources. The findings provided a thorough understanding of PAH pollution, especially its major emission sources, in a typical region with pollution-intensive industries after the implementation of strict environmental management.

The potential risks of phthalates affecting human and animal health as well as the environment are emerging as serious concerns worldwide. However, the mechanism by which phthalates induce developmental effects is under debate. Herein, we found that embryonic exposure of zebrafish to di-(2-ethylhexyl) phthalate (DEHP) and di-butyl phthalate (DBP) increased the rate of heart defects including abnormal heart rate and pericardial edema. Changes in the transcriptional profile demonstrated that genes involved in the development of the heart, such as tbx5b, nppa, ctnt, my17, cmlc1, were significantly altered by DEHP and DBP at 50 μg/L, which agreed with the abnormal cardiac outcomes. Methylated DNA immunoprecipitation sequencing (MeDIP-Seq) further showed that significant hypomethylation of nppa and ctnt was identified after DEHP and DBP exposure, which was consistent with the up-regulation of these genes. Notably, hypermethylation on the promoter region (<1 kb) of tbx5b was found after DEHP and DBP exposure, which might be responsible for its decrease in transcription. In conclusion, phthalates have the potential to induce cardiac birth defects, which might be associated with the transcriptional regulation of the involved developmental factors such as tbx5b. These findings would contribute to understand the molecular pathways that mediated the cardiac defects caused by phthalates.

In parts of developing countries, the over-exploitation of sands from inland waters has led to serious environmental concerns. However, understanding of the impacts of commercial sand dredging on inland water ecosystem functions remains limited. Herein, we assess the effects of this activity on the functional structure of the macroinvertebrate community and its recovery processes based on a 4-year survey, in the South Dongting Lake in China. Our result showed a simplified macroinvertebrate functional structures within the dredged area, as evidenced by a loss of certain trait categories (e.g., oval and conical body form) and a significant reduction in trait values due to the direct removal of macroinvertebrates and indirect alternations to physical environmental conditions (e.g., water depth and %Medium sand). Moreover, clear increases were observed in certain trait categories (e.g., small body size and swimmer) resulting from the dredging-related disturbance (e.g., increased turbidity) within the adjacent area. Furthermore, one year after the cessation of dredging, a marked recovery in the taxonomic and functional structure of macroinvertebrate assemblages was observed with most lost trait categories returning and an increase in the trait values of eight categories (e.g., body size 1.00–3.00 cm and oval body form) within the dredged and adjacent area. In addition, dispersal processes and sediment composition were the main driver for the structuring of the macroinvertebrate taxonomic and functional assemblages during the dredging stages, whilst water environmental conditions dominated the taxonomic structure and dispersal processes determined the functional structure during the recovery stage. Implications of our results for monitoring and management of this activity in inland waters are discussed.

Polychlorinated biphenyls (PCBs) are notorious environmental pollutants. For their hydrophobic and lipophilic capability, they are wildly spread to environment to threat human health thus attracts more attention. In this study, we observed increasing numbers of CD163 positive (CD163⁺) macrophages in aortic valve of ApoE⁻/⁻ mice after 2,3,5-trichloro-6-phenyl-[1,4]-benzoquinone (PCB29-pQ) treatment, the metabolite of polychlorinated biphenyl. In addition, in vitro studies identified that PCB29-pQ exposure significantly provoked the shifting of RAW264.7 macrophages and bone marrow derived monocytes (BMDMs) to CD163⁺ macrophages. Upon PCB29-pQ administration, CD163 and CD206 levels were enhanced in RAW264.7 cells as well as in BMDMs. However, the concentration of iron and total cholesterol (TC) were reduced due to the boosting of ferroportin (Fpn) and ATP binding cassette transporter, subfamily A, member 1 (ABCA1) which are efflux transporters of iron and cholesterol individually. Further investigation on mechanism indicated that PCB29-pQ exposure induced reactive oxygen species (ROS), which may result in activation of nuclear factor erythroid 2-related factor 2 (Nrf2), a protein responsible for macrophage polarization. After that, we blocked Nrf2 through Nrf2 shRNA and ROS scavenger NAC, which significantly reversed the shifting of macrophage to CD163⁺ sub-population. These results confirmed the importance of Nrf2 in inducing macrophage polarization. In short, our study uncovered that PCB29-pQ could promote macrophage/monocyte polarization to CD163⁺ macrophage which would be a potential incentive to accelerate atherosclerosis through Nrf2 signaling pathway.

Whereas non-extractable residue (NER) formation is recognized as an important process affecting the ecological risk of organic contaminants in soils, it is commonly neglected in regional-scale multi-media models assessing chemical environmental fate and risk. We used a combined field and modeling study to elucidate the relative importance of NER formation to the reduction in available organic contaminants compared with fate processes commonly considered in risk assessment models (volatilization, leaching, and biodegradation). Specifically, four polycyclic aromatic hydrocarbons (PAHs), i.e., phenanthrene (Phe), pyrene (Pyr), benzo[a]pyrene (BaP), and benzo[ghi]perylene (BghiP), were spiked and measured in a one-year field pot experiment at four sites with diverse environmental conditions on the eastern Tibetan Plateau. The rate of NER formation was derived as the difference between the overall rate of decline in total-extractable PAH concentrations, obtained by fitting a biphasic first-order model to the measured concentrations, and the sum of the calculated rates of volatilization, leaching, and biodegradation. Our work shows that the total-extractable PAH concentration undergoes a rapid decline and a slow decline, with shorter overall half-lives (especially for BaP and BghiP) than those observed in earlier studies. Generally, NER formation was assessed to be the dominant contributor (64 ± 33%) to the overall decline of PAHs, followed by biodegradation (35 ± 32%); volatilization and leaching were the smallest contributors. In particular, heavier PAHs (i.e. BaP and BghiP) tend to have shorter half-lives in the rapid and the overall decline phase, indicating that the erroneous estimation of environmental fate and risks might be more pronounced for organic contaminants with a large molecular size. The trend of overall decline rates of PAHs displayed a combined effect of NER formation and biodegradation. This work indicates the need to consider NER formation as a process in multi-media models of chemical fate and risk.

While considerable efforts have been made to address the relationship between urbanization and environmental issues, few of them focus on household emissions. Thus, this work aimed at evaluating the effect of urbanization on household wastewater emissions, and made a distinction between the efficient- and the inefficient-emissions. We compiled a China’s provincial dataset over the period 2005–2017, and estimates indicated that a 1% rise in the urbanization was correlated with a 0.581 increase of efficient emissions, while inefficient emissions decreased by 7.606. As of 2017, the sample period end year, the inefficient emissions accounted for 10.5% of the total emissions of China as a whole, which was relatively large and could not be overlooked. Meanwhile, a back-of-the-envelope estimate revealed that urbanization had a negative impact on China’s total emissions, with a marginal effect ranging from −0.226 to −1.354 over the sample period. The above findings, however, did not imply that urbanization would spontaneously reduce the inefficient- and total-emissions. Thus, the government in the process of urbanization should strengthen environmental education, municipal facilities, and others.

Recently, chemical compounds containing lanthanides were used in various fields of biology and medicine. It has been described that such compounds can be applied in scanning electron microscopy (SEM) to increase the contrast and simplify the sample preparation process due to the process of replacing calcium with lanthanides in cell. However cell death by different mechanisms under influence of lanthanides seems possible. Here, we described that mummification process is a cell death physiologically realized in time: some time after lanthanide contrasting, the cell remains metabolically active and is able to biochemically transform neodymium-containing contrast, oxidize it and form large agglomerates. A distinctive feature of mummification induced by neodymium-containing contrast (NCC) is the formation of a high-rigid oxygen-containing “shield” on the surface of a neutrophil granulocyte.

To compare aquatic organisms’ responses to the toxicity of copper oxide (CuO) nanoparticles (NPs) with those of CuO microparticles (MPs) and copper (Cu) ions, a global metabolomics approach was employed to investigate the changes of both polar and nonpolar metabolites in microalga Chlorella vulgaris after 5-day exposure to CuO NPs and MPs (1 and 10 mg/L), as well as the corresponding dissolved Cu ions (0.08 and 0.8 mg/L). Unchanged growth, slight reactive oxygen species production, and significant membrane damage (at 10 mg/L CuO particles) in C. vulgaris were demonstrated. A total of 75 differentiated metabolites were identified. Most metabolic pathways perturbed after CuO NPs exposure were shared by those after CuO MPs and Cu ions exposure, including accumulation of chlorophyll intermediates (max. 2.4–5.2 fold), membrane lipids remodeling for membrane protection (decrease of phosphatidylethanolamines (min. 0.6 fold) and phosphatidylcholines (min. 0.2–0.7 fold), as well as increase of phosphatidic acids (max. 1.5–2.9 fold), phosphatidylglycerols (max. 2.2–2.3 fold), monogalactosyldiacylglycerols (max. 1.2–1.4 fold), digalactosylmonoacylglycerols (max. 1.9–3.8 fold), diacylglycerols (max. 1.4 fold), lysophospholipids (max. 1.8–3.0 fold), and fatty acids (max. 3.0–6.2 fold)), perturbation of glutathione metabolism induced by oxidative stress, and accumulation of osmoregulants (max. 1.3–2.6 fold) to counteract osmotic stress. The only difference between metabolic responses to particles and those to ions was the accumulation of fatty acids oxidation products: particles caused higher fold changes (particles/ions ratio 1.9–3.0) at 1 mg/L and lower fold changes (particles/ions ratio 0.4–0.7) at 10 mg/L compared with ions. Compared with microparticles, there was no nanoparticle-specific pathway perturbed. These results confirm the predominant role of dissolved Cu ions on the toxicity of CuO NPs and MPs, and also reveal particle-specific toxicity from a metabolomics perspective.

Trace elements (TEs) in the insoluble particles of surface snow are less affected by melting processes and can be used as environmental proxies to reveal natural and anthropogenic emissions. Here the first comprehensive study of the 16 TEs (Al, As, Ba, Bi, Cr, Cu, Fe, Mn, Ni, Pb, Sn, Sr, Ti, U, V, and Zn) in insoluble particles (>0.45 μm) from surface snow samples collected at Urumqi Glacier No. 1 (UG1), Eastern Tien Shan, China, from February 2008 to January 2010 were presented. Results show that concentrations of most insoluble particulate TEs (TEs ᵢₙₛₒₗ) in the snow were higher in summer while lower in winter, due to the increasing particle inputs and melting processes. The abundances of As, Cr, Cu, Ni, Pb, and Zn in some samples were higher than those in surrounding urban soils, which might due to these TEs have further anthropogenic input beyond the already contaminated re-suspended urban soil particles and TEs were mainly enriched in particles with small grain size. Based on enrichment factor (EF) and principal component analysis (PCA), our results suggest that eight TEs (Al, Fe, Ti, Ba, Mn, Sr, U, and V) mainly came from mineral dust, while the remaining eight TEs (As, Bi, Cr, Cu, Ni, Pb, Sn, and Zn) were affected by coal combustion, mining and smelting of non-ferrous metals, traffic emissions, and the steel industry. The Hybrid Single-Particle Lagrangian Integrated Trajectory (HYSPLIT) model suggests that pollutants might originate from Xinjiang province, Kazakhstan, and Kyrgyzstan. Moreover, UG1 received more significant inputs of particle-bound pollutants in summer than in winter due to the stronger convection and the prevailing valley wind that transports pollutants from the city of Urumqi.
