Human hepatoma cells exposed to estuarine sediment contaminant extracts permitted the differentiation between cytotoxic and pro-mutagenic fractions

Complex toxicant mixtures present in estuarine sediments often render contaminant screening unfeasible and compromise determining causation. HepG2 cells were subjected to bioassays with sediment extracts obtained with a series of progressively polar solvents plus a crude extract. The sediments were collected from an impacted area of an estuary otherwise regarded as pristine, whose stressors result mostly from aquaculture effluents and hydrodynamic shifts that enhance particle deposition. Compared to a reference scenario, the most polar extracts yielded highest cytotoxicity while higher genotoxicity (including oxidative damage) was elicited by non-polar solvents. While the former caused effects similar to those expected from biocides, the latter triggered effects compatible with known pro-mutagens like PAHs, even though the overall levels of toxicants were considered of low risk. The results indicate that the approach may constitute an effective line-of-evidence to infer on the predominant set of hazardous contaminants present in complex environmental mixtures.

Air pollution has been accepted as an important contributor to asthma development and exacerbation. However, the evidence is limited in China. In this study, we investigated the acute effect of air pollution on asthma hospitalization in Shanghai, China. We applied over-dispersed generalized additive model adjusted for weather conditions, day of the week, long-term and seasonal trends. An interquartile range increase in the moving average concentrations of PM10, SO2, NO2 and BC on the concurrent day and previous day corresponded to 1.82%, 6.41%, 8.26% and 6.62% increase of asthmatic hospitalization, respectively. The effects of SO2 and NO2 were robust after adjustment for PM10. The associations appeared to be more evident in the cool season than in the warm season. Our results contribute to the limited data in the scientific literature on acute effects of air pollution on asthma in high exposure settings, which are typical in developing countries.

Ground-level ozone (O3) levels are usually lower in urban centers than nearby rural sites. To compare trends in O3 levels during the period 1990–2010, we obtained monitoring data from paired urban and rural sites from the European Environment Agency and the US Environmental Protection Agency. Ozone peaks decreased at both station types, with no significant differences between urban and rural stations. Ozone annual averages increased at both urban and rural sites, with a faster rate of increase for urban centers. The overall trend was for convergence between urban and rural O3 data. Ozone levels exceeded the criteria established for the protection of human and vegetation health at both urban and rural sites.

Polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) belong to the group of persistent organic pollutants, highly toxic environmental pollutants that include hydrophobic compounds with the tendency to bioaccumulate. Earthworms (Eisenia andrei) were exposed to PCDD/Fs-contaminated soil, and changes in their lipophilic structures and the gene expression of their defense molecules were followed. Damage to the intestinal wall and adjacent chloragogenous tissue was observed. Further, the up-regulation of the expression of several genes was detected. On the basis of these results, the mechanism of the impact of PCDD/Fs on earthworms has been proposed. Dioxins that accumulate in the lipophilic structures cause an increase in reactive oxidative species that triggers oxidative stress followed by the gene expression of two molecules that play a role in protection against oxidant toxicity, calreticulin (CRT) and Hsp70. Moreover, the effect of microbial biomass on the expression of coelomic cytolytic factor (CCF), a pattern recognition receptor, was also observed.

In Asia, the evaluation of black carbon (BC) using global aerosol-transport models has been incomplete due to a lack of available measurements. Recently, new measurements and datasets at various Asian sites have become available for use in model validation. In this study, we compared the BC mass concentrations obtained by an aerosol-coupled global nonhydrostatic model adopting a uniform-grid system with in-situ measurements and other models over Asia. The results revealed that our model, as well as other global models, was unable to reproduce the observed BC values at most sites in China and India, most likely due to strong local hotspots. To overcome the inconsistency between the models and measurements, we developed a new aerosol-transport model using a stretched-grid system for high-resolution simulations with up to approximately 10 km grids. Our model can be used as a global-to-regional seamless aerosol-transport model for low to high horizontal resolution simulations.

This paper reports the results of an Open-Top Chambers experiment on the responses of two durum wheat cultivars (Neodur and Virgilio) exposed to two different levels of ozone (charcoal-filtered air and ozone-enriched air) and irrigation water salinity (tap water as control and a 75 mM NaCl solution once a week).The stomatal conductance of the flag leaves was measured on four dates during May. Flag leaf samples were collected to detect ozone visible leaf injuries. At the end of the growing season, the yield/biomass and grain quality parameters were assessed. Saline irrigation caused significant reductions in gs, yield and grain quality in Neodur, while Virgilio was more tolerant. The yield response to ozone was almost negligible, with Virgilio, despite the higher susceptibility to visible leaf injuries, being more productive than Neodur.The responses to the combined stress were not consistent, with the main tendencies undoubtedly driven by the saline irrigation factor.

In the Metropolitan Region of Campinas (MRC), Brazil, high levels of primary pollutants contribute to ozone (O3) formation. However, little is known regarding the O3 effects in the tropics. Objectives in this study were to characterize the present levels of O3 pollution and to evaluate the relevance of current concentration-based indices for assessing the phytotoxic potential of O3. Changes in O3 concentrations and precursors at 5 monitoring stations within towns of MRC were analyzed. The daily O3 profile was typical for urban sites and showed little yearly variation. Given the permanently foliated forest canopy, yearly rather than seasonal O3 indices were thus more appropriate for estimating the effective ozone dose. With yearly SUM00, SUM60 and AOT40 of 156, 16 and 14 ppm h and confirmed by evidence of O3 injury in foliage, oxidative stress in the MRC has reached levels high enough to affect trees from the Atlantic Semi-deciduous Forest.

Coronene is a high molecular weight polycyclic aromatic hydrocarbon with seven aromatic rings. It, more specifically a lower ratio of Benzo[a]pyrene to Coronone (BaP/COR), is suggested as a marker for vehicle emission. In the present study, emissions of Coronene were measured from residential combustions of wood, crop straw, and pellets. The detection of COR in non-vehicle emission sources, and comparable BaP/COR ratios between the solid fuel combustion and vehicle emissions indicated that the generality of COR or the BaP/COR ratio as markers for the vehicle emission would be questionable, especially for the area where solid fuel combustion dominated the PAHs emission.

In this study, we demonstrate that partial least-squares regression analysis with full cross-validation of spectral reflectance data estimates the amount of polycyclic aromatic hydrocarbons in petroleum-contaminated tropical rainforest soils. We applied the approach to 137 field-moist intact soil samples collected from three oil spill sites in Ogoniland in the Niger Delta province (5.317°N, 6.467°E), Nigeria. We used sequential ultrasonic solvent extraction–gas chromatography as the reference chemical method. We took soil diffuse reflectance spectra with a mobile fibre-optic visible and near-infrared spectrophotometer (350–2500 nm). Independent validation of combined data from studied sites showed reasonable prediction precision (root-mean-square error of prediction = 1.16–1.95 mg kg−1, ratio of prediction deviation = 1.86–3.12, and validation r2 = 0.77–0.89). This suggests that the methodology may be useful for rapid assessment of the spatial variability of polycyclic aromatic hydrocarbons in petroleum-contaminated soils in the Niger Delta to inform risk assessment and remediation.

We investigated the effects of arsenic species on As accumulation, plant growth and rhizospheric changes in As−hyperaccumulator Pteris vittata (PV). PV was grown for 60−d in a soil spiked with 200 mg kg−1 arsenate (AsV−soil) or arsenite (AsIII−soil). Diffusive gradients in thin−films technique (DGT) were used to monitor As uptake by PV. Interestingly AsIII−soil produced the highest PV biomass at 8.6 g plant−1, 27% and 46% greater than AsV−soil and the control. Biomass increase was associated with As−induced P uptake by PV. Although AsIII was oxidized to AsV during the experiment, As species impacted As accumulation by PV, with 17.5% more As in AsIII−soil than AsV−soil (36 vs. 31 mg plant−1). As concentration in PV roots was 30% higher in AsV−soil whereas As concentration in PV fronds was 7.9% greater in AsIII−soil, suggesting more rapid translocation of AsIII than AsV. These findings were important to understand the mechanisms of As uptake, accumulation and translocation by PV.
